Use of synchronous first-derivative fluorimetry for determination of gentamycin is described. Gentamycin reacts with acetylacetone and formaldehyde in pH 5.6 HOAc/NaOAc buffer solution to form N-gentamyl-2,6-dimethyl-3,5-diacethyl-1,4-dihydropyridine[I] which is a fluorescent substance. Spectra of [I] and the reagent blank can be separated with synchronous derivative fluorimetry, and gentamycin can be determined directly. The synchronous spectral peaks of [I] and the reagent blank are at 434 and 411 nm, respectively. The first-derivative peak of [I] is at 425 nm. Effects of pH, foreign ions, buffer system, and heating time on the determination of gentamycin have been examined. The linear regression equation of the calibration graph is C=0.0513H-0.0416, with a correlation coefficient of linear regression of 0.9978. C means total potency of gentamycin: U ml(-1); H means peak height in the linear regression equation calibration graph. The linear range for the determination of gentamycin is from 0.00 to 3.00 U ml(-1). Recovery is from 95.06 to 112.0%, R.S.D. of 3.8%. The results determined by the fluorimetric method agreed roughly with those by the microbiological method. The method is simple and has low detection limit.  相似文献

Proteomic analysis of simulated occupational jet fuel exposure in the lung   总被引:3,自引:0,他引:3  
We analyzed protein expression in the cytosolic fraction prepared from whole lung tissue in male Swiss-Webster mice exposed 1 h/day for seven days to aerosolized JP-8 jet fuel at concentrations of 1000 and 2500 mg/m3, simulating military occupational exposure. Lung cytosol samples were solubilized and separated via large scale, high resolution two-dimensional electrophoresis (2-DE) and gel patterns scanned, digitized and processed for statistical analysis. Significant quantitative and qualitative changes in tissue cytosol proteins resulted from jet fuel exposure. Several of the altered proteins were identified by peptide mass fingerprinting, confirmed by sequence tag analysis, and related to impaired protein synthetic machinery, toxic/metabolic stress and detoxification systems, ultrastructural damage, and functional responses to CO2 handling, acid-base homeostasis and fluid secretion. These results demonstrate a significant but comparatively moderate JP-8 effect on protein expression and corroborate previous morphological and biochemical evidence. Further molecular marker development and mechanistic inferences from these observations await proteomic analysis of whole tissue homogenates and other cell compartment, i.e., mitochondria, microsomes, and nuclei of lung and other targets.  相似文献

测定了亲水性甲基丙烯酸羟烷基酯如HEMA、MHPMA分别与AN或AAM,在60℃不同溶剂中自由基聚合反应的竞聚率。AN(M_1)-HEMA(M_2)共单体,在DMSO或DMF溶剂以AIBN或KPS-IPA引发剂条件下共聚,用Kelen-Tüds法计算的竞聚率变化不大,r_1=0.22-0.25、r_2=0.97-1.05,说明在此均相溶液共聚中,所用的溶剂及引发剂对竞聚率的影响较小,这两种单体能很好共聚。但AAM-MHPMA或AAM-HEMA共单体时,r_1与r_2值相差很大,如前者r_1=0.0433、r_2=3.98,后者r_1=0.0535、r_2=1.89,说明不易共聚,共聚物中主要是MHPMA或HEMA组分。  相似文献

采用动态热重分析法在升温速率为6.2K·min ̄(-1)及惰性气流量100cm ̄3·min ̄(-1)的条件下,对大庆渣油族组分的热转化行为进行了考察,获得了族组分的热转化速率峰值、速率峰值处的反应温度、转化率和剧裂分解温度区间等动力学基本数据以及各族组分的生焦性能。数据分析表明,族组分的热反应活性顺序为:饱和分>胶质>芳香分>沥青质。在大庆渣油族组分热转化的两个温区内,采用分段一级动力学模型较好地拟合了实验数据,确定了各族组分在两个温区内的热反应动力学参数。  相似文献

有关Co(Ⅱ)、Ni(Ⅱ)萃取分离的研究一直受到人们的重视。Preston报导了硫氰酸季铵盐(Q~ SCN-)萃取Co(Ⅱ)、Ni(Ⅱ)的平衡研究,认为Co(Ⅱ)的萃合物组成为Q_2Co(SCN)_4.本文用上升液滴法技术考察了Co(Ⅱ)/0.10mol·L~(-1)SO_4~(2-)/QSCN-煤油体系在298±1K条件下萃取速率R与CoSO_4、SO_4~(2-)和QSCN浓度的关系,确定了正向初始萃取速率方程,拟定了速控步骤。  相似文献

Six new ent‐abietane diterpenoids, rubescensins I–M ( 1 – 5 ) and P ( 6 ), along with two related acetonide derivatives ( 7 and 8 ), were isolated from Isodon rubescens. Their structures were elucidated by detailed spectroscopic analysis. Compound 5 is the first N‐containing diterpenoid from the genus Isodon, exhibiting notable cytotoxicity against human tumor K562 cells.  相似文献

用TRP技术研究了以全硅MCM-41(Si-MCM-41)和HNO3交换的全硅MCM-41(H-MCM-41)为载体制备的Ni-Mo、Co-Mo和Ni-W加氢脱硫(HDS)催化剂的还原性能,并以0.8(wt)%二苯并噻吩(DBT)的十氢萘溶液为模型化合物,在高压固定床反应器上考察了上述催化剂的加氢脱硫(HDS)反应性能。结果表明,Si-MCM-41经稀HNO3交换后,所担载的Ni-Mo和Ni-W催化剂还原性能、HDS活性和加氢活性有显著变化,但对Co-Mo催化剂影响不大。这说明在Ni-Mo/H-MCM-41和Ni-W/H-MCM-41中可能存在氢溢流现象,DBT的HDS活性与载体表面酸性和氢溢流有关。  相似文献

A novel methodology based on electron-nuclear double resonance (ENDOR) spectroscopy is used for the direct determination of the water coordination number (q) of gadolinium-based magnetic resonance imaging (MRI) contrast agents. Proton ENDOR spectra can be obtained at approximately physiological concentrations for metal complexes in frozen aqueous solutions either in the presence or absence of protein targets. It is shown that, depending on the structure of the co-ligand, the water hydration number of a complex in aqueous solution can be significantly different to when the complex is noncovalently bound to a protein. From the ENDOR spectra of the exchangeable protons, precise information on the metal-proton distance can be derived as well. These essential parameters directly correlate with the efficacy of MRI contrast agents and should therefore aid the development of novel, highly efficient compounds targeted to various proteins.  相似文献

A new kind of multilayer of didodecyldimethylammonium bromide (DDAB) and 1:12 phosphomolybdic anions (PMo12) was achieved on the surface of a wax-impregnated graphite (WIG) electrode by ion exchange and electrostatic interaction. The characterization and electrochemical behavior of the multilayer films of DDAB/PMo12 is described in detail. The chemically modified electrode was shown to exhibit an excellent electrocatalytic activity toward the reduction of BrO3 anion in 0.5 M H2SO4 and possesses several attractive features, such as simple preparation, fast response, good stability, etc. Electronic Publication  相似文献
