The electrophilic abstraction of chloride ion by SbCl5 from diphenylchloroborane in CD3NO2 resulted in a transient species of which 11B NMR chemical shift appeared at 20 ppm. This was indicative of diphenylboron cation, stabilized with electron donor O-atom of two nitromethane molecules. The experimental 11B chemical shift for the solvated diphenylboron cation is in good agreement with that calculated by GIAO-HF/6-311+G(2d,p)//B3LYP/6-31+G(d).  相似文献

Six diterpenoids ( 1 – 6 ) belonging to the clerodane and tetracyclic diterpene types were isolated from the CHCl3 extract of Ballota limbata. The structures of the new compounds 1 and 4 – 6 (named ballatenolide A and limbatenolides A–C, resp.) were established by their spectral data, and their relative configuration was determined by 2D NMR. Compounds 2 and 3 were identified as known clerodanes. Compound 1 had a unique feature, i.e., an α,β‐unsaturated γ‐lactone moiety at C(4)/C(6), whereas limbatenolides A–C ( 4 – 6 ) belonged to the rare class of tetracyclic diterpenoids, which are known to occur only in the genus Ballota. All of the isolated compounds showed inhibitory potential in a cholinesterase‐inhibition assay.  相似文献

Two new tetracyclic triterpenoid trilocularol A and trilocularol A 3-glucoside and one pentacyclic triterpenoid tirlocularoside A were isolated from Corchorus trilocularis L., their structure were elucidated as 3beta,6alpha,16alpha,20(S),27-pentahydroxydammar-24(Z)-ene (1), 3beta-D-glucopyranosyloxy-6alpha,16alpha,20(S),27-tetrahydroxydammar-24(Z)-ene (2) and 2alpha,3beta,19alpha,30-tetrahydroxyurs-12-en-24,28-dioic acid 28-O-beta-D-glucopyranosyl ester (3). respectively, on the basis of detailed spectroscopic studies.  相似文献

The absorption, emission, and infrared spectra, metal (Ru) and ligand (PP) half-wave potentials, and ab initio calculations on the ligands (PP) are compared for several [L(n)()Ru(PP)](2+) and [[L(n)Ru]dpp[RuL'(n)]](4+) complexes, where L(n) and L'(n) = (bpy)(2) or (NH(3))(4) and PP = 2,2'-bipyridine (bpy), 2,3-bis(2-pyridyl)pyrazine (dpp), 2,3-bis(2-pyridyl)quinoxaline (dpq), or 2,3-bis(2pyridyl)benzoquinoxaline (dpb). The energy of the metal-to-ligand charge-transfer (MLCT) absorption maximum (hnu(max)) varies in nearly direct proportion to the difference between Ru(III)/Ru(II) and (PP)/(PP)(-) half-wave potentials, DeltaE(1/2), for the monometallic complexes but not for the bimetallic complexes. The MLCT spectra of [(NH(3))(4)Ru(dpp)](2+) exhibit three prominent visible-near-UV absorptions, compared to two for [(NH(3))(4)Ru(bpy)](2+), and are not easily reconciled with the MLCT spectra of [[(NH(3))(4)Ru]dpp[RuL(n)]](4+). The ab initio calculations indicate that the two lowest energy pi orbitals are not much different in energy in the PP ligands (they correlate with the degenerate pi orbitals of benzene) and that both contribute to the observed MLCT transitions. The LUMO energies calculated for the monometallic complexes correlate strongly with the observed hnu(max) (corrected for variations in metal contribution). The LUMO computed for dpp correlates with LUMO + 1 of pyrazine. This inversion of the order of the two lowest energy pi orbitals is unique to dpp in this series of ligands. Configurational mixing of the ground and MLCT excited states is treated as a small perturbation of the overall energies of the metal complexes, resulting in a contribution epsilon(s) to the ground-state energy. The fraction of charge delocalized, alpha(DA)(2), is expected to attenuate the reorganizational energy, chi(reorg), by a factor of approximately (1 - 4alpha(DA)(2) + alpha(DA)(4)), relative to the limit where there is no charge delocalization. This appears to be a substantial effect for these complexes (alpha(DA)(2) congruent with 0.1 for Ru(II)/bpy), and it leads to smaller reorganizational energies for emission than for absorption. Reorganizational energies are inferred from the bandwidths found in Gaussian analyses of the emission and/or absorption spectra. Exchange energies are estimated from the Stokes shifts combined with perturbation--theory-based relationship between the reorganizational energies for absorption and emission values. The results indicate that epsilon(s) is dominated by terms that contribute to electron delocalization between metal and PP ligand. This inference is supported by the large shifts in the N-H stretching frequency of coordinated NH(3) as the number of PP ligands is increased. The measured properties of the bpy and dpp ligands seem to be very similar, but electron delocalization appears to be slightly larger (10-40%) and the exchange energy contributions appear to be comparable (e.g., approximately 1.7 x 10(3) cm(-1) in [Ru(bpy)(2)dpp](2+) compared to approximately 1.3 x 10(3) cm(-1) in the bpy analogue).  相似文献

