A fast method for the determination of acidity constants by CZE has been recently developed. This method is based on the use of an internal standard of pK(a) similar to that of the analyte. In this paper we establish the reference pK(a) values of a set of 24 monoprotic neutral acids of varied structure that we propose as internal standards. These compounds cover the most usual working pH range in CZE and facilitate the selection of adequate internal standards for a given determination. The reference pK(a) values of the acids have been established by the own internal standard method, i.e. from the mobility differences between different acids of similar pK(a) in the same pH buffers. The determined pK(a) values have been contrasted to the literature pK(a) values and confirmed by determination of the pK(a) values of some acids of the set by the classical CE method. Some systematic deviations of mobilities have been observed in NaOH buffer in reference to the other used buffers, overcoming the use of NaOH in the classical CE method. However, the deviations affect in a similar degree to the test compounds and internal standards allowing thus, the use of NaOH buffer in the internal standard method. This fact demonstrates the better performance of the internal standard method over the classical method to correct mobility deviations, which together with its fastness makes it an interesting method for the routine determination of accurate pK(a) values of new pharmaceutical drugs and drug precursors.  相似文献

The adsorption behavior of highly charged cationic polyelectrolytes onto porous substrates is electrostatic in nature and has been shown to be highly dependent on the polyelectrolyte properties. Copolymers of acrylamide (AM) and diallyldimethylammonium chloride (DADMAC) were synthesized to have a range of macromolecular properties (i.e., charge density and molecular mass). Traditional titration methods have been complemented by fluorescence labeling techniques that were developed to directly observe the extent that fluorescently labeled poly(AM- co-DADMAC) adsorbs into the pore structure of a cellulosic substrate. Although contributing to the electrostatic driving force, the charge density acts to limit adsorption to the outermost surface under electrolyte-free conditions. However, adsorption into the pores can occur if both the molecular mass and charge density of poly(AM- co-DADMAC) are sufficiently low. Adsorption initially increases as the electrolyte concentration is increased. However, the electrostatic persistence length of poly(AM- co-DADMAC) restricts the polyelectrolyte from entering the pores. Therefore, changes in the adsorption behavior at moderate electrolyte concentrations have been attributed to swelling of the polyelectrolyte layer at the fiber exterior. The adsorption behavior changes again at high electrolyte concentrations such that poly(AM- co-DADMAC) could adsorb into the pore structure. This occurred when the electrolyte concentration was sufficient to screen the electrostatic persistence length of poly(AM- co-DADMAC), provided that the entropic driving force for adsorption still existed. It is suggested that adsorption into the pore structure is a kinetic process that is governed by localized electrostatic interactions between poly(AM- co-DADMAC) and the charges located within the pores.  相似文献

A set of 25 monoprotic bases is proposed as internal standards for pK(a) determination by capillary electrophoresis. The pK(a) of the bases is determined and compared with available literature data. The capillary electrophoresis internal standard method offers numerous advantages over other typical methods for pK(a) determination, especially of analysis time and buffer preparation. However, it requires disposing of appropriate standards with reference pK(a) value. The set of bases established in this work together with the set of acids previously established provide a reference set of compounds with well-determined acidity constants that facilitate the process of selecting appropriate internal standards for fast pK(a) determination by capillary electrophoresis in high throughput screening of pharmaceutical drugs. In addition, the performance of the method when acidic internal standards are used for the determination of acidity constants of basic internal standards has also been tested. Although higher errors may be expected in this case, good agreement is observed between determined and literature values. These results indicate that in most cases structural similarity between the analyte and the internal standard might not be an essential requirement in the internal standard method.  相似文献

Grape antioxidant dietary fiber (GADF) is a dietary supplement that combines the benefits of both fiber and antioxidants that help prevent cancer and cardiovascular diseases. The antioxidant polyphenolic components in GADF probably help prevent cancer in the digestive tract, where they are bioavailable. Mass spectrometry coupled to liquid chromatography is a powerful tool for the analysis of complex plant derivatives such as GADF. We use a combination of MS techniques, namely liquid chromatography/electrospray ionization time-of-flight mass spectrometry (LC/ESI-TOF-MS) and liquid chromatography/electrospray ionization tandem mass spectrometry (LC/ESI-MS/MS) on a triple quadrupole, for the identification of the polyphenolic constituents of the soluble fraction of GADF. First, we separated the mixture into four fractions which were tested for phenolic constituents using the TOF system in the full scan mode. The high sensitivity and resolution of the TOF detector over the triple quadrupole facilitate the preliminary characterization of the fractions. Then we used LC/ESI-MS/MS to identify the individual phenols through MS/MS experiments (product ion scan, neutral loss scan, precursor ion scan). Finally, most of the identities were unequivocally confirmed by accurate mass measurements on the TOF spectrometer. LC/ESI-TOF-MS combined with MS/MS correctly identifies the bioactive polyphenolic components from the soluble fraction of GADF. High-resolution TOF-MS is particularly useful for identifying the structure of compounds with the same LC/ESI-MS/MS fragmentation patterns.  相似文献

The properties of bulk salt solutions over wide concentration ranges are explored by a combination of simple physical theory and Monte Carlo (MC) simulations. The corrected Debye–Hückel (CDH) theory which incorporates ion size effects in a linear response approximation is extended to yield free energy and other thermodynamic properties by integration of the chemical potential over concentration. Charging integration which is usually used to obtain an electrostatic contribution of total free energy of electrolytes is avoided in this new direct approach. MC simulations are performed with a modified Widom particle insertion method, which also provides directly the ionic activity coefficients. The validity of the CDH theory is tested by comparison with the MC simulation data for 1:1, 2:1, 2:2 and 3:1 restricted primitive model (RPM) electrolytes over a wide concentration range and at various ion sizes. Mean ionic activity and osmotic coefficients calculated by the CDH theory in RPM approximation of electrolyte are fitted to experimental data by adjusting only a mean ionic diameter. Good fits up to 1 molal (m) concentration are obtained for a large number of salt solutions. MC simulations data for unrestricted primitive model (UPM) of 1:1 and 2:1 electrolytes are also fitted to the experimental data by varying the cation radius while keeping the anion radius fixed at a crystallographic value. The success of this approach is found to be salt specific. For example good fits up to 2 and 3.5 m concentrations were obtained for LiCl and LiBr, respectively. However in the case of less dissociated salts such as NaCl and KI the experimental data could only be fitted up to one molal concentration. Possibility of extending the applicability range of the CDH theory to concentrations >2 m is explored by including a concentration dependent dielectric constant as measured in experiments. Mean ionic activity coefficients for a number of salts could successfully be fitted up to 3 m concentration by adjusting only a mean ionic diameter. Difficulties encountered in simultaneously fitting the mean ionic activity and osmotic coefficients at salt concentrations >2 m are discussed.  相似文献
