In a clinical diagnosis microbiology laboratory, the current method of identifying bacterial isolates is based mainly on phenotypic characteristics, for example growth pattern on different media, colony morphology, Gram stain, and various biochemical reactions. These techniques collectively enable great accuracy in identifying most bacterial isolates, but are costly and time-consuming. In our clinical microbiology laboratory, we prospectively assessed the ability of matrix-assisted laser desorption ionization–time of flight mass spectrometry (MALDI–TOF MS) to identify bacterial strains that were routinely isolated from clinical samples. Bacterial colonies obtained from a total of 468 strains of 92 bacterial species isolated at the Department of Clinical Laboratory at Chiba University were directly placed on target MALDI plates followed by addition of CHCA matrix solution. The plates were then subjected to MALDI–TOF MS measurement and the microorganisms were identified by pattern matching with the libraries in the BioTyper 2.0 software. Identification success at the species and genus levels was 91.7% (429/468) and 97.0% (454/468), respectively. MALDI–TOF MS is a rapid, simple, and high-throughput proteomic technique for identification of a variety of bacterial species. Because colony-to-colony differences and effects of culture duration on the results are minimal, it can be implemented in a conventional laboratory setting. Although for some pathogens, preanalytical processes should be refined, and the current database should be improved to obtain more accurate results, the MALDI–TOF MS based method performs, in general, as well as conventional methods and is a promising technology in clinical laboratories.  相似文献

Ternary sodium borosilicide, Na(8)B(74.5)Si(17.5), was newly synthesized by heating a mixture of sodium, silicon and amorphous or crystalline boron at 1073-1273 K. The crystal structure of the black hexagonal prismatic single crystal obtained at 1273 K was analyzed. The X-ray diffraction reflections of the crystal were indexed with hexagonal cell parameters a = 10.2392(3) ? and c = 10.9215(4) ? (space group P6(3)/mmc, No. 194). The structural formula could be represented as Na(8)(B(12))(6)Si(16)[BSi](1.5)[B(2)](0.5). B(12) icosahedra form a three-dimensional framework having small triangular and large hexagonal channels along the c axis. Chains of [-Si-(Si-Si)(3)-Si-] surrounded by Na atoms are located in the large channels. Trigonal prism cages for Na atoms and for atom pairs of Si-B or B-B are alternately arrayed in the small channels. The ratio of Si-B and B-B pairs in the cage is around 3:1.  相似文献

End‐functionalized poly(phenylacetylene)s were synthesized by the polymerization of phenylacetylene (PA) using the well‐defined palladium catalysts represented as [(dppf)PdBr(R)] {dppf = 1,1′‐bis(diphenylphosphino)ferrocene}. The Pd catalysts having a series of R groups such as o‐tolyl, mesityl, C(Ph)?CPh2, C6H4o‐CH2OH, C6H4p‐CN, and C6H4p‐NO2 in conjunction with silver triflate polymerized PA to give end‐functionalized poly(PA)s bearing the corresponding R groups in high yields. The results of IR and NMR spectroscopies and MALDI‐TOF mass analyses proved the introduction of these R groups at one end of each polymer chain. The poly(PA) bearing a hydroxy end group was applied as a macroinitiator to the synthesis of a block copolymer composed of poly(PA) and poly(β‐propiolactone) moieties. © 2010 Wiley Periodicals, Inc. The magnetic property of Co2B2O5 and the optical property of M2B2O5 (M = Mn, Co) were investigated. Co2B2O5 showed antiferromagnetic behavior below the Néel temperature of TN ≈ 45 K, and the Weiss temperature was TW = +7.7 K. The effective magnetic moment of Co was 4.96 μB, which indicated that Co was divalent and in a high-spin state. Absorptions attributed to the d–d transitions in Mn2+ and Co2+ ions were observed at 250–650 nm in the diffuse reflection spectra. The optical absorption edges of Mn2B2O5 and Co2B2O5 were at 243 nm (5.11 eV) and 299 nm (4.15 eV), respectively.  相似文献

