A systematic approach to the implementation of a quality management system in the teaching laboratories of higher education institutions is described, based on the experience of the Faculty of Chemistry of the University of Barcelona. The main planning issues (definition of the aims and scope of the project, adoption of a model and of the appropriate strategy, leadership and adherence of personnel) are dealt with. Functionalised hydroxylamine derivatives of (S)-proline and pipecolic acid have been prepared using a Cope elimination. These hydroxylamines have been found to undergo oxidation to the nitrone either in the presence of air or a catalytic quantity of TPAP. An intramolecular 1,3-dipolar cycloaddition then occurs between the nitrone and pendant double bond to give tricyclic lactams and lactones with high diastereoselectivity.  相似文献

Obtaining an accurate theoretical model for the activation of dinitrogen by three-coordinate molybdenum amide complexes (e.g. Mo(NH2)3) is difficult due to the interaction of various high- and low-spin open-shell complexes along the reaction coordinate which must be treated with comparable levels of accuracy in order to obtain reasonable potential energy surfaces. Density functional theory with present-day functionals is a popular choice in this situation; however, the dinitrogen activation reaction energetics vary substantially with the choice of functional. An assessment of the reaction using specialized wave function based methods indicates that although current density functionals in general agree qualitatively on the mechanistic details of the reaction, a variety of high-level electron correlation methods (including CCSD(T), OD(T), CCSD(2), KS-CCSD(T), and spin-flip CCSD) provide a consistent but slightly different representation of the system.  相似文献

The syntheses of three decalin-based tert-thiols displaying varying degrees of solubility in aqueous milieu are described. The S-nitroso derivatives of these compounds have also been prepared and the structures of two of these determined by single crystal X-ray diffraction. These compounds have been designed for studying the interaction of nitric oxide (NO) with thiols under physiological conditions.  相似文献

A priori evaluations, using Hartree-Fock self-consistent-field (SCF) theory or density-functional theory (DFT), of the current passing between two electrodes through a single bridging molecule result in predicted conductivities that may be up to one to two orders of magnitude larger than observed ones. We demonstrate that this is, in part, often due to the improper application of the computational methods. Conductivity is shown to arise from tunneling between junction states of the electrodes through the molecule; these states are inherently either quasi two-fold or four-fold degenerate and always comprise the (highest occupied molecular orbital) HOMO band at the Fermi energy of the system. Frequently, in previous cluster based molecular conduction calculations, closed-shell SCF or Kohn-Sham DFT methods have been applied to systems that we demonstrate to be intrinsically open shell in nature. Such calculations are shown to induce artificial HOMO-LUMO (LUMO-lowest unoccupied molecular orbital) band splittings that Landauer-based formalisms for steady-state conduction interpret as arising from extremely rapid through-molecule tunneling at the Fermi energy, hence, overestimating the low-voltage conductivity. It is demonstrated that these shortcomings can be eliminated, dramatically reducing calculated current magnitudes, through the alternate use of electronic-structure calculations based on the spin-restricted open-shell formalism and related multiconfigurational SCF of DFT approaches. Further, we demonstrate that most anomalies arising in DFT implementations arise through the use of hybrid density functionals such as B3LYP. While the enhanced band-gap properties of these functionals have made them the defacto standard in molecular conductivity calculations, we demonstrate that it also makes them particularly susceptible to open-shell anomalies.  相似文献

The use of the solution redox species, [Os(bpy)2Cl2]+/0, [Os(bpy)2(MeIm)Cl]2+/+ and [Fe(CN)6]4−/3−, where bpy is 2,2-bipyridine and MeIm is N-methylimidazole, as electron mediators in the enzymatic reduction of oxygen by tyrosinase is investigated. Co-immobilization of both enzyme and an osmium redox mediator in a hydrogel on glassy carbon electrodes results in a biosensor for the ‘reagentless’ addressing of enzyme activity, consuming only oxygen present in solution. Immobilized enzyme inhibition biosensors can thus be constructed for the detection of tyrosinase inhibitors, such as sodium azide, using this approach. The enzyme inhibition biosensor can detect levels of azide as low as 5 × 10−6 mol dm−3 in solution and may be useful in environmental monitoring applications and as an early warning poison sensor.  相似文献

Super‐resolution microscopy (SRM) greatly benefits from the ability to install small photostable fluorescent labels into proteins. Genetic code expansion (GCE) technology addresses this demand, allowing the introduction of small labeling sites, in the form of uniquely reactive noncanonical amino acids (ncAAs), at any residue in a target protein. However, low incorporation efficiency of ncAAs and high background fluorescence limit its current SRM applications. Redirecting the subcellular localization of the pyrrolysine‐based GCE system for click chemistry, combined with DNA‐PAINT microscopy, enables the visualization of even low‐abundance proteins inside mammalian cells. This approach links a versatile, biocompatible, and potentially unbleachable labeling method with residue‐specific precision. Moreover, our reengineered GCE system eliminates untargeted background fluorescence and substantially boosts the expression yield, which is of general interest for enhanced protein engineering in eukaryotes using GCE.  相似文献

