Stereoselective syntheses of novel 5,6-difunctionalized-2-azabicyclo[2.1.1]hexanes containing 5-anti-fluoro or hydroxyl in one methano bridge and a variety of syn- or anti-chloro, fluoro, hydroxy, methyl, or phenyl substituents in the other methano bridge have been effected. Rearrangements of iodides to alcohols were initiated using Selectfluor. Rearrangement of alcohols to fluorides was initiated using Deoxo-Fluor. Ring opening of 2-azabicyclo[2.2.0]hex-5-ene exo-epoxide with organocopper reagents is regioselective at C(5).  相似文献

Understanding the conformation, orientation, and specific activity of proteins bound to surfaces is crucial for the development and optimization of highly specific and sensitive biosensors. In this study, the very efficient enzyme beta-lactamase is used as a model protein. The wild-type form was genetically engineered by site-directed mutagenesis to introduce single cysteine residues on the surface of the enzyme. The cysteine thiol group is subsequently biotinylated with a dithiothreitol (DTT)-cleavable biotinylation reagent. beta-Lactamase is then immobilized site-specifically via the biotin group on neutral avidin-covered surfaces with the aim to control the orientation of the enzyme molecule at the surface and study its effect on enzymatic activity using Nitrocefin as the substrate. The DTT-cleavable spacer allows the release of the specifically bound enzyme from the surface. Immobilization of the enzyme is performed on a monolayer of the polycationic, biotinylated polymer PLL-g-PEG/PEG-biotin assembled on niobium oxide (Nb2O5) surfaces via neutral avidin as the docking site. Two different assembly protocols, the sequential adsorption of avidin and biotinylated beta-lactamase and the immobilization of preformed complexes of beta-lactamase and avidin, are compared in terms of immobilization efficiency. In situ optical waveguide lightmode spectroscopy and colorimetric analysis of enzymatic activity were used to distinguish between specific and unspecific enzyme adsorption, to sense quantitatively the amount of immobilized enzyme, and to determine Michaelis-Menten kinetics. All tested enzyme variants turned out to be active upon immobilization at the polymeric surface. However, the efficiency of immobilized enzymes relative to the soluble enzymes was reduced about sevenfold, mainly because of impaired substrate (Nitrocefin) diffusion or restricted accessibility of the active site. No significant effect of different enzyme orientations could be detected, probably because the enzymes were attached to the surface through long, flexible PEG chain linkers.  相似文献

本文提出了人体内苯、甲苯和二甲苯的代谢产物苯酚、马尿酸和甲基马尿酸的反相高效液相色谱分析法,讨论了其保留机制和样品预处理技术.以ODS为固定相,甲醇-水-醋酸为流动相时可实现良好分离.方法回收率为97.3%,相对标准偏差为1.02%.提出的方法可用于尿样分析,适用于临床与职业病防治的监测分析.  相似文献

New sufficient conditions are derived to guarantee the global synchronization of the weighted cellular neural network with multiple time delays. Based on Lyapunov theory, this paper proposes an adaptive feedback controlling method to identify the exact topology of a rather general weighted cellular neural network system with time-varying delays and then considers the synchronization of the neural network with different nodes dynamics. The parameters in this paper are very few and experiments show that the methods presented in this paper are of high application in global synchronization.  相似文献

This paper deals with the inverse spectral problem for a non-self-adjoint Sturm–Liouville operator with discontinuous conditions inside the interval. We obtain that if the potential q is known a priori on a subinterval $$ \left[ b,\pi \right] $$ with $$b\in \left( d,\pi \right] $$ or $$b=d$$, then $$h,\,\beta ,\,\gamma $$ and q on $$\left[ 0,\pi \right] $$ can be uniquely determined by partial spectral data consisting of a sequence of eigenvalues and a subsequence of the corresponding generalized normalizing constants or a subsequence of the pairs of eigenvalues and the corresponding generalized ratios. For the case $$b\in \left( 0,d\right) $$, a similar statement holds if $$\beta ,\,\gamma $$ are also known a priori. Moreover, if q satisfies a local smoothness condition, we provide an alternative approach instead of using the high-energy asymptotic expansion of the Weyl m-function to solve the problem of missing eigenvalues and norming constants.  相似文献

A highly selective and efficient LC–MS/MS method was developed to determine the plasma concentration of magnolol, hesperidin, neohesperidin and geniposide following oral administration of Zhi‐Zi‐Hou‐Po decoction in normal and depressed rats. Plasma samples were pretreated by protein precipitation with methanol. Chromatographic separation was performed on an XTerra® MS C18 column using a gradient elution with a mobile phase composed of acetonitrile–0.1% aqueous formic acid. The proposed method was validated to be specific, accurate and precise for the analytes determination in plasma samples. The calibration curves displayed good linearity over definite concentration ranges for the analytes. The intra‐ and inter‐day precision of the proposed method at three different levels were all within <11.13% and the relative errors ranged from ?8.46 to 8.93%. The recovery of the four compounds ranged from 82.72 to 89.08% and no apparent matrix effect was observed during sample analysis. After full validation, the established method was successfully applied for comparing the pharmacokinetics of four components between normal and depressed rats. The results showed that the AUC and Cmax of four analytes in depressed rats were significantly different from those in normal rats and might provide helpful information to guide the clinical use of Zhi‐Zi‐Hou‐Po to treat depression.  相似文献

Herein we report a new ammoniation‐based chemical modification strategy for synthesis of continuous and uniform metal–organic framework (MOF)/polyvinylidene fluoride (PVDF) membranes with attractive performance. Ammoniation can promote the support PVDF membrane to produce amino groups, form a nanoparticle structure, and be well cross‐linked; therefore, the high‐density heterogeneous nucleation sites for MOFs growth were provided and the thermal stability and chemical resistance of composite membranes can be greatly improved. The high‐quality layers of representative Cu‐BTC and ZIF‐8 were synthesized on the chemically modified PVDF membranes. By ammoniation, ZIF‐7 can even be grown under harsh synthetic conditions such as in DMF precursor solutions at 403 K. The fabricated MOF/PVDF composite membranes with excellent hollow fiber structures and enhanced structural stability exhibited high H2 permselectivities for H2/CO2 and H2/N2.  相似文献

A long-chain surfactant, enzoylbenzyl-N,N-dimethyl-N-octadecylammonium bromide (BDOB) with a benzophenone group, was synthesized to modify the montmorillonites (MMT) for the preparation of nanocomposites via photo-induced polymerization. The BDOB-modified MMT was characterized by the fourier transform infrared spectrometer (FTIR), thermal gravimetric analyzer (TGA) and X-ray diffraction (XRD), and the results of XRD indicated that the intercalated structures of BDOB-modified MMT was obtained. The conversion of the bisphenol A epoxy diacrylate (EA) was quantified by the FTIR, and the results indicated that conversion increased with an increase in the amount of BDOB-modified MMT. The morphologies of the UV-cured EA/MMT nanocomposites prepared from this organically modified MMT were studied by means of XRD and TEM, and the results showed that all the samples contained an intercalated structure with partial exfoliated structure. The results of TGA and mechanical properties also indicated that the thermal and mechanical properties of UV-cured nanocomposites were significantly enhanced due to the presence of the long chain surfactant organically modified MMT.  相似文献
