Magnetic poly(N‐propargylacrylamide) (PPRAAm) microspheres were prepared by the precipitation polymerization of N‐propargylacrylamide (PRAAm) in a toluene/propan‐2‐ol medium in the presence of magnetic nanoparticles (oleic acid‐coated Fe3O4). The effects of several polymerization parameters, including the polarity of the medium, polymerization temperature, the concentration of monomer, and the amount of magnetite (Fe3O4) in the polymerization feed, were examined. The microspheres were characterized in terms of their morphology, size, particle‐size distribution, and iron content using transmission and scanning electron microscopies (TEM and SEM) and atomic absorption spectroscopy (AAS). A medium polarity was identified in which magnetic particles with a narrow size distribution were formed. As expected, oleic acid‐coated Fe3O4 nanoparticles contributed to the stabilization of the polymerized magnetic microspheres. Alkyne groups in magnetic PPRAAm microspheres were detected by infrared spectroscopy. Magnetic PPRAAm microspheres were successfully used as the anchor to enable a “click” reaction with an azido‐end‐functionalized model peptide (radiolabeled azidopentanoyl‐GGGRGDSGGGY(125I)‐NH2) and 4‐azidophenylalanine using a Cu(I)‐catalyzed 1,3‐dipolar azide‐alkyne cycloaddition reaction in water. © 2011 Wiley Periodicals, Inc. The supramolecular interactions between a Bradykinin Potentiating Peptide (BPP10c) and β-cyclodextrin (βCD) have been investigated by using several techniques. These new properties acquired by the inclusion phenomena are important in developing a strategy for pharmaceutical formulation. The BPP10c structural elucidation and its inclusion complex formed have been investigated using Nuclear Magnetic Resonance techniques. The peptide secondary structure was investigated using infrared spectroscopy in solution, Circular Dichroism and NMR. In addition, the thermodynamic parameters of the inclusion process were also evaluated using Isothermal Titration Calorimetry. The results obtained by these physical–chemical techniques suggested a 1:1 complex formed by interaction between the Tryptophan amino acid residue and the βCD cavity. The peptide secondary structure was not substantially modified for the inclusion process. In addition, the inclusion process proved to be spontaneous (ΔGº = ?2.53 kcal mol?1), with an enthalpy reduction (ΔHº = ?3.72 kcal mol?1) and a favored entropic variation (TΔSº = ?1.19 kcal mol?1).  相似文献

A new three‐component cyclisation reactions of methyl 3,3,3‐trifluoropyruvate, 2‐aminobenzylamine and oxo compounds afforded tetrahydropyrroloquinazolinones of the types 4 and 5 as mixtures of regio‐ and stereoisomers. Whereas standard 1D NMR spectroscopy was used for a facile assignment of the cyclization regioisomers, a combination of homo (proton–proton) and heteronuclear (proton–fluorine) NOE experiments allowed the determination of the relative configuration on stereogenic centres. The structure of some compounds was also confirmed by the X‐ray diffraction. Adaptation of the 1D double‐pulsed field‐gradient spin‐echo NOE for a heteronuclear case is presented. In the present work are performed the TG and DSC study of gallstone samples collected from patients submitted to cholecystectomy at the Hospital Universitário Onofre Lopes of Universidade Federal do Rio Grande do Norte, in Natal, Rio Grande do Norte State, Brazil. The gallstone samples were characterized by elemental analysis and infrared spectroscopy. Standard cholesterol was used for all analysis as comparison with the gallstones. Is verified that cholesterol is the main chemical component of all studied gallstone samples. The thermoanalytical study was performed on air and nitrogen atmospheres. The presence of cholesterol as the main chemical component of all gallstone samples is confirmed by TG and DSC results.  相似文献

Properties of beta-glucosidase produced by Aspergillus niger URM 6642 recently isolated from the Atlantic rainforest biome and its potential tolerance to saccharification of lignocellulosic biomass products and fermentation inhibitors was evaluated. The fungus was cultivated under solid state culture conditions at 37°C with different agro-industrial wastes. High levels of beta-glucosidase (3778.9 U g?1)from A. niger were obtained with rice meal as substrate under solid state culture conditions after ten days. Optimum pH for this particular beta-glucosidase activity was 4.0 although it was stable in the range of 4.0 to 7.0. The half-life (T½) of beta-glucosidase at 55°C is 3 h. However, at the optimum temperature of the enzyme, 65°C, T½ is 20 min. The enzyme showed tolerance to various compounds such as glucose, xylose, 5-hydroxymethyl furfural, furfural, coumarin, ethanol and acetic acid. Therefore, beta-glucosidase from the novel A. niger species may be of potential use in the saccharification of lignocellulosic biomass, as well as an additional enzyme supplement in cellulase cocktails used to increase the yield of fermentable sugars.  相似文献

