The second member of the family of single-isomer sulfated alpha-cyclodextrins, the sodium salt of hexakis(6-O-sulfo)-alpha-cyclodextrin (HxS), has been synthesized, analytically characterized, and used as the resolving agent for the capillary electrophoretic separation of the enantiomers of nonionic, weak-acid and weak-base analytes present in our initial screening kit. HxS interacted less strongly with many of the analytes tested than the larger-ring analogs, heptakis(6-O-sulfo)-beta-cyclodextrin (HS) and octakis(6-O-sulfo)-gamma-cyclodextrin (OS). For some of the analytes, the separation selectivities obtained with HxS were complementary to those observed with hexakis(2,3-di-O-acetyl-6-O-sulfo)-alpha-cyclodextrin (HxDAS), HS, and OS. For all analytes, the effective mobilities and separation selectivities as a function of the background electrolyte concentration of HxS followed the trends that were found for HxDAS, HS, and OS.  相似文献

The second member of the single-isomer, sulfated gamma-cyclodextrin family, the sodium salt of octa(6-O-sulfo)-gamma-cyclodextrin (OS) has been synthesized, characterized and used to separate the enantiomers of nonelectrolyte, acidic, basic, and ampholytic analytes by capillary electrophoresis in acidic aqueous background electrolytes. The anionic effective mobilities of the nonelectrolyte and anionic analytes increased with increasing concentration of OS. The effective mobilities of strongly complexing cationic analytes became anionic with very low OS concentrations and passed local anionic effective mobility maxima as the OS concentration, and along with it, the ionic strength, of the background electrolyte increased. The effective mobilities of the weakly binding cationic analytes became only slightly anionic at high OS concentration values and did not show the local anionic effective mobility maxima. For nonelectrolyte analytes, separation selectivities decreased with increasing OS concentration. For cationic analytes, separation selectivities were highest where the effective mobilities of the less mobile enantiomers approached zero. OS proved to be a broadly applicable chiral resolving agent.  相似文献

Our recent extended peak resolution equation of capillary electrophoresis has been combined with the multiple equilibria-based electrophoretic mobility model of chiral separations to describe peak resolution as a function of the composition of the background electrolyte (pH and the β-cyclodextrin concentration) and a function of the operating variables (effective portion of the applied potential, dimensionless electroosmotic flow coefficient). Using the previously determined model parameters, the resolution surfaces were calculated for a Type I chiral separation (ibuprofen), and a Type III chiral separation (homatropine). In Type I separations resolution can be obtained only over a narrow pH range in the vicinity of the pKa value, and above a minimum value, the concentration of β-cyclodextrin plays a lesser role. In Type III separations, the pH- and β-cyclodextrin concentration-dependent resolution surface has two lobes, on which the migration order of the enantiomers is opposite. This can be an advantage in trace component analysis. In both Type I and Type III separations, peak resolution varies strongly with the dimensionless electroosmotic flow coefficient when its value is changed in the − 1 to 1 range. The loci of the pH-dependent and the β-cyclodextrin concentration-dependent resolution maxima do not shift significantly when the dimensionless electroosmotic flow coefficient is changed. This fact provides the analyst with an additional resolution enhancement tool that does not alter the selectivity of the separation. The utility of the model and its theoretical predictions has been demonstrated by comparing measured and calculated Rs values for ibuprofen and homatropine.  相似文献

Acylation of the N-substituted diethyl aminomalonates 3a3d with diketene furnished the ring tautomers 6a6d of the expected acetoacetyl derivatives 5. By treatment with iodine and sodium ethoxide compounds 6a6d are smoothly converted into the β-lactam derivatives 2a2d. Deethoxycarbonylation of the ethylene ketals 7a7d of the latter furnishes mixtures of the corresponding diastereomeric monoesters 8 and10. The ethoxycarbonyl groups of the trans esters 8 are more reactive than those of the cis isomers 10. This permits, under appropriate conditions, selective alkaline hydrolysis and NaBH4 reduction of the trans esters 8 in the presence of the cis esters 10. Reduction of the cis ester 10c under more forceful conditions furnishes the trans hydroxymethyl derivative 11c.  相似文献

trans-Perhydro-1,4-benzoxazepin-3-ones 2a-c were synthesized and transformed to condensed-skeleton perhydro-trans-1,4-benzoxazepines 3a,b , the thiones 4a,b , the urea derivatives 5a,b , and N-acylated compounds 6a-e . Compounds 6b,d were ring-opened by hydrochloric acid in ethanol to yield trans-2-(1-carbethoxyethoxy)-1-acylaminomethylcyclohexane derivatives 7b,d . The 1H- and 13C-nmr investigation and X-ray analysis of 5b and 6c,d proved that the expected N-acylated derivatives were formed and that both rings of the trans anellated compounds have a chair conformation.  相似文献

The conformational properties of diastereomeric P-modified nucleotides are reported as reflected by different NMR parameters. Some conformational trends can be rationalized by consideration of the 3J(C4,P) and 3J(C2,P) coupling values of the isomers and the nature of the substituent on the phosphorus. Configurational assessment of the phosphorus is inferred from NOE experiments. The effects of temperature, solvent and size of substituents are presented.  相似文献

8-Aza-12-oxasteroids have been synthesized in a simple three-step process. Diesel exhaust is one of the major sources of fine and ultrafine particulate matter in urban air. Toxicity of diesel-powered engine emissions has been quite widely assessed, however, much less information is available on their ecotoxicity. In our study the kinetic version of the Vibrio fischeri bioluminescence inhibition bioassay, based on the ISO 21338:2010 standard, was used to characterise the ecotoxicity of diesel-powered cars. The method is sensitive enough to test the ecotoxic effect of the emission of individual vehicles. In general, significant positive correlation was found between ecotoxicity (expressed as Toxic Unit /TU/values) and total carbon (TC) as well as between TU and polycyclic aromatic hydrocarbon (PAH) concentrations.   相似文献
