Continuous 1-dimensional wavelet transform (WT) was applied to the quantitative analysis of a vitamin combination of thiamine hydrochloride (THI) and pyridoxine hydrochloride (PYR) with strongly overlapping signals. Absorbance data from the UV-Vis absorption spectrum of width 1150 were subjected to Gauss1 and Gauss2 WTs. Because of its flexibility, data processing, and its high signal amplitude, the continuous WT method is a powerful tool for analysis of multicomponent mixtures. By measuring the amplitude signals corresponding to the selected zero-crossing points of the transformed signal, we obtained the calibration curve. The validation of the calibration graphs was confirmed with different mixtures of THI and PYR at various concentration ratios. A brief explanation of the continuous wavelet method is given. MATLAB 6.5 software was used to perform the calculations. The results of our study were compared with those obtained by spectroscopic, chemometric, and liquid chromatographic methods, and good agreement was found. 相似文献
The study and extension of a simple automated clean-up method for polychlorinated dibenzo-p-dioxins (PCDD) and polychlorinated dibenzofurans (PCDF) to a broad range of polychlorinated biphenyls (PCB) is described. The isolation of seven PCDD, ten PCDF, and three coplanar PCB (cPCB) is extended to eight monoortho substituted PCB and seven so-called "marker PCB" (Aroclor 1260) for fatty food samples. This enables quantification of 35 compounds - including all congeners with a WHO toxic equivalent factor (TEF)--in a single extraction and single purification step. The chromatographic behaviour of mono-ortho PCB and marker PCB on a variety of adsorbents, including basic alumina, has been studied. Partitioning of analytes through multi-column sequences is described and correlated with their structural and electronic properties, by use of molecular modelling calculations. The fractionation process available with the Power-Prep automated clean-up system enables rapid independent analysis of the different groups of compounds. Gas chromatography with high resolution mass spectrometry (GC-HRMS) is used for the PCDD/F and cPCB fraction and quadrupole ion-storage tandem in time mass spectrometry (GC-QISTMS) for analysis of the remaining PCB. A comparison study was performed on quality-control samples and real fatty food samples to evaluate the robustness of the new strategy compared with a reference method. On the basis of this simultaneous clean-up, a rapid simplified strategy for PCDD/F and selected PCB analysis determination is proposed for fatty food samples. 相似文献

The use of crown ethers is a simple and rapid method for the separation and determination of strontium in various samples. After sample dissolution oxalate precipitation was carried out to remove K. Chromatographic separation of strontium from most inactive and radioactive interference was done with a crown ether. The chemical recovery was determined gravimetrically. Recent developments in trapping efficiency inside ion trap mass spectrometer permitted to lower instrument detection limit (IDL). An IDL of 200 fg μl−1 injected with a signal-to-noise ratio of 5:1 for tetrachlorodibenzo-p-dioxin (TCDD) was obtained by gas chromatography coupled to a quadrupole ion storage mass spectrometer in tandem mode (GC/MS/MS). Coupling large volume programmable temperature vaporizer (PTV-LV) injection to GC/MS/MS provides an alternative and complementary method to classical splitless-GC injection connected to high-resolution mass spectrometry (splitless-GC/HRMS) method for dioxin monitoring in food and feed.
