We report a multichannel surface plasmon resonance (SPR) sensor for detection of thrombin via DNA aptamers immobilized on the SPR sensor surface. A detailed investigation of the effect of the immobilisation method on the interaction between thrombin and DNA aptamers is presented. Three basic approaches to the immobilisation of aptamers on the surface of the SPR sensor are examined: (i) immobilisation based on chemisorption of aptamers modified with SH groups, (ii) immobilisation of biotin-tagged aptamers via previously immobilized avidin, neutravidin or streptavidin molecular linkers, and (iii) immobilisation employing dendrimers as a support layer for subsequent immobilisation of aptamers. A level of nonspecific binding of thrombin to immobilized human serum albumin (HSA) for each of the immobilisation methods is determined. Immobilisation of aptamers by means of the streptavidin–biotin system yields the best results both in terms of sensor specificity and sensitivity.  相似文献

Separations of metal cations on a column packed with the strongly acidic cation exchanger Separon SGX CX were investigated in the presence of -hydroxyisobutyric acid (HIBA) in the mobile phase. A retention model based on the general theory of side equilibria was elaborated and relations describing dependences of capacity factors of analytes on the compositon of the mobile phase were derived. Effects of HIBA concentration and pH of the mobile phase on the analyte retention were studied in detail. Stability constants of divalent metal cations (Cd2+, Co2+, Mn2+, Ni2+ and Zn2+) with HIBA were calculated from the experimental dependences of the reciprocal values of capacity factors on the ligand concentration.  相似文献

We study involutions in the Riordan group, especially those with combinatorial meaning. We give a new determinantal criterion for a matrix to be a Riordan involution and examine several classes of examples. A complete characterization of involutions in the Appell subgroup is developed. The present study advances a theoretical and experimental investigation of the frost growth and densification on flat surfaces. This study focuses on the most important factors affecting the frost formation process, i.e. the surrounding air temperature, humidity and velocity, and the surface temperature. The processes of frost growth and densification were investigated experimentally in order to provide a physical basis for the development of a theoretical model to predict the variation of the frost layer thickness and mass with time. The mathematical model was based on mass and energy balances within the frost layer, assuming the frost as a porous medium and accounting for the supersaturation of the moist air on the frost surface. The governing equations for mass and heat diffusion were integrated analytically, giving rise to a semi-algebraic formulation which requires numerical integration of only one time dependent ordinary differential equation. When compared with experimental data, the model predictions of the frost thickness as a function of time agreed to within ±10% error bands. The experimentally-validated model was then used to predict the frost layer growth and densification with respect to the operation conditions such as plate surface temperature, air stream temperature, humidity and velocity.  相似文献

We prove that if D=(g(x),f(x)) is an element of order 2 in the Riordan group then g(x)=±exp[Φ(x,xf(x)] for some antisymmetric function Φ(x,z). Also we prove that every element of order 2 in the Riordan group can be written as BMB-1 for some element B and M=(1,-1) in the Riordan group. These proofs provide solutions to two open problems presented by L. Shapiro [L.W. Shapiro, Some open questions about random walks, involutions, limiting distributions and generating functions, Adv. The compounds diethyl-N-carbazolylmethylphosphonate 1 and diethyl-2-oxymethylphosphonate carbazole 2 have been synthesised and characterized by a range of techniques including NMR, absorption and emission spectroscopy, infrared, and mass spectroscopy. Compound 1 forms a 2:1 complex 3 with calcium nitrate and the single crystal X-ray diffraction structure of 3 is described. Titrations of 1 with Zn2+ and Ca2+ in ethanol reveal that the intrinsic fluorescence is only slightly perturbed in the presence of these metal ions in micromolar ethanol solutions. Compounds 1 and 2 are readily taken up and visualized in L929 fibrosarcoma and DRG (Dorsal root ganglia) cell lines.  相似文献

Silver-ion high-performance liquid chromatography (HPLC) coupled to atmospheric pressure chemical ionization mass spectrometry (APCI-MS) is used for the regioisomeric analysis of triacylglycerols (TGs). Standard mixtures of TG regioisomers are prepared by the randomization reaction from 8 mono-acid TG standards (tripalmitin, tristearin, triarachidin, triolein, trielaidin, trilinolein, trilinolenin and tri-gamma-linolenin). In total, 32 different regioisomeric doublets and 11 triplets are synthesized, separated by silver-ion HPLC using three serial coupled chromatographic columns giving a total length of 75cm. The retention of TGs increases strongly with the double bond (DB) number and slightly for regioisomers having more DBs in sn-1/3 positions. DB positional isomers (linolenic vs. γ-linolenic acids) are also separated and their reverse retention order in two different mobile phases is demonstrated. APCI mass spectra of all separated regioisomers are measured on five different mass spectrometers: single quadrupole LC/MSD (Agilent Technologies), triple quadrupole API 3000 (AB SCIEX), ion trap Esquire 3000 (Bruker Daltonics), quadrupole time-of-flight micrOTOF-Q (Bruker Daltonics) and LTQ Orbitrap XL (Thermo Fisher Scientific). The effect of different types of mass analyzer on the ratio of [M+H-R(i)COOH](+) fragment ions in APCI mass spectra is lower compared to the effect of the number of DBs, their position on the acyl chain and the regiospecific distribution of acyl chains on the glycerol skeleton. Presented data on [M+H-R(i)COOH](+) ratios measured on five different mass analyzers can be used for the direct regioisomeric determination in natural and biological samples.  相似文献