Novel pyridinium salts [N‐(α‐phenylbenzyl)‐, N‐(1‐naphthylmethyl)‐, or N‐cinnamyl p‐ or o‐cyanopyridinium hexafluoroantimonates] were synthesized by the reaction of p‐ or o‐cyanopyridine and the corresponding bromides followed by anion exchange with KSbF6. These pyridinium salts polymerized epoxy monomers at lower temperatures than previously reported for N‐benzyl‐2‐cyanopyridinium hexafluoroantimonate. The o‐substituted pyridinium salts showed higher activity than the p‐substituted ones, and the crosslinked epoxy polymers cured with these initiators showed higher glass‐transition temperatures. These pyridinium salts photoinitiated radical polymerization as well as cationic polymerization. We have created a simple algorithm for automatically predicting the explicit solvent atom distribution of biomolecules. The explicit distribution is coerced from the three‐dimensional (3D) continuous distribution resulting from a 3D reference interaction site model (3D‐RISM) calculation. This procedure predicts optimal location of solvent molecules and ions given a rigid biomolecular structure and the solvent composition. We show examples of predicting water molecules near the KNI‐272 bound form of HIV‐1 protease and predicting both sodium ions and water molecules near the rotor ring of F‐adenosine triphosphate (ATP) synthase. Our results give excellent agreement with experimental structure with an average prediction error of 0.39–0.65 Å. Further, unlike experimental methods, this method does not suffer from the partial occupancy limit. Our method can be performed directly on 3D‐RISM output within minutes. It is extremely useful for examining multiple specific solvent–solute interactions, as a convenient method for generating initial solvent structures for molecular dynamics calculations, and may assist in refinement of experimental structures. © 2012 Wiley Periodicals, Inc.  相似文献

A highly sensitive, selective and simple method is described for the determination of histamine by high-performance liquid chromatography (HPLC) with fluorescence detection. The method is based on an intramolecular excimer-forming fluorescence derivatization of histamine with 4-(1-pyrene)butyric acid N-hydroxysuccinimide ester (PSE), followed by reversed-phase HPLC. Histamine, having two amino moieties in a molecule, was converted to the dipyrene-labeled derivative by reaction with PSE. The derivative afforded intramolecular excimer fluorescence (450-540 nm), which can clearly be discriminated from the monomer fluorescence (370-420 nm) emitted from PSE. Typically, a 10 micro L sample solution was mixed with 100 micro L of derivatization reagent solution, which was a mixture of 0.5 mm PSE in acetonitrile and 0.5 mm potassium carbonate in water (8:2, v/v). The derivatization was carried out at 100 degrees C for 90 min. The PSE derivative of histamine could be separated by reversed-phase ODS column with isocratic elution using acetonitrile:water (82:18, v/v) containing 0.03% triethylamine. The detection limit (singnal-to-noise ratio = 3) of histamine was 0.5 fmol for a 30 micro L injection. The method was successfully applied to the determination of histamine in human urine, and had enough selectivity and sensitivity for urinary histamine quantification.  相似文献

Novel aliphatic aminimides were synthesized from the corresponding carboxylic acid esters, 1,1‐dimethylhydrazine, and epoxides in 54–95% yields. Bulk polymerization of glycidyl phenyl ether (GPE) with 3 mol % of the aminimides was evaluated by DSC as a model process for curing of epoxy resin. All the aminimides showed no exothermic DSC peak below 120 °C but showed sharp exothermic peaks above 137 °C, indicating good thermal latency. Good relationships were observed between the calculated bond length from the carbonyl carbon to the α‐carbon of the aliphatic group (R C), DSC onset temperatures, and the thermal dissociation temperatures (Td 's) of the aminimides. The ability of chloroform solutions of p-diethylaminomethylthiacalix[4]arene 1 to extract platinum from HCl media was tested. As an ion-pair extractant, compound 1 had the highest platinum extraction ability among all the thiacalix[n]arene derivatives tested in this study. Compound 1 was a highly specific extractant for Pt(IV) ions from single-metal solutions and platinum-group metal solutions from automotive catalyst residue. Various conditions such as contact time, pH, concentration of 1, effect of chloride ions, effect of acidity and Pt(IV) concentration were studied. The distribution ratio of platinum ions (D) was calculated. The platinum complex stoichiometry ratio was obtained using slope analysis. The 1–Pt(IV) complex was characterised using X-ray photoelectron spectroscopy, FT-IR and 1H NMR. Finally, the Pt(IV) extraction percentage of compound 1 was compared with that of previously reported thiacalix[n]arene-based extractants.  相似文献
