Enzyme-based electron-transfer reactions involved in the cytochrome P450 monooxygenase system were investigated in nanostructural reverse micelles. A bacterial flavoprotein, putidaredoxin reductase (PdR), was activated and shown to be capable of catalyzing the electron transport from NADH to electron-carrier proteins such as cytochrome b5 (tCyt-b5) and putidaredoxin (Pdx) in reverse micelles. Ferric tCyt-b5 in reverse micelles was effectively converted to its ferrous form by the exogenous addition of separately prepared reverse micellar solution harboring PdR and NADH. The fact that direct interactions of macromolecular proteins should be possible in the reverse micellar system encouraged us to functionalize a multicomponent monooxygenase system composed of the bacterial cytochrome P450cam (P450cam), putidaredoxin (Pdx), and PdR in reverse micelles. The successful camphor hydroxylation reaction catalyzed by P450cam was significantly dependent on the coexistence of Pdx, PdR, and NADH but not H2O2, suggesting that the oxygen-transfer reactions proceeded via a "monooxygenation" mechanism. This is the first report of a multicomponent cytochrome P450 system exhibiting enzymatic activity in organic media.  相似文献

Soluble polystyrenes with crown ether structures and hydroxyl groups adjacent to the macrorings were prepared by the reaction of epoxide-containing polystyrenes with monoaza-15-crown-5 or monoaza-18-crown-6. Rate of formation of the polymer-bound alkali cation-crown alkoxide ion pair from the soluble polystyrenes and aqueous NaOH or KOH depended on the loading of crown ether and hydroxyl units and on the size of the macroring. The elimination of HCl from less reactive 2-chloroethylbenzene with aqueous NaOH or KOH in the presence of the soluble polystyrene catalysts under two-phase conditions was limited mainly by the intrinsic reaction rate. The elimination of HBr from more reactive 2-bromoethylbenzene in the presence of the less (11%) ring-substituted polymer catalyst with 18-crown unit was limited by the alkoxide formation rate. © 1994 John Wiley & Sons, Inc.  相似文献

A new quaternary layered carbide, Zr2[Al3.56Si0.44]C5, has been synthesized and characterized by X-ray powder diffraction, transmission electron microscopy and thermopower and electrical conductivity measurements. The crystal structure was successfully determined using direct methods, and further refined by the Rietveld method. The crystal is trigonal (space group R3m, Z=3) with lattice dimensions of a=0.331059(5), c=4.09450(5) nm and V=0.38864(1) nm3. The final reliability indices calculated from the Rietveld refinement were Rwp=6.24%, Rp=4.21% and RB=0.82%. The crystal structure is composed of electroconductive NaCl-type ZrC slabs separated by Al4C3-type [Al3.56Si0.44]C3 layers. This material had thermoelectric properties superior to those of the ternary layered carbides Zr2Al3C4 and Zr3Al3C5, with the power factor reaching 7.6×10−5W m−1 K−2.  相似文献

Copper-mediated atom transfer radical polymerization (ATRP) of a protected silanol group-holding methacrylate, methacryloxypropyltrimethoxysilane (MOPS), was investigated. In a dry condition using carefully distilled solvent and monomer, the polymerization proceeded in a living fashion providing a low-polydispersity polymer with a predicted molecular weight. The ATRP in conjunction with the sequential monomer addition of methyl methacrylate (MMA) and MOPS afforded a block copolymer of the type PMMA-b-poly(MMA-r-MOPS). The heat treatment of a solution of the block copolymer in the presence of a catalytic amount of ammonia gave a polymeric core-shell nanoparticle with a shell of PMMA moieties and a core of the poly(MMA-r-MOPS) blocks cross-linked via the condensation of the trimethoxysilane groups of the MOPS moieties.  相似文献

An exothermic phenomenon and a simultaneous rapid evolution of a small amount of carbon dioxide at ?500°C during thermal decomposition of hydromagnesite 4 MgCO3 · Mg(OH)2 · 4 H2O was studied by isothermal DSCTG in a carbon dioxide atmosphere. It was quantitatively confirmed that the exothermic phenomenon was due to crystallization of MgCO3 from the amorphous phase and that the evolution of carbon dioxide was due to decomposition of the MgCO3 by the heat of crystallization (?3.4 kcal mole?1.  相似文献

We investigated glucose and amino acid metabolism in tumors and other organs using whole body autoradiography with a short-lived positron emitter and a long-lived beta emitter. The radioactive compounds used were 2-deoxy-2-[18F]-fluoro-D-glucose (18F-FDG) with a half life of 109.8 min and L-[methyl-14C]-methionine (14C-MET) with a half life of 5,730 years. A Donryu rat weighing about 150 g was subcutaneously inoculated at the back with experimental tumors of AH109A and AH272. 74 MBq (2 mCi) of 18F-FDG and 740 kBq (20 microCi) of 14C-MET was administered and after 30 min, the rat was sacrificed. Whole body frozen sections were obtained using autocryotome. For the 18F-FDG autoradiogram, the frozen sections were exposed to an X-ray film for 6 h. After seven days, these frozen sections were again exposed to 14C-MET for a week. Cross-contamination was minimized by adjusting the exposure time, the interval of exposures and the administered dose. The accumulation of the tracers was represented as the optical density ratio of the tissue of interest to the muscle. The tumor ratios were 12.5 for 18F-FDG and 8.6 for 14C-MET showing the highest accumulation in the whole body autoradiogram. In contrast the inflammatory tissue ratios were 1.27 for 18F-FDG and 0.77 for 14C-MET showing very low amino acid metabolism. With the present double tracer whole body autoradiogram, 18F-FDG accumulation was seen in the brain and the heart but not to the liver as against 14C-MET accumulation which was seen to the liver but not to the brain and the heart.  相似文献

Whole skin lipid bio-liposomes (skin bio-liposomes), in size ranging from 2 to 8 microns, were prepared by a reverse phase evaporation technique using rat full thickness skin. Leakage of an encapsulated fluorescence probe, ANTS (delta-amino-1,3,6-naphthalene-trisulfonate), was measured by adding transdermal penetration enhancers (penetrants) into the medium where the skin bio-liposomes were present. Oleylamine induced a fast release of ANTS from the liposomes compared to lauryl-amine which showed a weak action. With these penetrants, the degree of ANTS release from the prepared bio-liposomes was found to correlate well with the results of frequency changes in the CH-asymmetric stretching band near 2920 cm-1 in the rat stratum corneum. The penetrant which caused relatively strong leakage of ANTS induced the significantly large shift of the peak toward the higher wave-numbers due to the perturbation in the structure of lipids of the stratum corneum. The skin bio-liposomes prepared from the rat full thickness skin could be useful in evaluating the penetrants.  相似文献
