LC-electron capture-APCI(-)-MS determination of nitrobenzoxadiazole derivatives

Nitrobenzoxadiazole (NBD) derivatives are determined with limits of detection ranging down to 20 nmol l(-1) using liquid chromatography-mass spectrometry (LC-MS) with electron capture (EC) ionisation. An atmospheric pressure chemical ionisation (APCI) interface operated in the negative ion mode is used as ionisation source. Amine derivatives of 4-chloro-7-nitro-2,1,3-benzoxadiazole (NBDCl) as well as the isocyanate derivatives of 4-nitro-7-piperazino-2,1,3-nitrobenzoxadiazole (NBDPZ) have been analysed using this technique. The parameters favouring electron capture mechanisms have been investigated thoroughly under consideration of the competing mechanism of deprotonation to allow a better understanding of the electron capture process and to improve selectivity of the analysis.     

New approach to calibrating conductivity meters in the low conductivity range

There is currently a major issue with the calibration of conductivity meters used for high purity water: the lack of availability of a reference material or reference methods for low conductivity ranges (conductivity below 1 S cm^–1 at 25.0 °C, resistivity >1 M cm at 25.0 °C). This paper describes the current status of conductivity measurements in high purity water. A new and improved approach, currently being investigated, should allow us to make the calibration of conductivity meters used for low conductivity ranges traceable to the SI.Milipore, Milli-Q and Elix are registered trademarks of Millipore Corporation.     

Identification of the function of gene lndM2 encoding a bifunctional oxygenase-reductase involved in the biosynthesis of the antitumor antibiotic landomycin E by Streptomyces globisporus 1912 supports the originally assigned structure for landomycinone

The angucycline antibiotic family of the landomycins displays potent antitumor activity. To elucidate early post polyketide synthase (PKS) tailoring steps of the landomycin E biosynthetic pathway in Streptomyces globisporus 1912, the mutant S. globisporus M12 was prepared through gene replacement experiment of lndM2. It encodes an enzyme with putative oxygenase and reductase domains, according to sequencing of the gene and its counterpart lanM2 from S. cyanogenus S136 landomycin A biosynthetic gene cluster. The isolation of the novel shunt products 11-hydroxytetrangomycin and 4-hydroxytetrangomycin along with the well-known angucyclines tetrangomycin and tetrangulol from the culture of S. globisporus M12 provides evidence for the involvement of lndM2 in the early biosynthetic pathway of the landomycins, in particular in the formation of the alicyclic 6-hydroxy function of the landomycin aglycon. We therefore propose LndM2 to be responsible for both hydroxylation of the 6-position and its subsequent reduction. These reactions are necessary before the glycosylation reactions can occur. The results are in agreement with the originally published structure of landomycin but do not support the recently suggested revised structure.     

Über ternäre Thalliumchalkogenide mit Thalliumselenidstruktur

On Ternary Thallium Chalcogenides with Thallium Selenide Structure. By means of single crystal investigations, it has been confirmed that the compounds TlGaTe₂, TIInSe₂, and TIInTe₂ crystallize in the TISe type (B37). As an univalent cation thallium is surrounded by eight chalcogen atoms. Gallium and indium are trivalent with tetrahedral coordination and covalent bonds. The interatomic distances as well as coordination and bonding resulting from them are discussed.     

Determination of the activity of diamine oxidases, urease and histidine ammonia-lyase by enzymatic assay of ammonia

Summary The activities of diamine oxidases (DAO) from submaxillary glands, parotid glands and kidneys, from human serum during the pregnancy and from pig serum and of DAO from pea seedlings and the activities of urease and histidine ammonia-lyase have been determined by measuring NH₄ ⁺ released from the corresponding substrates. Ammonia has been determined with the aid of glutamate dehydrogenase reaction in the coupled optic test. The specifity and sensitivity of the method, by which still 3×10^–9 Mol of NH₄ ⁺ could be measured, is excellent, the performance simple and rapid. The sensitivity could be enhanced by fluorometric determination of NAD⁺ in strong alkali solution. A method for the preparation of tissues for the spectrophotometric test of DAO is described. The importance of DAO determination in different diseases is briefly discussed.

Zusammenfassung Die Aktivität von Diaminoxydasen aus Speicheldrüsen und Nieren, aus menschlichem Serum während der Schwangerschaft und Schweineserum und aus Erbsenkeimlingen sowie die Aktivität der Urease und Histidase wurden anhand der NH₃-Abspaltung aus ihren entsprechenden Substraten bestimmt. Das entstehende Ammoniak wurde dabei mit Hilfe der Glutamatdehydrogenasereaktion im gekoppelten optischen Test gemessen. Spezifität und Empfindlichkeit der Methode, mit der noch 3 · 10^–9 Mol NH₄ ⁺ bestimmt werden können, sind ausgezeichnet, die Durchführung einfach und rasch. Die Empfindlichkeit konnte außerdem noch durch die fluorimetrische Bestimmung von NAD⁺ gesteigert werden. Für die Bestimmung der Diaminoxydasen im Gewebe wird eine Methode der Aufarbeitung des Gewebes angegeben, die praktisch keinen Verlust an Gesamtaktivität gegenüber dem Ausgangshomogenat aufweist. Die Bedeutung der Diaminoxydasebestimmung für die Klinik wird kurz diskutiert.

Markierungsexperimente zur fragmentierung von 1,2,3-thiadiazolen

The generation of thiirenes in the thermal and photochemical nitrogen elimination of 1,2,3-thiadiazoles is discussed in relation to oxirene formation. Isotope labelling experiments were performed with 4-phenyl-[4-¹³C]-1,2,3-thiadiazole ( 7 ) as the substrate.     

Reduction of 1,4-dichlorobut-2-yne by titanocene to a 1,2,3-butatriene. Formation of a 1-titanacyclopent-3-yne and a 2,5-dititanabicyclo[2.2.0]hex-1(4)-ene

The 2,5-dititanabicyclo[2.2.0]hex-1(4)-ene (bis-titanocene-mu-(Z)-1,2,3-butatriene complex)3 is formed starting from [Cp2Ti(eta2-Me3SiC2SiMe3)] by in situ generated titanocene and 1,4-dichlorobut-2-yne via the 1-titanacyclobut-3-yne (2).
