In this study, an original method of macromolecular design was used to develop a hyaluronidase‐1 (HYAL1) inhibitor from its principal substrate, hyaluronic acid (HA). HA‐based nanoparticles (HA‐NP) were obtained by copolymer self‐assembly and their effects on HYAL1 activity were investigated by combining different analytical tools. Compared to HA, HA‐NP exhibited an enhanced stability against HYAL1 degradation while maintaining its interaction with the HA receptors CD44 and aggrecan. HA‐NP displayed a strong and selective inhibition of HYAL1 activity and retarded the hydrolysis of higher‐molar‐mass HA in solution. A co‐nanoprecipitation process was used to formulate a range of hybrid nanoparticle samples, which demonstrated the specificity and efficiency of HA‐NP in HYAL1 inhibition.  相似文献

Click chemistry at a tetrazine core is useful for bioorthogonal labeling and crosslinking. Introduced here are two new classes of doubly clickable s-aryl tetrazines synthesized by Cu-catalyzed cross-coupling. Homocoupling of o-brominated s-aryl tetrazines leads to bis(tetrazine)s structurally characterized by tetrazine cores arranged face-to-face. [N]8 π-stacking interactions are essential to the conformation. Upon inverse electron demand Diels–Alder (iEDDA) cycloaddition, the bis(tetrazine)s produce a unique staple structure. The o-azidation of s-aryl tetrazines introduces a second proximal intermolecular clickable function that leads to double click chemistry opportunities. The stepwise introduction of fluorophores and then iEDDA cycloaddition, including bioconjugation to antibodies, was achieved on this class of tetrazines. This method extends to (thio)etherification, phosphination, trifluoromethylation and the introduction of various bioactive nitrogen-based heterocycles.  相似文献

Integral cross sections and pressure broadening coefficients have been measured for the acetylene-krypton complex, by molecular beam scattering and by high resolution IR spectroscopy, respectively. A new potential energy surface (PES) is proposed to describe structure and dynamical properties of this prototypical weakly bound complex. The PES has been parametrized exploiting a novel atom-bond pairwise additive scheme and has been fitted to the experimental data. A similar PES has been obtained for the acetylene-xenon system by a proper scaling of the interaction parameters of the krypton case, based on empirical considerations. These PESs together with that recently proposed by the same authors [J. Phys. Chem. 109, 8471 (2005)] for the acetylene-argon case have been employed for close coupling calculations of the pressure broadening cross sections and for a characterization of the rovibrational structure of the complexes.  相似文献

In arbitrary dimension, in the discrete setting of finite-differences we prove a Carleman estimate for a semi-discrete parabolic operator, in which the large parameter is connected to the mesh size. This estimate is applied for the derivation of a (relaxed) observability estimate, that yield some controlability results for semi-linear semi-discrete parabolic equations. This article presents an empirical study of 13 derivative markets for commodities and financial assets. The study goes beyond statistical analysis by including the maturity as a variable for the daily returns of futures contracts from 1998 to 2010, and for delivery dates up to 120 months. We observe that the mean and variance of the commodities follow a scaling behavior in the maturity dimension with an exponent characteristic of the Samuelson effect. The comparison between the tails of the probability distribution according to the expiration dates shows that there is a segmentation in the fat tails exponent term structure above the Lévy stable region. Finally, we compute the average tail exponent for each maturity, and we observe two regimes of extreme events for derivative markets, reminiscent of a phase diagram with a sharp transition at the 18th delivery month.  相似文献

We consider the incompressible Euler equations in a (possibly multiply connected) bounded domain Ω of R2, for flows with bounded vorticity, for which Yudovich (1963) proved in [29] global existence and uniqueness of the solution. We prove that if the boundary ∂Ω of the domain is C (respectively Gevrey of order M?1) then the trajectories of the fluid particles are C (respectively Gevrey of order M+2). Our results also cover the case of “slightly unbounded” vorticities for which Yudovich (1995) extended his analysis in [30]. Moreover if in addition the initial vorticity is Hölder continuous on a part of Ω then this Hölder regularity propagates smoothly along the flow lines. Finally we observe that if the vorticity is constant in a neighborhood of the boundary, the smoothness of the boundary is not necessary for these results to hold.  相似文献

We tackle the issue of the inviscid limit of the incompressible Navier–Stokes equations when the Navier slip-with-friction conditions are prescribed on impermeable boundaries. We justify an asymptotic expansion which involves a weak amplitude boundary layer, with the same thickness as in Prandtl’s theory and a linear behavior. This analysis holds for general regular domains, in both dimensions two and three.  相似文献

A family of water-soluble colorimetric chemosensors incorporating an anthraquinone signalling subunit functionalized with a polyamine chain that bears hydrophilic diethoxyphosphoryl moieties was prepared with the aim of assaying metal cations. The outstanding UV-Vis absorption properties of the 1-aminoanthraquinone chromophore allowed the efficient visual detection and quantification of copper(II) ions by chelators L(1)-L(3) in buffered aqueous solution. Moreover, the visible response of L(2) is not interfered by addition of large excesses of 13 common metal ions, whereas chemosensor L(3) produces also a color change in the presence of equimolar amounts of lead(II). Considering the 134 nm gap between both absorption maxima, simultaneous colorimetric quantification of lead and copper can be envisaged. Detailed potentiometric and spectrophotometric analysis of Cu(2+) complexation by L(2) and L(3), as well as Pb(2+) and Cd(2+) by L(3) was undertaken in order to gain a deeper insight into the pH-dependent speciation and understanding the color changing process. Furthermore, the inner coordination sphere of the [PbL(3)](2+) complex was probed by NMR spectroscopy.  相似文献

Organic–inorganic hybrid membranes of poly(vinylidene fluoride)–cohexafluoropropylene (PVdF–HFP) and mesostructured silica containing sulfonic acid groups were synthesized by using the sol‐gel process. These hybrid membranes were prepared by in situ co‐condensation of tetraethoxysilane and an organically modified silane (ormosil) by a self‐assembly route using organic surfactants as templates for tuning the architecture of the hybrid organosilica component. In this paper, we describe the elaboration and characterization of hybrid membranes all the way from the precursor solution to the evaluation of the fuel cell performances. These hybrid materials were extensively characterized by using NMR and IR spectroscopy, electron microscopy, or impedance spectroscopy so as to determinate their physicochemical and electrochemical properties. Even though the ion‐exchange capacity (IEC) was quite weak, the first fuel cell tests performed with these hybrid membranes show promising results relative to optimized Nafion 112 thanks to great water management of the silica inside the hydrophobic polymer.  相似文献