Nickel(II) complexes with 2,3-dihydroxybenzaldehyde N4-substituted thiosemicarbazone ligands (H3L1–H3L4) have been synthesized and characterized with the aim of evaluating the effect of N4 substitution in the thiosemicarbazone moiety on their coordination behavior and biological activities. Two series of nickel(II) complexes with the general formulae [Ni(H3L)(H2L)]ClO4 and [Ni2(HL)2] were characterized by analytical and spectral techniques. The molecular structure of one of the complexes, namely, [Ni(H3L4)(H2L4)]ClO4 was established by single crystal X-ray diffraction studies. The crystal structure of this complex revealed that two H3L4 ligands are coordinated to nickel(II) in different modes; one as a neutral tridentate ONS ligand and the other is as a monoanionic tridentate (ONS?) ligand. The antimicrobial activities of the compounds were tested against 25 bacterial strains via the disc diffusion method, and their minimum inhibitory concentration (MIC) and minimum microbicidal concentration were evaluated using microdilution methods. With a few exceptions, most of the compounds exhibited low-to-moderate inhibitory activities against the tested bacterial strains. However, the complexes [Ni2(HL3)2] (7) and [Ni2(HL4)2] (8) indicated higher inhibitory activity against Salmonella enterica ATCC 9068 (MIC values 15.7 and <15.7 μg/ml, respectively), compared with gentamicin as the positive control (MIC 25 μg/ml). Complex (7) also inhibited Streptococcus pneumoniae more efficiently (MIC 31.2 μg/ml), compared with gentamicin (MIC > 50 μg/ml). The toxicities of the compounds were tested on brine shrimp (Artemia salina), where no meaningful toxicity level was noted for both the free ligands and the complexes. The cytotoxicities of the compounds on cell viability were determined on MCF7, PC3, A375, and H413 cancer cells in terms of IC50; complexes [Ni(H3L3)(H2L3)]ClO4 (3), [Ni2(HL3)2] (7) and [Ni2(HL4)2] (8) exhibited significant cytotoxicity on the tested cell lines.  相似文献

When quantifying information in metabolomics, the results are often expressed as data carrying only relative information. Vectors of these data have positive components, and the only relevant information is contained in the ratios between their parts; such observations are called compositional data. The aim of the paper is to demonstrate how partial least squares discriminant analysis (PLS‐DA)—the most widely used method in chemometrics for multivariate classification—can be applied to compositional data. Theoretical arguments are provided, and data sets from metabolomics are investigated. The data are related to the diagnosis of inherited metabolic disorders (IMDs). The first example analyzes the significance of the corresponding regression parameters (metabolites) using a small data set resulting from targeted metabolomics, where just a subset of potential markers is selected. The second example—the approach of untargeted metabolomics—was used for the analysis detecting almost 500 metabolites. The significance of the metabolites is investigated by applying PLS‐DA, accommodated according to a compositional approach. The significance of important metabolites (markers of diseases) is more clearly visible with the compositional method in both examples. Although cyclometalated IrIII complexes have emerged as promising photosensitizers for photodynamic therapy, some key drawbacks still hamper clinical translation, such as operability in the phototherapeutic window and reactive oxygen species (ROS) production efficiency and selectivity. In this work, a cyclometalated IrIII complex conjugated to a far‐red‐emitting coumarin, IrIII–COUPY, is reported with highly favourable properties for cancer phototherapy. IrIII–COUPY was efficiently taken up by HeLa cells and showed no dark cytotoxicity and impressive photocytotoxicity indexes after irradiation with green and blue light, even under hypoxia. Importantly, a clear correlation between cell death and intracellular generation of superoxide anion radicals after visible light irradiation was demonstrated. This strategy opens the door to novel fluorescent photodynamic therapy agents with promising applications in theragnosis.  相似文献

Preparation of affinity polyacrylamide gels containing immobilized Fe(III) ions for the separation of proteins exhibiting metal ion binding properties is described. The presented method enables uniform distribution of immobilized metal ions in the affinity part of the polyacrylamide separating gel. Affinity gels prepared by this way are suitable to follow the effect of different concentrations of metal ions immobilized in polyacrylamide gel on a protein electrophoretic behavior. Polyacrylamide gels containing immobilized Fe(III) ions were used to study the electrophoretic behavior of two model proteins differing in their phosphate group content: chicken ovalbumin and bovine α‐casein. For the electrophoretic separation, both the native and the denaturating conditions were used.  相似文献

An efficient synthesis of three novel stigmasterol-amino acid (glycine, L-leucine and L-phenylalanine) conjugates as stimuli responsive gelators is reported. The gelation properties of the prepared compounds were investigated in a variety of organic as well as aqueous solvents. The most striking finding of our investigation was that the hydrochloride salts of the prepared conjugates acted as gelators, whereas the neutral conjugates were either non-gelators or formed only a weak gel in anisole. The hydrochloride salts of stigmasteryl glycinate and L-leucinate form gels in n-alcohols (n=4-10) and in ethane-1,2-diol, and that of stigmasteryl L-phenylalaninate forms gels in aromatic solvents and in tetrachloromethane. These unique properties of the gelators were explored to prepare stimuli responsive, "acid-base" triggered reversible sol-gel transitions. The gelators and their gels were characterized by liquid and solid-state NMR as well as FT-IR. The morphology of their corresponding xerogels was investigated by SEM.  相似文献
