Microarray-guided discovery of two-photon (2P) small molecule probes for live-cell imaging of cysteinyl cathepsin activities

A microarray immobilized with 105 aldehyde-containing small molecules was screened against mammalian cell lysates over-expressing cathepsin L to identify two potent inhibitors, which were subsequently converted into cell-permeable probes capable of live-cell imaging of endogenous cysteinyl cathepsin activities by two-photon fluorescence microscopy.

Chemical examination of the fresh rhizomes of Tupistra wattii HOOK. f. led to the isolation of three new steroidal saponins, wattoside G (1), H (2), and I (3), together with one known steroidal saponin, (25S)-1beta,3beta,4beta-trihydroxyspirotan-5beta-yl-O-beta-D-glucopyranoside (4). The structures of 1-3 were established to be (25R)-1beta,2beta,3beta,5beta-tetrahydroxyspirostan-4beta-yl-O-beta-D-xylopyranoside (1), (24S,25S)-24-[(beta-D-glucopyranosyl)oxy]-1beta,2beta,3beta,4beta,5beta,7beta-hexahydroxyspirostan-6-one (2), and (24S,25S)-1beta,3beta-dihydroxy-5beta-spirostan-24-yl-O-beta-D-glucopyranosyl-(1-->6)-beta-D-glucopyranoside (3) on the basis of detailed analyses of physical, chemical, and spectral data. The isolated compounds were evaluated for cytotoxic activity against the cancer cell line K562 in vitro.

The adsorption of decylmethylsulfoxide (DEMS) onto carbon black and the effects of temperature, salt (NaCl) and acid (HCl) have been measured.Typical two plateaux type adsorption isotherms were obtained. Applying the two step model of surfactant adsorption on solid/liquld interface and the general adsorption isotherm equation[9]the experimental results were interpreted qualitatively and quantitatively.

Synthesis, crystal structure of bis(arylamido)lanthanide methyl complexes and their catalytic behavior for the polymerization of methyl methacrylate

Reactions of [N(C₆H₃i-Pr₂-2,6)(SiMe₃)]₂LnCl(THF) (Ln=Nd, Yb) with two equivalents of MeLi in a mixture solution of toluene and Et₂O gave [N(C₆H₃i-Pr₂-2,6)(SiMe₃)]₂LnCH₃(μ-CH₃)Li(THF)₃·PhCH₃ (Ln=Nd (1), Yb (2)) in good isolated yields as crystalline solids. The single-crystal structural analysis of 2 revealed that the coordination geometry of ytterbium ion is best described as a distorted pseudo-tetrahedron. Both 1 and 2 are active for the polymerization of methyl methacrylate to give syndiotactic-rich and high molecular weight polymers (M_n>10⁴) with relatively narrow molecular weight distributions (M_w/M_n<2).

Solid-phase linked o-iodoanilines (4) and o-iodophenol (9) reacted with 1,3- and 1,4-dienes in the presence of palladium acetate to generate highly substituted indolines, tetrahydroquinolines, hydrobenzofurans and hydrobenzopyrans, which provided an efficient way for making heterocyclic molecule libraries.