The assembly of proteins into highly organized fibrillar aggregates is a key process in biology, biotechnology, and human disease. It has been shown that proteins retain a small, yet significant propensity to aggregate when they are folded into compact globular structures, and this may be physiologically relevant, particularly when considering that proteins spend most of their lifespan into such compact states. Proteins from the acylphosphatase-like structural family have been shown to aggregate via different mechanisms, with some members forming native-like aggregates as a first step of their aggregation process and others requiring unfolding as a first necessary step. Here we use the acylphosphatase from Sulfolobus solfataricus to show that assembly of folded protein molecules into native-like aggregates is prevented by single-point mutations that introduce structural protections within one of the most flexible region of the protein, the peripheral edge beta-strand 4. The resulting mutants do not form native-like aggregates, but can still form thioflavin T-binding and beta-structured oligomers, albeit more slowly than the wild-type protein. The kinetic data show that formation of the latter species proceeds via an alternative mechanism that is independent of the transient formation of native-like aggregates.  相似文献

Separation science plays a crucial role in the isolation of novel compounds contained in complex matrices. Yet their rationale employment needs preliminary structure elucidation, which usually requires sufficient aliquots of grade substances to characterize the molecule by nuclear magnetic resonance experiments. In this study, two peculiar oxa-tricycloundecane ethers were isolated by means of preparative multidimensional gas chromatography from the brown alga species Dictyota dichotoma (Huds.) Lam., aiming to assign their 3D structures. Density functional theory simulations were carried out to select the correct configurational species matching the experimental NMR data (in terms of enantiomeric couples). In this case, the theoretical approach was crucial as the protonic signal overlap and spectral overcrowding were preventing any other unambiguous structural information. Just after the identification through the density functional theory data matching of the correct relative configuration it was possible to verify an enhanced self-consistency with the experimental data, confirming the stereochemistry. The results obtained further pave the way toward structure elucidation of highly asymmetric molecules, whose configuration cannot be inferred by other means or strategies.  相似文献

A straightforward to assemble catalytic system for the intermolecular hydroacylation reaction of beta-S-substituted aldehydes with activated and unactivated alkenes and alkynes is reported. These catalysts promote the hydroacylation reaction between beta-S-substituted aldehydes and challenging substrates, such as internal alkynes and 1-octene. The catalysts are based upon [Rh(cod)(DPEphos)][ClO(4)] (DPEphos=bis(2-diphenylphosphinophenyl)ether, cod=cyclooctadiene) and were designed to make use of the hemilabile capabilities of the DPEphos ligand to stabilise key acyl-hydrido intermediates against reductive decarbonylation, which results in catalyst death. Studies on the stoichiometric addition of aldehyde (either ortho-HCOCH(2)CH(2)SMe or ortho-HCOC(6)H(4)SMe) and methylacrylate to precursor acetone complexes [Rh(acetone)(2)(DPEphos)][X] [X=closo-CB(11)H(6)Cl(6) or [BAr(F) (4)] (Ar(F)=3,5-(CF(3))(2)C(6)H(3))] reveal the role of the hemilabile DPEphos ligand. The crystal structure of [Rh(acetone)(2)(DPEphos)][X] shows a cis-coordinated diphosphine ligand with the oxygen atom of the DPEphos distal from the rhodium. Addition of aldehyde forms the acyl hydride complexes [Rh(DPEphos)(COCH(2)CH(2)SMe)H][X] or [Rh(DPEphos)(COC(6)H(4)SMe)H][X], which have a trans-spanning DPEphos ligand and a coordinated ether group. Compared to analogous complexes prepared with dppe (dppe=1,2-bis(diphenylphosphino)ethane), these DPEphos complexes show significantly increased resistance towards reductive decarbonylation. The crystal structure of the reductive decarbonylation product [Rh(CO)(DPEphos)(EtSMe)][closo-CB(11)H(6)I(6)] is reported. Addition of alkene (methylacrylate) to the acyl-hydrido complexes forms the final complexes [Rh(DPEphos)(eta(1)-MeSC(2)H(4)-eta(1)-COC(2)H(4)CO(2)Me)][X] and [Rh(DPEphos)(eta(1)-MeSC(6)H(4)-eta(1)-COC(2)H(4)CO(2)Me)][X], which have been identified spectroscopically and by ESIMS/MS. Intermediate species in this transformation have been observed and tentatively characterised as the alkyl-acyl complexes [Rh(CH(2)CH(2)CO(2)Me)(COC(2)H(4)SMe)(DPEphos)][X] and [Rh(CH(2)CH(2)CO(2)Me)(COC(6)H(4)SMe)(DPEphos)][X]. In these complexes, the DPEphos ligand is now cis chelating. A model for the (unobserved) transient alkene complex that would result from addition of alkene to the acyl-hydrido complexes comes from formation of the MeCN adducts [Rh(DPEphos)(MeSC(2)H(4)CO)H(MeCN)][X] and [Rh(DPEphos)(MeSC(6)H(4)CO)H(MeCN)][X]. Changing the ligand from DPEphos to one with a CH(2) linkage, [Ph(2)P(C(6)H(4))](2)CH(2), gave only decomposition on addition of aldehyde to the acetone precursor, which demonstrated the importance of the hemiabile ether group in DPEphos. With [Ph(2)P(C(6)H(4))](2)S, the sulfur atom has the opposite effect and binds too strongly to the metal centre to allow access to productive acetone intermediates.  相似文献