A new coumarinolignoid 8'-epi-cleomiscosin A (1) together with the new glycoside 8-O-beta-D-glucopyranosyl-6-hydroxy-2-methyl-4H-1-benzopyrane-4-one (2) have been isolated from the aerial parts of Rhododendron collettianum and their structures determined on the basis of spectroscopic evidences. Tyrosinase inhibition study of these compounds and their structure-activity relationship (SAR) were also investigated. The compounds exhibited potent to mild inhibition activity against the enzyme. Especially, the compound 1 showed strong inhibition (IC50=1.33 microM) against the enzyme tyrosinase, as compared to the standard tyrosinase inhibitors kojic acid (IC50=16.67 microM) and L-mimosine (IC50=3.68 microM), indicating its potential used for the treatment of hyperpigmentation associated with the high production of melanocytes.  相似文献

ABSTRACT

Stopping cross sections (SCS) for protons, alphas and Li ions are calculated with a modified form of our earlier work by incorporating a different electron density distribution of target materials; this involves four parameters – two projectile dependent and the rest two remain fixed. The prosed model has been tested for three stripped ion (H+, He2 + and Li3 +) projectiles and found that it describes quite satisfactorily the experimental SCS data from low energies with projectile velocities nearing v = Z1v0 (with Z1 as the atomic number and v0 the Bohr velocity) up to 100.0 MeV over a wide range of stopping media with atomic numbers Z2 =3–100.  相似文献

In this study, the anti-tumour-promoting and thermal-induced protein denaturation inhibitory activities of β-sitosterol (1) and lupeol (2), isolated from Diospyros lotus L., were explored. Compound 1 showed a marked concentration-dependent inhibition against 12-O-tetradecanoylphorbol-13-acetate (20 ng/32 pmol)-induced Epstein–Barr virus early antigen activation in Raji cells with IC50 of 270 μg/ml, without significant toxicity (70% viability). Compound 2 showed significant anti-tumour-promoting effect with IC50 of 412 μg/ml, without significant toxicity (60% viability). In heat-induced protein denaturation assay, compound 1 exhibited a concentration-dependent attenuation with a maximum effect of 73.5% at 500 μg/ml with EC50 of 117 μg/ml, while compound 2 exhibited a maximum effect of 59.2% at 500 μg/ml with EC50 of 355 μg/ml. Moreover, in silico docking studies against the phosphoinositide 3-kinase enzyme also show the inhibitory potency of these compounds. In short, both the compounds exhibited a marked anti-tumour-promoting and potent inhibitory effect on thermal-induced protein denaturation.  相似文献

Three-dimensional quantitative structure-activity relationship (3D-QSAR) models have been constructed using the comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) for a series of structurally related steroidal alkaloids as butyrylcholinesterase (BuChE) inhibitors. Docking studies were employed to position the inhibitors into the BuChE active site to determine the most probable binding mode. The strategy was to explore multiple inhibitor conformations in producing a more reliable 3D-QSAR model. These multiple conformations were derived using the FlexS program. The conformation selection step for CoMFA was done by genetic algorithm. The genetic algorithm based CoMFA approach was found to be the best. Both CoMFA and CoMSIA yielded significant cross-validated q(2) values of 0.701 and 0.627 and the r(2) values of 0.979 and 0.982, respectively. These statistically significant models were validated by a test set of five compounds. Comparison of CoMFA and CoMSIA contour maps helped to identify structural requirements for the inhibitors and serves as a basis for the design of the next generation of the inhibitor analogues. The results demonstrate that the combination of ligand-based and receptor-based modeling with use of a genetic algorithm is a powerful approach to build 3D-QSAR models. These data can be used for the lead optimization process with respect to inhibition enhancement which is important for the drug discovery and development for Alzheimer's disease.  相似文献

Ab initio calculations were carried out to understand the effect of electron donating groups (EDG) and electron withdrawing groups (EWG) at the C5 position of cytosine (Cyt) and saturated cytosine (H2Cyt) of the deamination reaction. Geometries of the reactants, transition states, intermediates, and products were fully optimized at the B3LYP/6-31G(d,p) level in the gas phase as this level of theory has been found to agree very well with G3 theories. Activation energies, enthalpies, and Gibbs energies of activation along with the thermodynamic properties (ΔE, ΔH, and ΔG) of each reaction were calculated. A plot of the Gibbs energies of activation (ΔG) for C5 substituted Cyt and H2Cyt against the Hammett σ-constants reveal a good linear relationship. In general, both EDG and EWG substituents at the C5 position in Cyt results in higher ΔG and lower σ values compared to those of H2Cyt deamination reactions. C5 alkyl substituents ( H,  CH3,  CH2CH3,  CH2CH2CH3) increase ΔG values for Cyt, while the same substituents decrease ΔG values for H2Cyt which is likely due to steric effects. However, the Hammett σ-constants were found to decrease at the C5 position of cytosine (Cyt) and saturated cytosine (H2Cyt) on the deamination reaction. Both ΔG and σ values decrease for the substituents Cl and Br in the Cyt reaction, while ΔG values increase and σ decrease in the H2Cyt reaction. This may be due to high polarizability of bromine which results in a greater stabilization of the transition state in the case of bromine compared to chlorine. Regardless of the substituent at C5, the positive charge on C4 is greater in the TS compared to the reactant complex for both the Cyt and H2Cyt. Moreover, as the charges on C4 in the TS increase compared to reactant, ΔG also increase for the C5 alkyl substituents ( H,  CH3,  CH2CH3,  CH2CH2CH3) in Cyt, while ΔG decrease in H2Cyt. In addition, analysis of the frontier MO energies for the transition state structures shows that there is a correlation between the energy of the HOMO–LUMO gap and activation energies.  相似文献
