TNF-related apoptosis-inducing ligand (TRAIL) has been proposed as a promising cancer therapy that preferentially induces apoptosis in cancer cells, but not most normal tissues. However, many cancers are resistant to TRAIL by mechanisms that are poorly understood. In this study, we showed that tunicamycin, a naturally occurring antibiotic, was a potent enhancer of TRAIL-induced apoptosis through downregulation of survivin. The tunicamycin-mediated sensitization to TRAIL was efficiently reduced by forced expression of survivin, suggesting that the sensitization was mediated at least in part through inhibition of survivin expression. Tunicamycin also repressed expression of cyclin D1, a cell cycle regulator commonly overexpressed in thyroid carcinoma. Furthermore, silencing cyclin D1 by RNA interference reduced survivin expression and sensitized thyroid cancer cells to TRAIL; in contrast, forced expression of cyclin D1 attenuated tunicamycin-potentiated TRAIL-induced apoptosis via over-riding downregulation of survivin. Collectively, our results demonstrated that tunicamycin promoted TRAIL-induced apoptosis, at least in part, by inhibiting the expression of cyclin D1 and subsequent survivin. Of note, tunicamycin did not sensitize the differentiated thyroid epithelial cells to TRAIL-induced apoptosis. Thus, combined treatment with tunicamycin and TRAIL may offer an attractive strategy for safely treating resistant thyroid cancers.  相似文献

To analyze the effect of redox state changes on the second-order nonlinear optical (NLO) responses of organoimido-functionalized Keggin-type heteropolyanions, the excitation properties and static second-order polarizabilities of fully oxidized state, the first and second reduced states were calculated by means of the time-dependent density functional theory (TDDFT) method combined with the sum-over-states (SOS) formalism. The incorporation of extra electrons causes significant enhancement in the second-order NLO activity. The reduced complexes show more than three times the efficiency of fully oxidized state. Moreover, the NLO activities for PW11ReVNPh system can also be modified by controlling the spin multiplicity. The high spin state (3 3) has twice larger β vec value than the low spin state (1 3). The characteristic of the charge-transfer transition corresponding to the dominant contributions to the β vec values indicates that metal-centered redox processes influence the intramolecular donor or acceptor character, which accordingly leads to the variations in the computed β values. Owing to the reversible and manipulable redox processes, these kinds of the POM-based hybrid complexes could comprise a promising family of three-state redox-switchable molecular device combining chromic, magnetic, and NLO output.  相似文献

A series of functional polyacetylenes (PAs) bearing diphenyl oxadiazole pendant groups ( P1 – P4 ) were prepared, and the resultant polymers are completely soluble in common organic solvents. Their structures and properties were characterized and evaluated by DSC, TGA, UV, PL, CV, and EL analyses. The results show that all the resulting polymers possesses low LUMO energy level and high thermal stability, and the resultant functional polyacetylenes without spacer group between the polyacetylene conjugated main chain and oxadiazole pendant groups ( P1 – P3 ) show lower LUMO energy level (~?3.87 eV) and higher thermal properties (Tg) than that ( P4 ) with a flexible spacer. The resultant polymer ( P2 ) was applied as an ETM in bilayer electroluminescent devices and effectively enhances external quantum efficiency and the brightness of device, and decreases turn‐on voltages of devices. © 2010 Wiley Periodicals, Inc. We have synthesized a narrow‐bandgap conjugated polymer ( PCTDPP ) containing alternating cyclopentadithiophene (CT) and diketo‐pyrrolo‐pyrrole (DPP) units by Suzuki coupling. This PCTDPP exhibits a low band gap of 1.31 eV and a broad absorption band from 350 to 1000 nm, which allows it to absorb more available photons from sunlight. A bulk heterojunction polymer solar cell incorporating PCTDPP and C70 at a blend ratio of 1:3 exhibited a high short‐circuit current of 10.87 mA/cm2 and a power conversion efficiency of 2.27%. © 2010 Wiley Periodicals, Inc. A series of lead‐sensitive poly(N‐isopropylacrylamide) microgels with pendant crown ether groups were prepared. Their cation‐sensitive behaviors were studied by dynamic light scattering. When ionic strength is not controlled, adding salts causes the microgel particles to deswell. However, when the salt effect is ruled out by keeping a constant ionic strength, adding Pb2+ results in much larger swelling. The Pb2+‐induced swelling was explained by the formation of host–guest complex between Pb2+ and the pendant crown ether groups, which increases the hydrophilicity of the polymer and accordingly the degree of swelling. The lead sensitivity of the microgels increases with increasing crown ether content. For the modified microgel with the highest crown ether content, it swells to ~430% of its original volume at [Pb2+] = 10 mM. Other cations also increase the swelling degree of the modified microgels. The extent of the cation‐induced swelling mainly depends on their affinity to the pendant crown ether groups. © 2010 Wiley Periodicals, Inc. A new octameric water cluster was observed in the complex Co2(dptc)(bipy)2(H2O)6 · 4H2O ( 1 ) (H4dptc = diphenyl‐3,3′,4,4′‐tetracarboxylic acid; bipy = 2,2′‐bipyridine), which was characterized by single‐crystal X‐ray diffraction, elemental analysis and IR spectroscopy. The centrosymmetric octamer consists of a water hexamer in the chair form and two water molecules and brings to light a novel mode of the cooperative association of water molecules. Those complex units are connected into a 2D infinite layer framework through hydrogen bonding. Consequently, the 2D layers are further aggregated by hydrogen bonding with octameric subunits and π ··· π stacking interactions to form a 3D supramolecular architecture.  相似文献

