Liposomes can be effectively deposited on the inner surface of a capillary wall by flushing the electrophoretic system with a liposome suspension followed by air-drying of the capillary and removal of the excess of loosely bound liposomes by a 0.1 M NaOH wash. It was demonstrated that capillaries prepared in this way could be used for studies of analyte (drug)–liposome binding. The results were expressed as free binding energy changes [Δ(ΔG0)] relatively to an arbitrarily selected standard (acetylsalicylic acid). The results were compared to [Δ(ΔG0)] changes obtained from binding studies effected by capillary electrophoresis using a stable liposome plug in a capillary with minimized endoosmotic flow. Good agreement of data reported in the literature (without correction for the residual endoosmotic flow), our previous data obtained in a similar way (however, after the correction for the residual endoosmotic flow) and data obtained by the immobilized liposome affinity electrochromatography reported in this communication was achieved.  相似文献

We report density functional molecular dynamic simulations to determine the early chemical events of hot (T=3000 K) and dense (rho=1.97 g/cm(3), V/V(0)=0.68) nitromethane (CH(3)NO(2)). The first step in the decomposition process is an intermolecular proton abstraction mechanism that leads to the formation of CH(3)NO(2)H(+) and the aci ion H(2)CNO(2) (-). This event is also confirmed to occur in a fast annealing simulation to a final temperature of 4000 K at rho=2.20 g/cm(3). An intramolecular hydrogen transfer that transforms nitromethane into the aci acid form, CH(2)NO(2)H, accompanies this event. To our knowledge, this is the first confirmation of chemical reactivity with bond selectivity for an energetic material near the Chapman-Jouget state of the fully reacted material. We also report the decomposition mechanism followed up to the formation of H(2)O as the first stable product. We note that similarities in the global features of reactants, intermediates, and products of the reacting fluid seem to indicate a threshold for similar chemistry in the range of high densities and temperatures reported herein.  相似文献

The reaction of open-chain and cyclohexane sym-2-dimethylaminomethylene-1,3-diones with amidines and guanidine in refluxing ethanol gave, generally in good yields, a series of 5-acylpyrimidines and 7,8-dihydroquinazolin-5(6H)-ones, respectively. With formamidine (and in part acetamidine), 2-formylimino-1,3-diones were formed in general, as sole products or mixtures with the corresponding pyrimidines or dihydroquinazolinones.  相似文献

We present the results of Car-Parrinello (CP) simulations of water at ambient conditions and under pressure, using a rigid molecule approximation. Throughout our calculations, water molecules were maintained at a fixed intramolecular geometry corresponding to the average structure obtained in fully unconstrained simulations. This allows us to use larger time steps than those adopted in ordinary CP simulations of water, and thus to access longer time scales. In the absence of chemical reactions or dissociation effects, these calculations open the way to ab initio simulations of aqueous solutions that require time scales substantially longer than presently feasible (e.g., simulations of hydrophobic solvation). Our results show that structural properties and diffusion coefficients obtained with a rigid model are in better agreement with experiment than those determined with fully flexible simulations. Possible reasons responsible for this improved agreement are discussed.  相似文献

The O-exchange reaction between CO2+ and CO is triggered by two different entrance channels in 13C-labeled systems. The unimolecular dissociation of the C13CO3(+) reactive intermediate is investigated at varying energies by mass spectrometric techniques. An almost "one-way" O-exchange at energies close to the dissociation threshold discloses a remarkably large intramolecular 12C/13C kinetic isotope effect ranging from 6.0 to 7.3 +/- 0.5.  相似文献

The HS3 radical and the HS3(-) anion, the sulfur analogues of HO3 and HO3(-), have been detected for the first time by mass spectrometric experiments performed in the gas phase. The structural and energetic features of HS3 and HS3(-) have been investigated by ab initio calculations. Both HS3 and HS3(-) are characterized by HSSS open-chain structures, stable toward the dissociation into S2 and HS(0/-). HS3 adds to HS and HS2 as the known HS n species, and HS3(-) is the conjugate base of a strong Br?nsted acid, the trisulfane HSSSH.  相似文献

New monodentate H8‐binaphthol based phosphoramidites 6 b–i have been prepared. Starting from (S)‐3,3′‐dibromo‐5,5′,6,6′,7,7′,8,8′‐octahydro‐1,1′‐binaphthyl‐2,2′‐diol 3 , a general protocol for the synthesis of ligands 6 is presented. A small ligand library bearing aryl substituents in the 3,3′‐position of the binaphthol core was synthesized and successfully tested in the iridium‐catalyzed asymmetric hydrogenation of 2‐amidocinnamates to obtain different α‐amino acid derivatives in up to 99 % ee.  相似文献

The rhodium‐catalyzed asymmetric hydrogenation of different enamides, in particular, dihydro‐β‐carboline derivates, was investigated in the presence of chiral phosphorus ligands. Enantioselectivities of up to 99 % ee were obtained after ligand screening and optimization of the reaction conditions. The scope and limitation of the catalysts were shown in the synthesis of optically active tetrahydro‐β‐carbolines and other benchmark N‐acyl‐1‐aryl ethylamines.  相似文献

Ribes nigrum L. (blackcurrant) leaf extracts, due to high levels of flavonols and anthocyanins, have been shown to exhibit beneficial effects in inflammatory diseases. However, whereas their traditional use has been investigated and validated in several models of inflammation and oxidative stress, the possible impact on skin disorders is still largely unknown. The purpose of this work was to elucidate the effects of R. nigrum leaf extract (RNLE) on keratinocyte-derived inflammatory mediators, elicited by a Th1 or Th2 cytokine milieu. HaCaT cells were challenged with TNF-α, either alone or in combination with the costimulatory cytokines IFN-γ or IL-4, and the release of proinflammatory cytokines and mediators (IL-8, IL-6, s-ICAM-1, and TSLP) was evaluated. The results showed that RNLE preferentially interferes with IFN-γ signaling, demonstrating only negligible activity on TNF-α or IL-4. This effect was attributed to flavonols, which might also account for the ability of RNLE to impair TNF-α/IL-4-induced TSLP release in a cAMP-independent manner. These results suggest that RNLE could have an antiallergic effect mediated in keratinocytes via mechanisms beyond histamine involvement. In conclusion, the discovery of RNLE preferential activity against IFN-γ-mediated inflammation suggests potential selectivity against Th1 type response and the possible use in Th1 inflammatory diseases.  相似文献
