A simple method for the determination of the two most abundant sialic acids released from glycoproteins based on CZE-MS is presented. Several parameters like BGE with various organic modifiers and sheath liquids were studied with respect to their suitability for the fast and easy analysis of the selected compounds by CZE-MS. Finally, a BGE containing 10 mM ammonium acetate allowed the quantification of N-acetylneuraminic acid (Neu5Ac) and N-glycolylneuraminic acid (Neu5Gc) in glycoproteins as well as human plasma samples. LOD and LOQ were 2 microg/mL and 6 microg/mL, respectively.  相似文献

A novel heterometallic 1D coordination polymer [{Ni(en)2}2(micro-NCS)4Cd(NCS)2](n) x nCH3CN (en = ethylenediamine) has been prepared using the self-assembly process in a one-pot reaction of cadmium oxide, nickel and ammonium thiocyanates with an acetonitrile solution of ethylenediamine. The complex consists of an uncommon cis-Cd(SCN)4(NCS)2(4-) fragment and a rare combination of cis-Ni(en)2(2+) and trans-Ni(en)2(2+) building blocks linked by micro(1,3)-NCS bridges into a double-stranded zigzag chain structure. Each chain is comprised of [Ni2Cd2(micro-NCS-N,S)4)] macrocycles with chair-like and rectangular-like shapes arrayed alternately. The shortest intrachain CdCd separations are 9.535(1) and 10.868(2) A, while the nearest NiNi distances are 5.418(1) and 6.612(2) A. A network of weak N-HS hydrogen bonds, involving the terminal NCS ligands and NH2-groups of en, links the infinite chains and results in the formation of an extended supramolecular three-dimensional framework. Variable-temperature (1.8-300 K) magnetic susceptibilities show a slight change of the micro(B) value at low temperature, indicative of weak antiferromagnetic interactions (J = 1.55 cm-1) between magnetic canters. High-field, high-frequency (100-400 GHz) EPR spectra were simulated using S = 1 ground state for separate Ni2+ ions with the spin Hamiltonian parameters g = 2.165, D = 0.45 cm-1 and E = 0.03 cm(-1). According to DFT calculations, the D and E parameters are -0.35 cm(-1) and 0.049 cm-1 for the cis arrangement of Ni2+ and 0.58 cm(-1) and 0.012 cm(-1) for trans.  相似文献

A family of distorted heterometallic cubanes, [Mn (III) 3Ni (II)(hmp) 3O(N 3) 3(O 2CR) 3], where O 2CR (-) is benzoate ( 1), 3-phenylpropionate ( 2), 1-adamantanecarboxylate ( 3), or acetate ( 4) and hmp (-) is the anion of 2-pyridinemethanol, was synthesized and structurally as well as magnetically characterized. These complexes have a distorted-cubane core structure similar to that found in the S = 9/2 Mn 4 cubane family of complexes. Complexes 1, 3, and 4 crystallize in rhombohedral, hexagonal, and cubic space groups, respectively, and have C 3 molecular symmetry, while complex 2 crystallizes in the monoclinic space group Cc with local C 1 symmetry. Magnetic susceptibility and magnetization hysteresis measurements and high-frequency electron paramagnetic resonance (HFEPR) spectroscopy established that complexes 1-4 have S = 5 spin ground states with axial zero-field splitting (ZFS) parameters ( D) ranging from -0.20 to -0.33 cm (-1). Magnetization versus direct-current field sweeps below 1.1 K revealed hysteresis loops with magnetization relaxation, definitely indicating that complexes 1-4 are single-molecule magnets that exhibit quantum tunneling of magnetization (QTM) through an anisotropy barrier. Complex 2 exhibits the smallest coercive field and fastest magnetization tunneling rate, suggesting a significant rhombic ZFS parameter ( E), as expected from the low C 1 symmetry. This was confirmed by HFEPR spectroscopy studies on single crystals that gave the following parameter values for complex 2: gz = 1.98, gx = gy = 1.95, D = -0.17 cm (-1), B 4 (0) = -6.68 x 10 (-5) cm (-1), E = 6.68 x 10 (-3) cm (-1), and B 4 (2) = -1.00 x 10 (-4) cm (-1). Single-crystal HFEPR data for complex 1 gave g z = 2.02, gx = gy = 1.95, D = -0.23 cm (-1), and B 4 (0) = -5.68 x 10 (-5) cm (-1), in keeping with the C 3 site symmetry of this Mn 3Ni complex. The combined results highlight the importance of spin-parity effects and molecular symmetry, which determine the QTM rates.  相似文献

