A series of perylene‐3,4,9,10‐bis(dicarboximide) (PDI) dimers linked through the bay regions was systematically synthesized to examine the electronic structures and photophysical properties in dependence on the distance and orientation between the two PDI units. The spectroscopic and electrochemical measurements suggested that the coupling value of a directly linked PDI dimer (PDI)2 is much larger than those of para‐ and meta‐phenylene‐bridged PDI dimers p‐(PDI)2 and m‐(PDI)2. The width of Davydov splitting was quantitatively evaluated to compare the coupling values between the two PDI units in these dimers by absorption spectroscopy in frozen 2‐methyl‐THF. Excimer formation of PDI dimers induced the strong fluorescence quenching and large red‐shifts. Femtosecond transient absorption revealed a broad absorption derived from an excimer in the range from about 600 nm to the near‐IR region. The rate constants of formation and decay of the excimer are strongly dependent on the coupling values. Time‐resolved measurements on ferrocene‐linked p‐(PDI)2 revealed a competition between the photoinduced processes of electron transfer and excimer formation in PhCN, which is in sharp contrast with the sole electron‐transfer process in toluene.  相似文献

(N-Ethylcarbazol-3-yl)methyl acrylate and methacrylate were polymerized anionically and radically. Anionically-polymerized polyacrylate using ethylmagnesium chloride-benzalacetophenone as catalyst is an isotactic rich polymer with 85% isotactic dyad. All polymers except anionically polymerized polymethacrylate showed good film forming property. Hole drift mobilities measured by the time-of-flight method at room temperature and at 5 × 105 V/cm are in the following order: isotactic polyacrylate (1.0 × 10-5 cm2/V s) > atactic polyacrylate (1.8 × 10-6 cm2/V s) > atactic polymethacrylate (1.2 × 10-6 cm2/V s) at 5 × 105 V/cm. These results are discussed in terms of the hopping model between localized sites.  相似文献

cDNAs encoding geranylgeranyl diphosphate synthase (GGPPS) of two diterpene-producing plants, Scoparia dulcis and Croton sublyratus, have been isolated using the homology-based polymerase chain reaction (PCR) method. Both clones contained highly conserved aspartate-rich motifs (DDXX(XX)D) and their N-terminal residues exhibited the characteristics of chloroplast targeting sequence. When expressed in Escherichia coli, both the full-length and truncated proteins in which the putative targeting sequence was deleted catalyzed the condensation of farnesyl diphosphate and isopentenyl diphosphate to produce geranylgeranyl diphosphate (GGPP). The structural factors determining the product length in plant GGPPSs were investigated by constructing S. dulcis GGPPS mutants on the basis of sequence comparison with the first aspartate-rich motif (FARM) of plant farnesyl diphosphate synthase. The result indicated that in plant GGPPSs small amino acids, Met and Ser, at the fourth and fifth positions before FARM and Pro and Cys insertion in FARM play essential roles in determination of product length. Further, when a chimeric gene comprised of the putative transit peptide of the S. dulcis GGPPS gene and a green fluorescent protein was introduced into Arabidopsis leaves by particle gun bombardment, the chimeric protein was localized in chloroplasts, indicating that the cloned S. dulcis GGPPS is a chloroplast protein.  相似文献

A new apparatus based on the circulation method was developed to measure the solubility of metal complexes in supercritical carbon dioxide (scCO2) at a wide range of temperatures and pressures. A UV–vis spectrometer, which was connected to a small saturation cell through optical fibers, was used to determine solubility. The solubilities of cobalt(III) acetylacetonate (Co(acac)3) and chromium(III) acetylacetonate (Cr(acac)3) in scCO2 were measured to check the validity of both the apparatus and the method and to accumulate new solubility data. The solubility data for Cr(acac)3 obtained in this study were in good agreement with the data reported in the literature.The measured solubilities of Co(acac)3 and Cr(acac)3 were also correlated with the empirical equation including the three adjustable parameters, based on the equation proposed by Chrastil. The parameters were determined by fitting the equation to the experimental data for each metal complex and the calculated results closely replicated the experimental data.  相似文献

Cortisol levels in body fluids are useful for monitoring the function of the pituitary-adrenal axis. Here, we established an “enzyme-linked immunometric assay” (a noncompetitive-type ELISA) for cortisol based on idiotype-anti-idiotype reactions. Six different anti-idiotype monoclonal antibodies that recognized the variable regions of a newly established anti-cortisol antibody were generated using hybridoma technology; these were two β-type and four α-type anti-idiotype antibodies, recognizing the paratope and framework regions, respectively. An immunometric assay was established using a combination of a selected α-type and a selected β-type antibody. The analyte (cortisol) was captured by an excess amount of anti-cortisol antibody immobilized on microplates, and the unoccupied paratope was saturated with the β-type antibody. Hapten-occupied anti-cortisol antibody, with less steric hindrance, was then selectively bound by the α-type antibody, labeled with biotin. The amount of biotin residue on the microplates was colorimetrically monitored using a peroxidase-labeled streptavidin. This assay had an approximately threefold higher sensitivity (detection limit: 90 pg = 248 fmol cortisol) than a competitive ELISA using the same anti-cortisol antibody, as well as a practical specificity for providing reasonable determination of normal urinary cortisol levels.  相似文献

Measurement of cortisol levels in body fluids is important for monitoring pituitary gland and adrenal functions. To develop a specific and standardized enzyme-linked immunosorbent assay (ELISA), a novel monoclonal anti-cortisol antibody has been generated using a reasonably designed haptenic derivative. Spleen cells were prepared from the BALB/c or A/J mouse, which had repeatedly been immunized with a conjugate of 4-(2-carboxyethylthio)cortisol (CET) and bovine serum albumin, to be fused with P3/NS1/1-Ag4-1 myeloma cells. After four fusion experiments, one hybridoma clone secreting a practical antibody has been established. The resulting monoclonal antibody CS#38 (isotype gamma1, kappa) showed an affinity constant (K(a)) for cortisol of 1 x 10(9) M(-1) and provided a practical calibration curve (detection limit, 0.26 ng per assay) in a homologous ELISA system employing horseradish peroxidase-labeled CET as a labeled antigen. Cross-reactivities with related C-21 steroids were acceptably low: 11-deoxycortisol (4.3%), cortisone (4.0%), corticosterone (1.9%), progesterone (1.6%), 17alpha-hydroxyprogesterone (12%), 6beta-hydroxycortisol (8.4%), and tetrahydrocortisol (< 0.1%). Urinary and serum cortisol levels of healthy volunteers were determined by this method after methylene chloride extraction to be 39.0 +/- 17.0 microg/day (n = 7) and 80.8 +/- 38.9 ng/mL (n = 10), respectively, both of which are in the reference range.  相似文献