An injection volume of 10 μl was found to be the best compromise between the sensitivity requirements and the robustness required for a high throughput method. PTV-LV-GC/MS/MS and Splitless-GC/HRMS were compared by performing analysis on five different matrices such as beef fat, yolk eggs, milk powder, animal feed and serum samples covering a concentration range of two orders of magnitude (i.e. 0.2–25 ng WHO-TEQ kg−1). An analysis of variance (ANOVA) was carried out. Fisher tests pointed out that the method effect for all the 2,3,7,8 congeners was not significant, indicating that the null hypothesis (H0: μ1=μ2=…=μn) was not rejected. Moreover, the interaction effects between methods and matrices were not significant for most of the 2,3,7,8 congeners. However, three congeners (2,3,7,8-TCDF; 1,2,3,4,7,8-HxCDD and 1,2,3,4,6,7,8-HpCDD) were characterized by P-values lower than the significance level (=0.05). In toxic equivalence (TEQ), the study showed that no significant bias was observed between the two methods. Consequently, PTV-LV-GC/MS/MS is an attractive technique and can be used as a cost effective complementary method to HRMS for dioxin levels monitoring in food and feed. 相似文献
An accurate gas-phase acidity for germane (enthalpy scale, equivalent to the proton affinity of GeH3?), ΔHacido(GeH4) = 1502.0 ± 5.1 kJ mol?1, is obtained by constructing a consistent acidity ladder between GeH4, and H2S by using Fourier transform-ion cyclotron resonance spectrometry, and 0 and 298.15 K values for the first bond dissociation energy of GeH4 are proposed: D0o(H3Ge-H) = 352 ± 9 kJ mol?1; Do(H3Ge-H) = 358 ± 9 kJ mol?1, respectively. These results are compared with experimental and theoretical data reported in the literature. Methylgermane was found to be a weaker acid than germane by approximately 35 kJ mol?1: ΔHacido = 1536.6 kJ mol?1. 相似文献
A new chelating sorbent has been developed using Amberlite XAD-2 resin anchored with pyrocatechol through –N=C– group. This sorbent, characterised by elemental analysis and infrared (IR) spectra, was used as packing for the minicolumn in an on-line system preconcentration system for cadmium, cobalt, copper and nickel determination. Metal ions were sorbed in the minicolumn, from which it could be eluted directly to the nebulizer–burner system of the flame atomic absorption spectrometer (FAAS). Elution of all metals from minicolumn can be made with 0.50 mol L−1 HCl or HNO3. The enrichment factors obtained were 16 (Cd), 24 (Co), 15 (Cu) and 19 (Ni), for 60 s preconcentration time, and 39 (Cd), 69 (Co), 36 (Cu) and 41 (Ni), if used 180 s preconcentration time. Under the optimum conditions, the proposed procedure allowed the determination of cadmium, cobalt, copper and nickel with detection limits of 0.31, 0.32, 0.39 and 1.64 μg L−1, respectively, when used preconcentration periods of 180 s. The accuracy of the developed procedure was sufficient and evaluated by the analysis of the certified reference materials NIST 1515 apple leaves and NIST 1570a spinach leaves. Formulations of the tin etiopurpurin (SnET2) have been observed to undergo a spectral change on storage in dimethylformamide solution. This results in an alteration in the action spectrum, with enhanced photodynamic activity at lower wavelengths and decreased activity at 660 nm. On the basis of structural considerations, a reduced analog of SnET2 was prepared with an absorbance maximum at 640 nm. Formation of this product, termed SnET2H(2) relieves steric strain inherent in the parent molecule. Nuclear magnetic resonance, spectral and photodynamic data are consistent with the formation of SnET2H(2) during SnET2 storage. Slight modifications in the original synthesis are also reported, resulting in improved yields of intermediate products. 相似文献
The reactions of meta—para-substituted aryl isocyanates with phosphorus-containing 1,3-zwitterions, which proceed with the CN migration of the CO2Et group to form the corresponding carbamates, were extended to ortho-substituted aryl isocyanates. Over the past few years, a large number of studies have been prepared that describe the analysis of peptides and proteins using capillary electrophoresis (CE) and laser-induced fluorescence (LIF). These studies have focused on two general goals: (i) development of automatic, selective and quick separation and detection of mixtures of peptides or proteins; (ii) generation of new methods of quantitation for very low concentrations (nm and subnanomolar) of peptides. These two goals are attained with the use of covalent labelling reactions using a variety of dyes that can be readily excited by the radiation from a commonly available laser or via the use of noncovalent labelling (immunoassay using a labelled antibody or antigen or noncovalent dye interactions).