We probe the local electronic structure at solvated Na+ ions in 1 M aqueous NaCl solutions as a function of pH. A dramatic change in the Na+ white line intensity in X-ray absorption is observed for high pH values, reflecting a changing local electronic structure at the Na+ ions when OH- is present. Given the relative abundance of sodium and hydroxide ions, we conclude that one OH- affects at least 2.4+/-0.6 Na+ ions in an electronically noticeable way at pH 13. From the experimental data we infer that spatially extended clusters or networks incorporating Na+ and OH- can exist in the electrolyte solution. The experimental data are complemented by molecular dynamics simulations, which indicate the presence of structured clusters incorporating Na+, OH-, and solvent molecules.  相似文献

Identification of ancient biological samples from the 1991-discovered and more than 5300-year-old Tyrolean mummy, also called iceman or Oetzi, is very difficult. The species of origins of four animal-hair-bearing samples of the accoutrement of the mummy not yet diagnosed were identified by a special proteomics method. Ha 43/91/130 and Ha 6/91, two samples from his coat, and Ha 5/91, a sample from his leggings, were assigned to sheep. The upper leather of his moccasins, Ha 2/91, was made from cattle. Despite the enormous age of these samples with partial (bio)chemical alterations, reliable identification was possible using a recently developed matrix-assisted laser desorption/ionization time-of-flight mass spectrometric ((MALDI-TOF MS)-based analytical method. The method is exclusively based on the analysis of proteins and uses minute amounts of peptides directly derived from tryptic hair digests without any separation or enrichment steps. Unknown species are identified by comparison of their peptide ion patterns with known spectra stored in existing databases. Hereby, the correlation distance, a form of Euclidean distance, and deduced parameters are used to measure similarities.If more than one potential hit remains, specific diagnostic peptide ions are used to stepwise exclude incorrect matches. These ions are specific for orders, families, subfamilies/genera and/or even species. Peptide mass fingerprinting data combined with those from collision-induced dissociation spectra (combined MS & MS/MS) were used for interpretation with the MASCOT search engine and the NCBI database to find the potential parentage of hair proteins. The cyano-substituted metallocenes [M(C5H4CN)2] (M=Fe, 1; Co, 2; Ni 3) and [M(C5Me5)(C5H4CN)] (M=Fe, 4; Co, 5; Ni, 6) were synthesized in yields up to 58 % by treating K(C5H4CN) or Tl(C5H4CN) with suitable transition-metal precursors. Cyclic voltammetry indicated that the oxidation and reduction potentials of all the cyanometallocenes were shifted to positive values by up to 0.8 V. Single-crystal X-ray structure analysis showed that 1 had eclipsed ligands, formed planes in the lattice, and--unlike usual metallocenes--lined up in stacks perpendicular to these planes. Powder X-ray studies established that 1 and 2 are isotypic. The 1H and 13C NMR spectra were recorded for all the new compounds. Signal shifts of up to delta=1500 ppm were recorded for the paramagnetic molecules 2 and 3 and were, at a given temperature, strikingly different for solution and solid-state spectra. These results pointed to antiferromagnetic interactions as a consequence of molecular ordering in the lattice, as confirmed by magnetic measurements. The temperature-dependent susceptibilities were reproduced by Heisenberg spin-chain models (H=-J sum n- 1 i=1 SiSi+1), thus yielding J=-28.3 and -10.3 cm(-1) for 2 and 3, respectively, whereas J=-11.8 cm(-1) was obtained for 3 from the Ising spin-chain model. In accordance with molecular orbital (MO) considerations, much spin density was found to be delocalized not only on the cyclopentadienyl ligand but also the cyano substituents. The magnetic interaction was interpreted as a Heitler-London spin exchange and was analyzed based on how the interaction depends on the singly occupied MOs and the shift of parallel metallocenes relative to each other.  相似文献

