A sensitive bioassay for estimating the activity of Trichosanthin was developed by using intraoocyte microinjection as the technique and germinal vescicle breakdown as a marker. The GVBD inhibitory activity assay was million-fold more sensitive than the routinely used abortifacient activity assay. The detectable amount of Trichosanthin ranged from 0.5 to 100 pg. Since the least detectable dose was 0.5 pg, it made the assay suitable to analyse the microquantity of pure Trichosanthin used for studying the structure and function of the drug.  相似文献

A home-made high sensitive photoacoustic transducer was used to determine the photoacoustic signal in a solution. The photoacoustic spectra of Ho in different solvents were measured and comparedwith its absorption spectrum. The affection factors of photoacousticsensitivity are discussed. The trivalent rare earth ions Ho3+ and Nd3+in an aqueous acetonitrile solution were determined. The detection limit is 5×10-8 mol/L for Ho3+ and 1.0 ×10-7 mol/L for Nd3+, and corresponds to the absorbance of 1.5×10 and 6.3×10-7.respectively.  相似文献

Angiotensin I‐converting enzyme (ACE) inhibitory peptide was isolated from the hen ovotransferrin hydrolysate using chymotryptic hydrolysis by two steps of reverse‐phase high‐performance liquid chromatography. The amino sequence of this novel peptide was identified as Lys‐Val‐Arg‐Glu‐Gly‐Thr‐Thr‐Tyr that inhibited ACE activity in vitro in a concentration‐dependent manner with an effective concentration (IC50) of 102.8 μM. Also, this inhibition was identified as noncompetitive using the Lineweaver‐Burk plot. Moreover, the antihypertensive action of this novel peptide was investigated by an intravenous injection into spontaneously hypertensive rats (SHR). A dose‐dependent reduction of systolic blood pressure by this peptide was observed after 40 min of treatment and it decreased the blood pressure markedly at the maximal dose (1 nmol/mL/kg). The maximal blood pressure lowering activity of this peptide was calculated as 163% of captopril (10 pmol/mL/kg) that was used as positive control. In conclusion, the obtained data suggests that Lys‐Val‐Arg‐Glu‐Gly‐Thr‐Thr‐Tyr has an ability to inhibit ACE activity and decrease the systolic blood pressure in hypertensive animals.  相似文献

[Eu(ABA)(phen)2(H2O)3](ClO4)3·3phen·4.5H2O (1) and [Eu(Val)(phen)2(H2O)3](ClO4)3·3phen·2H2O (2) are two new europium complexes with amino acids and 1,10-phenanthroline (phen=1,10-phenanthroline, ABA=-amino butyl acid, Val= -valine). Their crystal structures were measured by X-ray crystallography. Europium atoms in both complexes are nine-coordinated with bidentate 1,10-phenanthroline and carboxylate anion of amino acids, and water molecules. In the solid state, 1 and 2 have a structure involving aromatic stacking of the coordinated and non-coordinated 1,10-phenanthroline and the oxygen atoms of non-coordinated perchlorate anions being H-bond acceptors connect [Eu(ABA)(phen)2(H2O)3]3+·3phen·4.5H2O or [Eu(Val)(phen)2(H2O)3]3+·3phen·2H2O in their structures. In their interactions, several C–HO bonds play an important role. Owing to their different amino acid ligands and the number of lattice water molecules, there is some difference in their hydrogen bond patterns in 1 and 2. The side chain of -valine is involved in the formation of C–HO bonds. Hydrogen bond and π–π interactions determine the supramolecular formation of three-dimensional net works of both complexes.  相似文献

The chiral phosphine‐oxazoline ligands 3 and 4 bearing 4‐alkoxymethyl substituents on the oxazoline ring with (R)‐configuration were prepared from L‐serine methyl ester in 66% and 33% yields, respectively. Along this synthetic pathway, the β‐hydroxylamides derived from L‐serine methyl ester and 2‐halobenzoyl chlorides were expediently converted to the corresponding oxazolines by using diethylaminosulfur trifluoride as the activation agent. Potassium diphenylphosphide was the reagent of choice for replacing the bromine atom on the phenyl ring, giving the desired oxazoline‐phosphine ligands 3 and 4 . Together with [Pd(η3‐allyl)Cl]2, ligands 3 and 4 induced an enantioselective allylic substitution reaction of 1,3‐diphenyl‐2‐pro‐penyl acetate by dimethyl malonate. Although ligands 3 and 4 exhibit the (R)‐configuration, differing from the (S)‐configuration of Pfaltz‐Helmchen‐Williams phosphine‐oxazoline ligands, all these ligands led to the same enantiotopic preference in the allylic substitution reaction. To facilitate the recovery and reuse of the phosphine‐oxazoline ligand, immobilization on Merrifield resin was attempted, albeit in low loading.  相似文献

在pH=3.8的磷酸柠檬酸盐缓冲体系中,铟(Ⅲ)-向红菲(口罗)啉络合物在单扫描示波极谱仪上于-0.67 V(vs.SCE)处产生一灵敏的吸附还原波,其二阶导数波灵敏度高,波形好.峰电位Ⅰ~O_P与钢(Ⅲ)浓度在 2.0X10~(-8)~2.0X10~(-6)mol/L范围内呈良好的线性关系,其检测限为8X10~(-9)mol/L铟(Ⅲ).该法用于试剂氧化锌和镀膜玻璃中铟(Ⅲ)的恻定,结果较好.  相似文献
