Baccharis dracunculifolia is a plant native from Brazil, commonly known as 'Alecrim-do-campo' and 'Vassoura' and used in alternative medicine for the treatment of inflammation, hepatic disorders and stomach ulcers. Previous studies reported that artepillin C (ArtC, 3-{4-hydroxy-3,5-di(3-methyl-2-butenyl)phenyl}-2(E)-propenoic acid), is the main compound of interest in the leaves. This study was undertaken to assess the mutagenic effect of the ethyl acetate extract of B. dracunculifolia leaves (Bd-EAE: 11.4-182.8 μg/plate) and ArtC (0.69-10.99 μg/plate) by the Ames test using Salmonella typhimurium strains TA98, TA97a, TA100 and TA102, and to compare the protective effects of Bd-EAE and ArtC against the mutagenicity of a variety of direct and indirect acting mutagens such as 4-nitro-O-phenylenediamine, sodium azide, mitomycin C, benzo[a]pyrene, aflatoxin B1, 2-aminoanthracene and 2-aminofluorene.The mutagenicity test showed that Bd-EAE and ArtC did not induce an increase in the number of revertant colonies indicating absence of mutagenic activity. ArtC showed a similar antimutagenic effect to that of Bd-EAE in some strains of S. typhimurium, demonstrating that the antimutagenic activity of Bd-EAE can be partially attributed to ArtC. The present results showed that the protective effect of whole plant extracts is due to the combined and synergistic effects of a complex mixture of phytochemicals, the total activity of which may result in health benefits.  相似文献

According to the World Health Organization, half of the World's population, approximately 3.3 billion people, is at risk for developing malaria. Nearly 700,000 deaths each year are associated with the disease. Control of the disease in humans still relies on chemotherapy. Drug resistance is a limiting factor, and the search for new drugs is important. We have designed and synthesized new 2-(trifluoromethyl)[1,2,4]triazolo[1,5-a]pyrimidine derivatives based on bioisosteric replacement of functional groups on the anti-malarial compounds mefloquine and amodiaquine. This approach enabled us to investigate the impact of: (i) ring bioisosteric replacement; (ii) a CF? group substituted at the 2-position of the [1,2,4]triazolo[1,5-a]pyrimidine scaffold and (iii) a range of amines as substituents at the 7-position of the of heterocyclic ring; on in vitro activity against Plasmodium falciparum. P. falciparum dihydroorotate dehydrogenase (PfDHODH) through strong hydrogen bonds. The presence of a trifluoromethyl group at the 2-position of the [1,2,4]triazolo[1,5-a]pyrimidine ring led to increased drug activity. Thirteen compounds were found to be active, with IC?? values ranging from 0.023 to 20 μM in the anti-HRP2 and hypoxanthine assays. The selectivity index (SI) of the most active derivatives 5, 8, 11 and 16 was found to vary from 1,003 to 18,478.  相似文献

Intact cell mass spectrometry biotyping, a collection of methods for classification of cells based on mass spectrometric fingerprints, is an established method in clinical and environmental microbiology. It has recently also been applied to the investigation of mammalian cells including primary blood cells and cultured cells. However, few automated procedures suitable for higher throughput and little analytical standardization of mammalian biotyping approaches have been reported so far. Here, we present a novel automated method that robustly classifies as few as 250 cells per spot. Automatically acquired cell fingerprints from cultured and primary cells show high technical (R?>?0.95) and biological reproducibility (R?=?0.83–0.96), with a median peak variance below 12?%. Ion suppression is shown to be a major concern at higher cell numbers and needs to be carefully monitored. We demonstrate that intact cell mass spectrometric signatures of different cell lines start to resemble each other at higher trifluoroacetic acid (TFA) concentrations and that therefore low concentrations of TFA in the matrix solution are preferred. We show that in vitro differentiation of HL-60 cells into a neutrophil-like phenotype can be rapidly and robustly monitored. We utilize the method for global analysis of person-to-person differences in mass spectral signatures of intact polymorphonuclear neutrophils and monocytes obtained from healthy volunteers. Our data suggest that automated MALDI mass spectrometry cell biotyping could be a useful complementary approach in clinical cell analysis.

Polybenzimidazolium hydroxides are of potential interest for the use in alkaline anion exchange membrane fuel cells (AAEMFC). Introduction of an ether group in para-position of the 2-phenyl substituent improves the mesomeric stabilization of imidazolium cations, and O-PBI seems to be more stable than meta-PBI under alkaline conditions. NMR, IR and XPS analysis show a structural change when the iodide form is exchanged into the hydroxide form. Surprisingly, this structural change is subject to a pH sensitive equilibrium and the pure imidazolium form is retained immediately when the pH is lowered. The molecular structure at high pH is either the 2-carbinol or the amine-amide. Further hydrolysis necessitates excess hydroxide.  相似文献

Analytical characterization of dimethacrylate-tyrosine-lysine-tyrosine (DMTLT, a new biodegradable acrylic cross-linker synthesized at our laboratory) is carried out using CE-MS. DMTLT is a pseudopeptide composed by tyrosine-lysine-tyrosine amino acids linked through urea bonds with two methacrylic groups, one at each end of the molecule, making this compound an excellent cross-linker for polymerization reactions and for obtaining new biodegradable materials. A new CE-MS method is developed for the characterization of DMTLT and its products of degradation after basic hydrolysis. In order to carry out an exhaustive examination of such degradation products methods based on CE coupled to IT and TOF-MS are employed. Based on CE-IT-MS results and the elemental composition of the degradation products obtained by CE-TOF-MS, conclusions on the mechanism and kinetic of hydrolysis of DMTLT are obtained confirming both the usefulness of CE-MS to characterize new biomaterials and the applicability of DMTLT for preparing new biodegradable polymers. These results are corroborated through the CE-MS detection of the identified products of degradation in a dimethyl acrylamide polymer cross-linked with DMTLT.  相似文献

