Mathematical models based on chemometric analyses of the coffee beverage sensory data and NIR spectra of 51 Arabica roasted coffee samples were generated aiming to predict the scores of acidity, bitterness, flavour, cleanliness, body and overall quality of coffee beverage. Partial least squares (PLS) were used to construct the models. The ordered predictor selection (OPS) algorithm was applied to select the wavelengths for the regression model of each sensory attribute in order to take only significant regions into account. The regions of the spectrum defined as important for sensory quality were closely related to the NIR spectra of pure caffeine, trigonelline, 5-caffeoylquinic acid, cellulose, coffee lipids, sucrose and casein. The NIR analyses sustained that the relationship between the sensory characteristics of the beverage and the chemical composition of the roasted grain were as listed below: 1 - the lipids and proteins were closely related to the attribute body; 2 - the caffeine and chlorogenic acids were related to bitterness; 3 - the chlorogenic acids were related to acidity and flavour; 4 - the cleanliness and overall quality were related to caffeine, trigonelline, chlorogenic acid, polysaccharides, sucrose and protein. 相似文献

Immobilized metal ion affinity chromatography (IMAC) has been widely used for the enrichment of phosphopeptides, whereas no report exists describing the use of IMAC columns for the enrichment of sulfopeptides. In this study, we used IMAC-Ga microcolumns for the enrichment of sulfopeptides from a complex mixture of peptides, extracted from skin secretions of the Pachymedusa dacnicolor frog. The enriched fraction obtained by IMAC-Ga was analyzed by liquid chromatograpy/electrospray ionization mass spectrometry (LC/ESI-MS) in an Orbitrap XL and by matrix-assisted laser desorption/ionization time-of-flight time-of-flight (MALDI-TOF/TOF) in an ABI 4800 instrument. From this fraction, different sulfated and non-sulfated peptides belonging to the caerulin and bradykinin families were structurally characterized. Other interesting negatively charged groups, such as phosphate adducts of dermaseptins and pyridoxal phosphate attached to a protease inhibitor, were also characterized. Unexpectedly, some dermaseptin antimicrobial peptides were also enriched by IMAC-Ga and a Sauvatine-like peptide was also fully sequenced. Furthermore, neutral loss of sulfated peptides and their fragmentation patterns in the gas phase were also compared using collision-induced dissociation (CID) and high-energy collision dissociation (HCD). Our present study provides evidence that IMAC-Ga enrichment is a fast, useful and promising method for high-throughput analysis of sulfated-peptides, since high-resolution mass spectrometers can be used for this purpose. 相似文献

The development of new methods and concepts to visualize massive amounts of data holds the promise to revolutionize the way scientific results are analyzed, especially when tasks such as classification and clustering are involved, as in the case of sensing and biosensing. In this paper we employ a suite of software tools, referred to as PEx-Sensors, through which projection techniques are used to analyze electrical impedance spectroscopy data in electronic tongues and related sensors. The possibility of treating high dimension datasets with PEx-Sensors is advantageous because the whole impedance vs. frequency curves obtained with various sensing units and for a variety of samples can be analyzed at once. It will be shown that non-linear projection techniques such as Sammon's Mapping or IDMAP provide higher distinction ability than linear methods for sensor arrays containing units capable of molecular recognition, apparently because these techniques are able to capture the cooperative response owing to specific interactions between the sensing unit material and the analyte. In addition to allowing for a higher sensitivity and selectivity, the use of PEx-Sensors permits the identification of the major contributors for the distinguishing ability of sensing units and of the optimized frequency range. The latter will be illustrated with sensing units made with layer-by-layer (LbL) films to detect phytic acid, whose capacitance data were visualized with Parallel Coordinates. Significantly, the implementation of PEx-Sensors was conceived so as to handle any type of sensor based on any type of principle of detection, representing therefore a generic platform for treating large amounts of data for sensors and biosensors. 相似文献

