Isothermal crystallization studies of low-density polyethylene (LDPE) and high-density polyethylene (HDPE) using differential scanning calorimetry (DSC) were performed using different sample thicknesses to determine the effect of non-ideal heat-transfer. Polyethylene was chosen because of its importance, its extensive coverage in the literature, and its fast crystallization kinetics. Thermal gradients were found to significantly affect the measured crystallization exotherm; slower crystallization rates were observed for thicker samples measured at lower temperatures (greater supercoolings). Differences between different sample thicknesses disappeared at higher temperatures, consistent with finite heat-transfer rates being responsible for the effect. A power-compensation and a heat-flux DSC were used; these experiments also enabled the determination that the performance of the latter was acceptable for this study. Finally, thickness-independent Avrami parameters have been calculated.  相似文献

Continuous outbreaks of avian influenza (AI) in recent years with increasing threat to animals and human health have warranted the urgent need for rapid detection of pathogenic AI viruses. In this study, an impedance immunosensor based on an interdigitated array (IDA) microelectrode was developed as a new application for sensitive, specific and rapid detection of avian influenza virus H5N1. Polyclonal antibodies against AI virus H5N1 surface antigen HA (Hemagglutinin) were oriented on the gold microelectrode surface through protein A. Target H5N1 viruses were then captured by the immobilized antibody, resulting in a change in the impedance of the IDA microelectrode surface. Red blood cells (RBCs) were used as biolabels for further amplification of the binding reaction of the antibody-antigen (virus). The binding of target AI H5N1 onto the antibody-modified IDA microelectrode surface was further confirmed by atomic force microscopy. The impedance immunosensor could detect the target AI H5N1 virus at a titer higher than 103 EID50/ml (EID50: 50% Egg Infective Dose) within 2 h. The response of the antibody-antigen (virus) interaction was shown to be virus titer-dependent, and a linear range for the titer of H5N1 virus was found between 103 and 107 EID50/ml. Equivalent circuit analysis indicated that the electron transfer resistance of the redox probe [Fe(CN)6]3−/4− and the double layer capacitance were responsible for the impedance change due to the protein A modification, antibody immobilization, BSA (bovine serum albumin) blocking, H5N1 viruses binding and RBCs amplification. No significant interference was observed from non-target RNA viruses such as Newcastle disease virus and Infectious Bronchitis disease virus. Background  

This study compared the transduction efficiencies of an adeno-associated viral (AAV) vector, which was pseudotyped with an AAV1 capsid and encoded the green fluorescent protein (GFP), with a lentiviral (LV) vector, which was pseudotyped with a VSV-G envelop and encoded the discosoma red fluorescent protein (dsRed), to investigate which viral vector transduced the lateral hypothalamus or the amygdala more efficiently. The LV-dsRed and AAV1-GFP vector were mixed and injected into the lateral hypothalamus or into the amygdala of adult rats. The titers that were injected were 1 × 108 or 1 × 109 genomic copies of AAV1-GFP and 1 × 105 transducing units of LV-dsRed.  相似文献

The double proton transfer in the formamide dimer is characterized computationally by combining density functional theory and ab initio methods. The intrinsic reaction coordinate (IRC) is obtained at the B3LYP level of theory. Energies of several points along the IRC are treated by the more rigorous focal point method to test the validity of the B3LYP functional. The reaction mechanism is examined in terms of the energy profile, the reaction force, the chemical potential, and the reaction electronic flux. The energy profile for the activation process of the formamide dimer to the imino ether product obtained with the B3LYP functional is in agreement with the results of the focal point method. Together with the reaction force analysis and the reaction electronic flux a precise assignment of the structural and electronic contributions to the activation barrier becomes possible. The results show that the reaction starts with a structural rearrangement, where the two dimers approach each other, and is followed by electronic changes before the system reaches the transition state. This electronic contribution to the activation barrier steers the activation process. After the transition state is reached, deviations of the B3LYP functional from the more accurate focal point energies become apparent, where the errors may be rationalized in terms of the treatment of exchange. The inconsistency could be assigned to the incapacity of the functional to describe delocalization effects over the whole system.  相似文献

The objective of this study was to determine if topical anesthesia to the larynx and pharynx affects vocal fold motion during dynamic voice evaluation with transnasal flexible endoscopy. Transnasal dynamic laryngeal examinations of 10 patients with no voice complaints were evaluated by five blinded fellowship-trained laryngologists. Each patient was examined before and after application of topical anesthetic. Reviewers rated briskness of right and left vocal fold movement and longitudinal tension on a visual analogue scale. Statistical comparisons were made between individual subject scores before and after anesthetic application. Inter-rater reliability was also assessed. No statistical difference was observed between subject scores before and after anesthetic application. Average intraclass correlation coefficients were 0.643 and 0.591 for pre- and postanesthesia scores, respectively. Application of topical anesthesia to the larynx and pharynx does not affect vocal fold motion.  相似文献

Uric acid, 3-ribosyluric acid, 1-methyluric acid, 1,3,7-trimethyluric acid, 2,8-dihydroxyadenine, and 2-imidazolone reacted with galvinoxyl, a stable free radical, to form galvinol. The reaction was followed by the decrease in absorbance at 550 nm of an ethanolic solution of galvinoxyl. Related compounds such as inosine, xanthosine, 2-hydroxybenzimidazole, lumichrome, and lumazine did not react with galvinoxyl. The data reported support the proposal that urates are important antioxidants and free-radical scavengers.  相似文献