In this work, 0.08 mmol L−1 of phthalic acid was introduced as a mobile phase additive to quantify free amino acids (AAs) by hydrophilic interaction liquid chromatography (HILIC) coupled with electrospray ionization tandem mass spectrometry (ESI-MS/MS). The addition of phthalic acid significantly increased the signal intensity of protonated AA ions, resulting from the decrease of the relative abundance of AA sodium adducts. Meanwhile, the chromatographic peak shapes of AAs were optimized. As a consequence, there was a noticeable increase in the sensitivity of detection for AAs. The limits of detection (LOD) and quantification (LOQ) of the AAs ranged from 0.0500 to 20.0 ng mL−1 and from 0.100 to 50.0 ng mL−1, respectively, which were 4–50 times lower compared to the values measured without the addition of phthalic acid. The enhanced detection and separation of AAs were obtained by merely adding phthalic acid to the mobile phase without changing other conditions. Eventually, this simple method was validated and successfully applied to the analysis of twenty-four kinds of free AAs in human thyroid carcinoma and para-carcinoma tissues, demonstrating a significant increase of most AAs in thyroid carcinoma tissues (p < 0.05).  相似文献

Structural stability of the template is one of the most important considerations during the preparation of protein imprinting technology. To address this limitation, we propose a novel and versatile strategy of utilizing macromolecularly functional monomers to imprint biomacromolecules. Results from circular dichroism and synchronous fluorescence experiments reflect the macromolecularly functional monomers tendency to interact with the protein surface instead of permeating it and destroying the hydrogen bonds that maintain the protein’s structural stability, therefore stabilizing the template protein structure during the preparation of imprinted polymers. The imprinted polymers composed of macromolecularly functional monomers or their equivalent micromolecularly functional monomers over silica nanoparticles were characterized and carried out in batch rebinding test and competitive adsorption experiments. In batch rebinding test, the imprinted particles prepared with macromolecularly functional monomers exhibited an imprinting factor of 5.8 compared to those prepared by micromolecularly functional monomers with the imprinting factor of 3.4. The selective and competitive adsorption experiments also demonstrated the imprinted particles made by macromolecularly functional monomers possessed much better selectivity and specific recognition ability for template protein. Therefore, using macromolecularly functional monomers to imprint may overcome the mutability of biomacromolecule typically observed during the preparation of imprinted polymers, and thus promote the further development of imprinting technology.  相似文献

An aligned and laminated sulfur‐absorbed mesoporous carbon/carbon nanotube (CNT) hybrid cathode has been developed for lithium–sulfur batteries with high performance. The mesoporous carbon acts as sulfur host and suppresses the diffusion of polysulfide, while the CNT network anchors the sulfur‐absorbed mesoporous carbon particles, providing pathways for rapid electron transport, alleviating polysulfide migration and enabling a high flexibility. The resulting lithium–sulfur battery delivers a high capacity of 1226 mAh g−1 and achieves a capacity retention of 75 % after 100 cycles at 0.1 C. Moreover, a high capacity of nearly 900 mAh g−1 is obtained for 20 mg cm−2, which is the highest sulfur load to the best of our knowledge. More importantly, the aligned and laminated hybrid cathode endows the battery with high flexibility and its electrochemical performances are well maintained under bending and after being folded for 500 times.  相似文献
