Screening method for organic aciduria by spectrofluorometric measurement of total dicarboxylic acids in human urine based on intramolecular excimer-forming fluorescence derivatization

A simple screening method of organic aciduria by spectrofluorometric measurement of total dicarboxylic acids in human urine is described. This method is based on an intramolecular excimer-forming fluorescence derivatization with a pyrene reagent, 4-(1-pyrene)butanoic acid hydrazide (PBH). Dicarboxylic acids in urine were converted to the corresponding dipyrene-labeled derivatives by reaction with PBH in the presence of 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide and pyridine, and the derivatives afforded intramolecular excimer fluorescence (420-540 nm) which can clearly be discriminated from the normal fluorescence (360-420 nm) emitted from reagent blanks. The technique is so selective that it permits spectrofluorometric measurement of total amount of dicarboxylic acids by the direct derivatization of diluted urine samples. The same reaction mixture has also served as a liquid chromatographic (LC) sample for the separative determination of individual dicarboxylic acids. The spectrofluorometric data did not contradict with the LC data. These methods were usefully applied to preliminary screening test of glutaric aciduria. In conclusion, the present derivatization method allows rapid and direct determination of total amount of dicarboxylic acids in human urine samples.     

Syntheses of TiO2(B) nanowires and TiO2 anatase nanowires by hydrothermal and post-heat treatments

TiO₂(B) nanowires and TiO₂ anatase nanowires were synthesized by the hydrothermal processing in 10 M NaOH aq. at 150 °C followed by the post-heat treatment at 300-800 °C. As-synthesized Na-free titanate nanowires (prepared by the hydrothermal treatment and repeated ion exchanging by HCl (aq.) were transformed into TiO₂(B) structure with maintaining 1-D morphology at 300-500 °C, and further transformed into anatase structure at 600-800 °C with keeping 1-D shape. At 900 °C, they transformed into rod-shaped rutile grains. Microstructure of these 1-D TiO₂ nanomaterials is reported.     

Preparation of18F source for slow positron beam by proton bombardment of18O-water

Fluorine-18 produced by the¹⁸O(p,n)¹⁸F reaction on¹⁸O-water has proved to be highly useful as a source for a slow positron beam. About 70 GBq of¹⁸F is produced routinely by an ultra-compact cyclotron. The¹⁸F formed in H₂ ¹⁸O target is sent through a fine pipe to the site of positron slowing-down, fixed on a small spot by adsorption or drying, and then placed close to the moderator foil. An automatic apparatus has been set up for the entire process including the recovery of H₂ ¹⁸O.     

Synthesis and pharmacological evaluation of pyrroloazepine derivatives as potent antihypertensive agents with antiplatelet aggregation activity

A series of 1-aminoalkyl-pyrrolo[2,3-c]azepin-8-one derivatives was synthesized and evaluated as alpha 1 adrenergic and serotonin 2 (5-HT2) receptor antagonists, with the aim of finding a novel antihypertensive agent potently exhibiting both activities. Some compounds with a 4-[4-(4-fluorobenzoyl)piperidino]butyl group at the 1-position exhibited both activities, and varied significantly in terms of the substituents at the 4-position of the pyrroloazepine ring. Among the compounds obtained in this study, (E)-1-[4-[4-(4-fluorobenzoyl)piperidino]-butyl]-4-hydroxyimino-7- methyl-1,4,5,6,7,8-hexahydropyrrolo[2,3-c]azepin-8-one (15a, SUN9221) displayed potent alpha 1-adrenergic antagonistic activity (pA2 = 8.89 +/- 0.21) and 5-HT2 antagonistic activity (pA2 = 8.74 +/- 0.22) in isolated guinea pig arteries. This compound exhibited antihypertensive activity and a duration of action equivalent to orally administered prazosin or doxazosin, 3 mg/kg, in conscious spontaneously hypertensive rats, as well as potent antiplatelet aggregation activity.     

Iodoetherification of 4-penten-1,3-diols: stereoselective synthesis of cis 2-iodomethyl-3-hydroxytetrahydrofurans

cis 2-Iodomethyl-3-hydroxytetrahydrofurans have been obtained in high yields and in high selectivities by treatment of 4-penten-1,3-diols with iodine (ether-H₂O, NaHCO₃).     

Analysis of free fatty acids by a micro-liquid chromatograph directly coupled with a mass spectrometer through a vacuum nebulizing interface

A liquid chromatograph directly coupled with a quadrupole mass spectrometer through a vacuum nebulizing interface was applied to the analysis of various free fatty acids. Chemical ionization mass spectra of the C₇? C₂₂ free fatty acids were first examined using either methanol or benzene as the reagents. Then the practical compositional analysis of the fatty acids were performed with various biological samples such as bean oil, rape oil, palm oil and milk fat where most of the fatty acids are included as their triglycerides.     

Reversible and Stepwise Single-Crystal-to-Single-Crystal Transformation of a Platinum(II) Complex with Vapochromic Luminescence

The vapochromic single-crystal-to-single-crystal (SCSC) transformation of a highly luminescent Pt^II complex bearing an N-heterocyclic carbene [Pt(CN)₂(tBu-impy)] (tBu-impyH⁺=1-tert-butyl-3-(2-pyridyl)-1H-imidazolium) is reported. The trihydrate form of the complex, which exhibits blue ³MMLCT emission owing to weak Pt⋅⋅⋅Pt interactions, changed its luminescence color from blue to yellowish-green upon the desorption of water molecules while keeping the high emission quantum yield of more than 0.45. Variable-temperature and continuous in-situ tracking of single-crystal X-ray diffraction measurements revealed that the SCSC transformation proceeds reversibly by the release and reabsorption of water molecules, thereby changing the stacked structure slightly. As a result, the dynamics of vapor-induced SCSC transformation were elucidated: that the anhydrous form returned to the original trihydrate form in a two-step process under a water vapor atmosphere. In addition, the Pt^II complex exhibited a similar SCSC response accompanied by a luminescence color change in the presence of methanol vapor, while being inactive toward ethanol vapor.
