Background  

Nudix hydrolases form a protein family whose function is to hydrolyse intracellular nucleotides and so regulate their levels and eliminate potentially toxic derivatives. The genome of the radioresistant bacterium Deinococcus radiodurans encodes 25 nudix hydrolases, an unexpectedly large number. These may contribute to radioresistance by removing mutagenic oxidised and otherwise damaged nucleotides. Characterisation of these hydrolases is necessary to understand the reason for their presence. Here, we report the cloning and characterisation of the DR0975 gene product, a nudix hydrolase that appears to be unique to this organism.  相似文献

The uptake of the three species of the drug model fluorescein (fluorescein sodium salt (FNa), fluorescein free acid (F), and fluorescein diacetate (FDA)) by zeolite NaX and the mesoporous zeotype MCM-41 was investigated as well as their release rates into solutions at pH 7 and pH 4.5. UV/Vis analysis was carried out at a wavelength of 490 nm. Uptakes of the sodium salt of 9 % for zeolite X and 14 % for MCM suggest little penetration of the pores. The use of ethanol as the loading solvent for F resulted in little uptake for both zeolitic materials due to the successful competition of the ethanol for binding sites. Use of acetone (weaker proton acceptor) as loading solvent significantly improved the uptake of F to 17 % and 12 % for zeolite X and MCM, respectively, whilst the uptake of FDA in acetone increased still further to 22 % and 17 % for zeolite X and MCM, respectively. Generally there was a large initial release of the fluorescein analogues from the surface of the zeolites with very little further increase over time. The prescence of an esterase enzyme in the release medium of FDA tripled the release from MCM to 15 % but left the release from zeolite X unaffected at 6 %. The results obtained show that uptake of fluorescein and its analogues is dependent on the loading solvent used, the amount released is influenced by not only the solvent but the pH and the presence of enzymes in the release medium.  相似文献

The nucleus of the carbacephem antibiotic loracarbef was synthesized in a highly efficient and enantioselective fashion from 2S,3S-2-amino-3-hydroxy-6-heptenoic acid (AHHA), which was derived from enzyme-catalyzed condensation of glycine and 4-pentenaldehyde. Retention properties of polyethylene glycol-phosphate aqueous two-phase systems in a spiral coil (5 mm I.D.) on Type-J synchronous counter-current chromatographic devices have been compared for the elution mode where the lower phase is the mobile phase and flows from the inside head terminal. This was achieved with the aid of digital imaging under stroboscopic illumination, an image analysis and measurement of the displaced volume of the stationary phase. For the spiral coil, high and stable stationary phase retention at mobile phase flow rates up to 64 ml/min has been obtained. Wave-like disturbance of the interface near the proximal point was observed and analyses have been made for possible use in protein separation.  相似文献

The reaction of [Ru 3 III 3-O)(μ-O2CCF3)6(H2O)3](O2CCF3) in methanol gives two solvates, [Ru3O(O2CCF3)6(DMSO)3] · 1/2H2O (I) and [Ru3O(O2CCF3)6(DMSO)3] · H2O (II), of a novel trinuclear mixedvalence Ru(II,III,III) trifluoroacetate complex, where two DMSO molecules are coordinated to the Ru atoms through the O atom, while the third DMSO molecule is coordinated through the S atom. According to the X-ray diffraction data, the complex can crystallize in two crystal systems: triclinic (I) (space group P \(\overline 1 \)) and monoclinic (II) (space group P21/m). The unit cell parameters for I are: a = 9.354, b = 11.005, c = 20.846 Å, α = 99.10, β = 96.38, γ = 92.17, Z = 2; R = 7.27%; for I are: a = 9.186, b = 17.044, c = 13.091 Å, β = 101.10, Z = 2; R = 14.18%.  相似文献

As part of a study initiating the development of an analytically validated Dietary Supplement Ingredient Database (DSID) in the United States (US), a selection of dietary supplement products were analyzed for their caffeine content. Products sold as tablets, caplets, or capsules and listing at least one caffeine-containing ingredient (including botanicals such as guarana, yerba mate, kola nut, and green tea extract) on the label were selected for analysis based on market share information. Two or three lots of each product were purchased and analyzed using high-pressure liquid chromatography (HPLC). Each analytical run included one or two National Institute of Standards and Technology (NIST) Standard Reference Materials (SRMs) and two products in duplicate. Caffeine intake per serving and per day was calculated using the maximum recommendations on each product label. Laboratory analysis for 53 products showed product means ranging from 1 to 829 mg caffeine/day. For products with a label amount for comparison (n = 28), 89% (n = 25) of the products had analytically based caffeine levels/day of between −16% and +16% of the claimed levels. Lot-to-lot variability (n = 2 or 3) for caffeine in most products (72%) was less than 10%.  相似文献

The dependence of the magnetization relaxation rate S = ?d lnM/dlnt on temperature T is measured in YBa2Cu3O7 ? δ samples with various oxygen concentrations. It is found that the S(T) curve changes qualitatively when oxygen deficit δ exceeds the threshold value δth = 0.37. For δ < δth (T c > 60 K, where T c is the superconducting transition temperature), function S(T) has the well-known peak at T/T c = 0.4. For δ > δth (at T c < 51 K), this peak transforms into a plateau and a new sharp peak appears at T/T c = 0.1. The threshold value δth of the oxygen deficit corresponds to the transition of the sample from the disordered state into the ordered state of oxygen vacancies. We consider the change in the shape of the S(T) curve as a macroscopic manifestation of this transition.  相似文献

The evolution of the spatial distribution of the magnetic induction in a superconductor after the action of the alternating magnetic field perpendicular to the trapped magnetic flux has been analyzed. The observed stabilization of the magnetic induction profile is attributed to the increase in the pinning force, so that the screening current density becomes subcritical. We report on a thorough optical investigation of BaFe2As2 over a broad spectral range and as a function of temperature, focusing our attention on its spin-density-wave (SDW) phase transition at TSDW = 135 K. While BaFe2As2 remains metallic at all temperatures, we observe a depletion in the far infrared energy interval of the optical conductivity below TSDW, ascribed to the formation of a pseudogap-like feature in the excitation spectrum. This is accompanied by the narrowing of the Drude term consistent with the dc transport results and suggestive of suppression of scattering channels in the SDW state. About 20% of the spectral weight in the far infrared energy interval is affected by the SDW phase transition.  相似文献
