Many studies have implicated cis-urocanic acid (cis-UCA) in UVB-induced immunomodulation. The strongest evidence came from studies in mice whereby a cis-UCA antibody blocked UVB-induced suppression of delayed-type hypersensitivity responses. Furthermore, in several studies, the cis-UCA antibody at least partially reversed UVB suppression of contact hypersensitivity responses. Previous reports suggested that cis-UCA was immunomodulatory through its effects on keratinocytes, Langerhans cells, fibroblasts, T lymphocytes, natural killer cells and monocytes/macrophages. As dermal mast cells were recently demonstrated to be critical to UVB-induced systemic suppression of certain delayed-type and contact hypersensitivity responses, we investigated whether they were involved in the processes by which cis-UCA was immunomodulatory. Not only was there a correlation between dermal mast cell prevalence and the degree of susceptibility of different strains of mice to the immunomodulatory effects of cis-UCA, there was also a functional link. Mast cell-depleted Wf/Wf mice were rendered susceptible to immunomodulation by cis-UCA injected subcutaneously only after their dorsal skin had been reconstituted with bone marrow-derived mast cell precursors. These studies suggest that mast cells are critical to the processes by which cis-UCA suppresses systemic contact hypersensitivity responses to the hapten, trinitrochlorobenzene, in mice.  相似文献

Folded polymers are used in Nature for virtually every vital process. Nonnatural folded polymers, or foldamers, have the potential for similar versatility, and the design and refinement of such molecules is of considerable current interest. Here we report a complete and systematic analysis of the relationship between side chain structure and the 14-helicity of a well-studied class of foldamers, beta(3)-peptides, in water. Our experimental results (1) verify the importance of macrodipole stabilization for maintaining 14-helix structure, (2) provide comprehensive evidence that beta(3)-amino acids branched at the first side chain carbon are 14-helix-stabilizing, (3) suggest a novel role for side chain hydrogen bonding as an additional stabilizing force in beta(3)-peptides containing beta(3)-homoserine or beta(3)-homothreonine, and (4) demonstrate that diverse functionality can be incorporated into a stable 14-helix. Gas- and solution-phase calculations and Monte Carlo simulations recapitulate the experimental trends only in the context of oligomers, yielding insight into the mechanisms behind 14-helix folding. The 14-helix propensities of beta(3)-amino acids differ starkly from the alpha-helix propensities of analogous alpha-amino acids. This contrast informs current models for alpha-helix folding, and suggests that 14-helix folding is governed by different biophysical forces than is alpha-helix folding. The ability to modulate 14-helix structure through side chain choice will assist rational design of 14-helical beta-peptide ligands for macromolecular targets.  相似文献

A mass spectrometry analysis has been performed on complex architecture polymeric material produced during reversible addition fragmentation chain transfer (RAFT) polymerizations yielding star polymers. Para‐acetoxystyrene (AcOSty) has been polymerized at 60 °C, using azobisisobutyronitrile (AIBN) as the thermally decomposing initiator, in the presence of the R‐group approach tetrafunctional RAFT agent (1,2,4,5‐tetrakis‐(2‐phenyl‐thioacetyl‐sulfanylmethyl)‐benzene). In addition to ideal star material, a variety of products unique to this mode of polymerization have been identified. These include star–star couples, stars terminated with initiator fragments, star–star couples terminated with initiator fragments and linear polymers, supporting the notion that these species are responsible for the structured molecular‐weight distributions measured for these systems when analyzed via gel permeation chromatography. The analysis begins with a study of AcOSty polymerizing (i) in the absence of any mediating agent and (ii) in the presence of a monofunctional RAFT agent, revealing the mode of termination of propagating poly(AcOSty) radicals as combination and that some ionization biases exist among variants of poly (AcOSty). The interpretation of the mass spectrometry data has been aided by a novel kinetic model of star polymerizations, allowing the rationalization of experimental observations with theoretical expectations. © 2008 Wiley Periodicals, Inc.
