Lactate oxidase from Pediococcus species can be rapidly and simply immobilized on a commercially available pre-activated membrane fixed to an amperometric probe detecting hydrogen peroxide, to provide a very sensitive practical l-lactate sensor. At 25°C, in a phosphate buffer pH 7.1, the detection limit is 1.25 × 10?7 M, calibration is linear between 2.5 × 10?7 and 2.5 × 10?4 M, the response time is <2 min, and the probe can be used for hundreds of assays over several weeks. With a microprocessor-based instrument including the same type of electrode, sample injections can be made at 90-s intervals, the response being displayed after only 30 s. High selectivity is achieved because the differential measurement system continuously subtracts currents at the chosen potential arising from the presence of electroactive species. Samples (20 μl) from sera and dairy products were successfully tested without pretreatment; a relative standard deviation between 1 and 3% was routinely obtained. The chemical investigation of the extract of the dried leaves of Rauvolfia caffra (Sond) (synonym Rauvolfia macrophylla) (Apocynaceae) led to isolation of a new glycoside derivative, rauvolfianine (1) as well as six known compounds: oleanolic acid (2), sitosterol-3-O-β-D-glucopyranoside (3), betulinic acid (4), vellosimine (5), sarpagine (6) and D-fructofuranosyl-β-(2→1)-α-D-glucopyranoside (7). Compounds 1, 2, 3, 4 and 7 were evaluated for antitubercular activity. Compounds 1 and 2 were the most active (MIC = 7.8125 and 31.25 μg/mL) towards the Isoniazid resistant strain of Mycobacterium tuberculosis AC45. The oxidation with air of cyclohexanone was conducted in the presence of synthetic carbons catalysts. The effect of carbon activation treatment (CO2 or air burnoff), phosphorus additive, platinum loading, and nature of the solvent (water or water/acetic acid mixture) were studied. Cyclohexanone oxidation at 140 degrees C yielded a mixture of C6, C5, and C4 dicarboxylic acids. Air activated carbons, including those containing phosphorus or those supporting platinum, resulted in a higher yield of adipic acid. The activity and selectivity was associated with the oxygenated functional groups, essentially carbonyl/quinone groups, created during air activation of the carbon. The incorporation of phosphorus into the carbon increased slightly the selectivity to 34.3%, probably because this additive increased the density of oxygenated functional groups. The deposition of platinum by impregnation and liquid-phase reduction with formaldehyde increased the reaction rate and improved the selectivity to adipic acid, where the highest figure was 38.8%. It was suggested that platinum contributed to molecular oxygen activation. In contrast platinum deposition by cationic exchange followed by reduction under H2 resulted often in a detrimental effect probably because the density of the oxygenated groups on carbon is decreased upon H2 reduction in the presence of platinum. When the oxidation of cyclohexanone was carried out in mixtures of water/acetic acid, the selectivity given by the different samples were quite close and generally smaller than those obtained in water which indicates that acetic acid interacts with the oxygenated surface functional groups responsible for the activity and selectivity. Oxidation experiments with methyl-labeled 4-methylcyclohexanone indicated that glutaric acid was produced by oxidative decarbonylation of both the C1 and C2 carbon atoms of the molecule. 相似文献