In this study the phase equilibrium behaviors of the binary system (CO2 + lauric acid) and the ternary system (CO2 + methanol + lauric acid) were determined. The static synthetic method, using a variable-volume view cell, was employed to obtain the experimental data in the temperature range of (293 to 343) K and pressures up to 24 MPa. The mole fractions of carbon dioxide were varied according to the systems as follows: (0.7524 to 0.9955) for the binary system (CO2 + lauric acid); (0.4616 to 0.9895) for the ternary system (CO2 + methanol + lauric acid) with a methanol to lauric acid molar ratio of (2:1); and (0.3414 to 0.9182) for the system (CO2 + methanol + lauric acid) with a methanol to lauric acid molar ratio of (6:1). For these systems (vapor + liquid), (liquid + liquid), (vapor + liquid + liquid), and (solid + fluid) transitions were observed. The phase equilibrium data obtained for the systems were modeled using the Peng–Robinson equation of state with the classical van der Waals mixing rule with a satisfactory correlation between experimental and calculated values. 相似文献

This paper presents the validation of a system for sample digestion using a digester block/cold finger to determine the lead content in vegetables by electrothermal atomic absorption spectrometry (ETAAS). After mineralization, lead contents were determined by ETAAS using a calibration curve based on aqueous standards prepared in 2.60 M nitric acid solutions containing 5 microg ammonium phosphate as chemical modifier. A pyrolysis temperature of 900 degrees C and atomization temperature of 2000 degrees C were used. This method allowed the determination of lead with a characteristic mass of 35 pg; LOD and LOQ of 0.6 and 2 nglg, respectively, were found. The precision was investigated in terms of reproducibility and repeatability. Reproducibility was estimated by analysis of nine different portions of a certified reference material (CRM) of spinach leaves, and the repeatability was determined through the analysis of nine aliquots of the same solution. The reproducibility and repeatability were found to be 4.27 and 2.94% RSD, respectively. The accuracy was confirmed by analysis of whole meal flour, spinach leaves, and orchard leaves CRMs, all furnished by the National Institute of Standards and Technology. Lead contents were measured using the newly developed technique in 11 different potato samples. The lead contents ranged from 12.80 to 69.27 ng/g, with an average value of 28.59 ng/g. The fluorescence properties of two bis(1,8-naphthalimides) with amino-containing spacers are investigated, giving special emphasis to the observation of excimer emission. It is found that a minor elongation of the spacer by two methylene units gives rise to a quantitative shut-down of the broad and red-shifted excimer emission. Furthermore, a switching of this emission is established through manipulation of a photoinduced electron transfer process, which involves the amino spacer. Protons as well as protic solvents lead to substantial excimer emission with lifetimes of 12 to 27 ns. The excimer quantum yield takes a maximum value of Φ(f) = 0.07 (acetonitrile with 1 equiv trifluoroacetic acid). The increased virtual Stokes shifts (ca. 150 nm) as compared to the fluorescence of monomeric 1,8-naphthalimides are an alternative approach to obtain colored, significant, and long-lived fluorescence from these chromophores. As an additional excited state pathway, the occurrence of homo-Fo?rster resonance energy transfer (homo-FRET) is established by fluorescence polarization measurements and calculation of the corresponding critical Fo?rster radius (R(0) ca. 13 ?). The average interchromophore distance between the naphthalimides is estimated as 7.5 ? and 9.5 ? for the dyad with the shorter and the longer spacer, respectively. These observations and the absence of a rise time component for excimer emission are in agreement with the formation of a "loose" ground state dimer, which upon excitation undergoes a fast geometrical adjustment to the excimer structure where the chromophores are at contact distance. 相似文献

Pequi (Caryocar brasiliense Camb.), baba?u (Orbignya phalerata Mart.), buriti (Mauritia flexuosa), and passion fruit (Passiflora edulis) oils were studied to determine their antibacterial, antioxidant and cytotoxic activities, as well as their total phenol and carotenoid contents. The fatty acid contents were determined by GC-MS. The three types of passion fruit oils studied were refined, cold pressed or extracted from seeds in a Soxhlet apparatus. The oils thus obtained showed differences in antioxidant activity and carotenoid content, but were similar in regard to total phenols. Buriti and pequi had the highest carotenoid contents, while refined and cold pressed passion fruit oil displayed the highest antioxidant activity. Pequi oil was the only oil to display antibacterial and cytotoxic activity. 相似文献
