Soluble guanylate cyclase(sGC) is a critical heme-containing enzyme involved in NO signaling.The dimerization of sGC subunits is necessary for its bioactivity and its mechanism is a striking and an indistinct issue.The roles of heme domain cysteines of the sGC on the dimerization and heme binding were investigated herein.The site-directed mutations of three conserved cysteines(C78A,C122A and C174S) were studied systematically and the three mutants were characterized by gel filtration analysis,UV-vis spectroscopy and heme transfer examination.Cys78 was involved in heme binding but not referred to the dimerization,while Cys174 was demonstrated to be involved in the homodimerization.These results provide new insights into the cysteine-related dimerization regulation of sGC.  相似文献

The effect of a gel polymer electrolyte (GPE) as the redox electrolyte used in dye-sensitized solar cells was studied. A GPE solution consisting of 0.5?M sodium iodide, 0.05?M iodine, and ethylene carbonate/propylene carbonate (1:1 w/w) binary solvents was mixed with increasing amounts of styrene–acrylonitrile (SAN). Bulk conductivity measurements show a decreasing trend from 4.54 to 0.83×10?3?S?cm?1 with increasing SAN content. The GPE exhibits Newtonian-like behavior and its viscosity increases from 0.041 to 1.093?Pa?s with increasing SAN content. A balance between conductivity (1.3?×?10?3?S?cm?1) and viscosity (1.4?Pa?s) is observed at 19?wt.% SAN. Fourier transform infrared spectroscopy detects elevated ring torsion at 706?cm?1 upon the addition of SAN into the liquid electrolyte. This indicates that SAN does not bond with the liquid electrolyte. Finally, the potential stability window of 19?wt.% SAN, which ranges from ?1.68 to 1.38?V, proves its applicability in solar cells.  相似文献

The titania monolith column has been synthesized through a template-free sol–gel route, and a simple and reliable method for the determination of benzoic acid by hydrophilic interaction liquid chromatography using the prepared titania monolith has been developed. The influences of acetonitrile, acetate buffer and buffer pH on the retention of benzoic acid were investigated. Benzoic acid in carbonated drinks and fruit beverages samples were determined within 5 min and quantitative analysis was carried out by external standard method with a correlation coefficient (R 2) of 0.9984. The relative standard deviation was 0.91 % and the recovery ranged from 92.5 to 101.3 %. The proposed method is suitable for the analysis of benzoic acid in beverage samples.

Myricetin, a naturally occurring flavonol, shows multifarious pharmacological activities, e.g., antidiabetic, antioxidant, anti-inflammatory, antitumor, and liver protection effects. In order to obtain an understanding of the myricetin’s metabolism in vivo, a rapid and sensitive method by high-performance liquid chromatography coupled with electrospray-ionization mass spectrometry (HPLC-MSn) techniques was employed to investigate the biotransformation in rats after oral administration of myricetin. Recognition and structural exposition of the metabolites were operated by comparing the changes in molecular mass (ΔM) and MSn spectra with the parent drug. As a result, the parent compound and seven metabolites were found in rat plasma, urine, and feces. In addition, besides 3,5-dihydroxyphenylacetic acid (M1) and 3,4,5-trihydroxyphenylacetic acid (M2), five other compounds were first discovered in the metabolite research of myricetin. These results indicated that, besides ring-fission, there were methylate (M3, M4, M5) and glucuronide (M6, M7) biotransformations of myricetin occurring in vivo.

A series of novel low band gap polymers containing conjugated side chains with 4,7‐dithien‐5‐yl‐2,1,3‐benzodiathiazole and different electron‐withdrawing end groups of aldehyde ( PT‐DTBTCHO ), 2‐ethylhexyl cyanoacetate ( PT‐DTBTCN ), 1,3‐diethyl‐2‐thiobarbituric acid ( PT‐DTBTDT ), and electron‐donating end group of 2‐methylthiophene ( PT‐DTBTMT ) have been designed and synthesized. All polymers exhibit good solubility in common organic solvents, film‐forming ability, and thermal stability. These conjugated polymers show the broad ultraviolet‐visible absorption and the narrow optical band gaps in the range of 1.65–1.90 eV. Through changing the end group of conjugated side chains, the photophysical properties and energy levels of the polymers were tuned effectively. Bulk heterojunction solar cells based on the blend of these polymers and (6,6)‐phenyl‐C61‐butyric acid methyl ester (PC61BM) reached the best power conversion efficiency (PCE) of 2.72%. © 2012 Wiley Periodicals, Inc.