(Hydroxypropyl)cellulose (HPC) dilute aqueous solutions in the presence of sodium cholate (CS), sodium deoxycholate (DC), and sodium dodecylsulphate (SDS) were investigated. The hydrophobicity parameter (I 1/I 3) from fluorescence has shown a critical aggregation concentration (CAC) lower than the critical micellar concentration (CMC). One or two breakpoints were observed in the curve conductivity vs surfactant concentration. The thermodynamic parameters of aggregation (, and ) and the degree of counterion dissociation were calculated. Evidences for the secondary aggregation of CS/water system were found. The relative viscosity increases for HPC/bile salt solutions only at high surfactant concentrations, whereas for HPC/SDS, it passes through a maximum. The cloud points of both HPC/bile salt solutions at higher surfactant concentrations reach a temperature plateau value around 324 K, while for HPC/SDS, it exceeds 373 K at low SDS concentrations. The functionalized flavylium salt 6-hexyl-7-hydroxy-4-methyflavylium chloride (HHMF) was employed to probe some of the fundamental features of proton transfer reactions at the surface of anionic sodium dodecyl sulfate (SDS) and cationic hexadecyltrimethylammonium chloride (CTAC) micelles. In contrast to most ordinary flavylium salts, HHMF is insoluble in water, but readily incorporates into SDS and CTAC micelles. In the ground state, the rate constant for deprotonation of the acid form (AH+) of HHMF decreases 100-fold upon going from CTAC (kd = 3.0 x 10(6) s(-1)) to SDS (kd = 1.4 x 10(4) s(-1)), consistent with the presence of an activation barrier for proton transfer in the ground state and reflecting, respectively, stabilization or destabilization of the AH+ cation by the micelle. Reprotonation of A is diffusion-controlled in both micelles (kp(SDS) = (2.1 x 10(11))[H+]aq s(-1) and kp(CTAC) = (3.7 x 10(8))[H+]aq s(-1)), the difference reflecting the rate of proton entry into the micelles. In the excited singlet state, the rate constants for deprotonation of the AH+* form of HHMF are similar in the two micelles (2.4 x 10(10) s(-1)), consistent with activationless proton transfer. Reprotonation of the excited A is dominated by fast geminate recombination of the photogenerated (A*-H+) pair at the micelle surface (k(rec)(SDS) = 6.1 x 10(9) s(-1) and k(rec)(CTAC) = 3.4 x 10(10) s(-1)) and the net efficiencies of geminate recombination are quite similar in SDS (0.89) and CTAC (0.86).  相似文献

Ab initio calculations at the MP2(full)/6-31++G**, RI-MP2(full)/6-31++G**, and RI-MP2(full)/6-311++G(2d,2p) levels of theory demonstrate important synergic effects between two noncovalent interactions that involve aromatic rings, that is, cation-pi and pi-pi interactions. The presence of a cation interacting with the pi cloud of an aromatic ring favors the face-to-face stacking interaction with additional aromatic rings. This effect is extended in the space up to five stacked aromatic rings.  相似文献

N-containing TiO(2)-based nanostructured materials (average particle size approximately 10 nm) with an anatase-type structure were investigated using oxygen (O) K-edge and titanium (Ti) K- and L-edge X-ray absorption near-edge spectroscopy (XANES). The Ti K pre-edge features indicate that samples predominantly contain ([6])Ti with some ([5])Ti, and there is no evidence for ([4])Ti. We observed that those samples with a larger fraction of Ti in a fivefold coordination, that is, with a significant number of oxygen vacancies, also present a modified Ti environment at the medium-range scale. The presence of these defects drastically modifies the electronic structure of the conduction band, as evidenced by the O K XANES spectra, but does not result in the presence of reduced Ti(3+) states. We discuss the influence of N-doping on titania nanoparticles and their structure, electronics and photocatalytic activity.  相似文献

Paracoccidioidomycosis (PCM) is the most prevalent human mycosis in Latin America. The infection is thought to take place firstly in the lungs and then may disseminate to other organs and tissues. Treatment by currently available antifungals is lengthy, the drugs may have undesirable side effects, and some are costly. Occasional resistant strains of Paracoccidioides brasiliensis, the causative agent of PCM, have been reported. So, the search for more efficient treatments or adjuvant therapies has to be continued. In this work, we evaluated the effects of HeNe laser irradiation on cutaneous inflammatory lesions caused by the inoculation of 5 x 10(6)/0.1ml yeasts cells into the back footpad of Balb/c mice. HeNe irradiation (lambda=632.8nm, 3mW, incident energy of 3J/cm(2)) was applied at days 7, 8 and 9 post-infection and histological and immunohistochemical analysis were done. Unirradiated animals were used as controls. The results showed that laser-treated mice presented reduction of footpad edema, faster cutaneous wound healing, confluent granuloma, diffuse- and more loosely distributed immunolabeling for TNF-alpha, enhanced labeling of IFN-gamma and any P. brasiliensis form detected, whereas multiple viable fungi were seen in diffuse widespread granulomas obtained from non-treated mice foot-pad. Fungi that were harvested from laser-treated animals presented no capability of growth in vitro as compared to those obtained from non-treated mice. We conclude that HeNe laser irradiation was able to inhibit the progress of inflammatory local reaction produced by P. brasiliensis infection and influence local cytokines production. We suggest that this treatment modality can be a useful coadjuvant tool to be combined with antifungal agents in the treatment of PCM ulcerations. The mechanisms involved in laser therapy of PCM lesions need further investigation.  相似文献