The quinonoid ligand-bridged diruthenium compounds [(acac)(2)Ru(mu-L(2-))Ru(acac)(2)] (acac(-)=acetylacetonato=2,4-pentanedionato; L(2-)=2,5-dioxido-1,4-benzoquinone, 1; 3,6-dichloro-2,5-dioxido-1,4-benzoquinone, 2; 5,8-dioxido-1,4-naphthoquinone, 3; 2,3-dichloro-5,8-dioxido-1,4-naphthoquinone, 4; 1,5-dioxido-9,10-anthraquinone, 5; and 1,5-diimido-9,10-anthraquinone, 6) were prepared and characterized analytically. The crystal structure analysis of 5 in the rac configuration reveals two tris(2,4-pentanedionato)ruthenium moieties with an extended anthracenedione-derived bis(ketoenolate) pi-conjugated bridging ligand. The weakly antiferromagnetically coupled {Ru(III)(mu-L(2-))Ru(III)} configuration in 1-6 exhibits complicated overall magnetic and EPR responses. The simultaneous presence of highly redox-active quinonoid-bridging ligands and of two ruthenium centers capable of adopting the oxidation states +2, +3, and +4 creates a large variety of possible oxidation state combinations. Accordingly, the complexes 1-6 exhibit two reversible one-electron oxidation steps and at least two reversible reduction processes. Shifts to positive potentials were observed on introduction of Cl substituents (1-->2, 3-->4) or through replacement of NH by O (6-->5). The ligand-to-metal charge transfer (LMCT) absorptions in the visible region of the neutral molecules become more intense and shifted to lower energies on stepwise reduction with two electrons. On oxidation, the para-substituted systems 1-4 exhibit monocation intermediates with intervalence charge transfer (IVCT) transitions of Ru(III)Ru(IV) mixed-valent species. In contrast, the differently substituted systems 5 and 6 show no such near infrared (NIR) absorption. While the first reduction steps are thus assigned to largely ligand-centered processes, the oxidation appears to involve metal-ligand delocalized molecular orbitals with variable degrees of mixing.  相似文献

The new inorganic-organic hybrid compound (dienH2)Cu2Sn2S6 (dien = diethylenetriamine) was synthesized under solvothermal conditions. It crystallizes in the tetragonal space group I4m2 with a = 7.8793(3) A, c = 24.9955(15) A, and V = 1551.80(13) A(3). The structure consists of anionic [Cu2Sn2S6](2-) layers extending in the (001) plane and protonated amine molecules as charge compensating ions sandwiched between the layers. The layered [Cu2Sn2S6](2-) anion is composed of a single layer of edge-sharing CuS4 tetrahedra which is joined above and below to straight chains constructed by corner-sharing SnS4 tetrahedra. The material is a semiconductor with an optical band gap of 1.51 eV. More interestingly, preliminary results demonstrate that the compound exhibits photoconductive properties with an increase of the conductivity by a factor of 3 when irradiated with UV light. Upon heating in an inert atmosphere the compound starts to decompose at about 256 degrees C.  相似文献

A microwave-assisted, environmentally friendly, high-yielding, time-saving synthesis of medicinally important 3-substituted isocoumarins was carried out in a single step by direct condensation of homophthalic acid with aryol and acyl chlorides under solvent-free conditions without any solid support. A new approach to the facile large-scale fabrication of robust silicon membranes with artificial proton conducting channels is presented. Ordered two-dimensional macroporous silicon was rendered proton conducting by growing a thick uniform polyelectrolyte brush using surface-initiated atom transfer radical polymerization throughout the porous matrix. The fabricated silicon-poly(sulfopropyl methacrylate) hybrid membranes were evaluated for their proton conductivity, ion exchange capacity, and water uptake. With proton conductivities in the range of 10(-2) S/cm, these proof-of-concept experiments highlight a promising alternative for producing tailorable proton conducting membranes. This approach constitutes a benchmark for the preparation and study of model systems and, in addition, for the large-scale fabrication of membranes suitable for a wide range of technological applications.  相似文献
