An extracellular, endo-??-1,4-xylanase was purified to homogeneity from the culture filtrate of the filamentous fungus Penicillium occitanis Pol6, grown on oat spelt xylan. The purified enzyme (PoXyn2) showed a single band on SDS?CPAGE with an apparent molecular weight of 30?kDa. The xylanase activity was optimal at pH?3.0 and 65?°C. The specific activity measured for oat spelt xylan was 2,368?U?mg?1. The apparent K m and V max values were 8.33?mg?ml?1 and 58.82???mol?min?1?ml?1, respectively, as measured on oat spelt xylan. Thin-layer chromatography experiments revealed that purified PoXyn2 degrades xylan in an endo-fashion releasing xylobiose as main end product. The genomic DNA and cDNA encoding this protein were cloned and sequenced. This PoXyn2 presents an open reading frame of 962?bp, not interrupted by any introns and encoding for a mature protein of 320 amino acids and 29.88?kDa.  相似文献

Resolution and penetration are primary criteria for image quality of diagnostic ultrasound. In theory (and usually in practice), the maximum depth of imaging in a tissue increases as power (pressure) is increased. Alternatively, at a particular effective penetration, an increased power may be used to allow a higher ultrasound frequency for higher resolution and tissue contrast. Recently, Karagoz and Kartal [29] proposed a safety parameter for thermal bioeffects of diagnostic ultrasound; that is, SUT (safe use time). The SUT model is constructed to determine how long one piece of tissue can be insonated safely according to a threshold exposure. Also, Karagoz and Kartal [29] suggested that an increase in acoustic intensity beyond the current US Food and Drug Administration (FDA) limit of intensity can be theoretically possible by using SUT model while staying within the safe limit. The present study was motivated particularly by the goals of higher resolution and/or deeper penetration by using SUT model. The results presented here suggest that the safe use of higher exposure levels than currently allowed by the FDA may be possible for obtaining substantial improvements in penetration depth and/or resolution. Also, the study reveals that image quality can be functionally related to exposure time in addition to acoustic energy and frequency.  相似文献

New series of 1H-perimidine-2-thiol derivatives and (2-substituted-1H-perimidin-1-yl)ethane-1,2-dione derivatives and their ligands (C24H14N4S2O2) H2L1 and (C26H18N4S2O2) H2L2 have been synthesized with transition metal ions, e.g., Copper (II), Silver (I), Cobalt (II) and Ruthenium (III) were prepared and evaluated for their antimicrobial, analgesic and anti-inflammatory activities. The synthesized compounds and their complexes were characterized by elemental analysis, 1H NMR, IR, MS, molar conductance, thermal gravimetric analysis and electronic spectra. All results revealed that compounds 3 and 13 exhibited high inhibitory effects against some bacterial strains by the disc diffusion method. On the other hand, compounds 2, 3, 7 and 12 displayed potent anti-inflammatory activity.  相似文献

Alfa grass pulping was successfully performed in hydro-organic acid medium under mild conditions (107°C, atmospheric pressure, cooking time: 3 h). Use of an acetic acid/formic acid/water mixture as pulping liquor was perfectly suitable for selective isolation of pulp, lignin, and hemicelluloses. The unbleached pulp obtained in good yield was first delignified by peroxyacids in organic acid medium and then bleached with hydrogen peroxide in a basic medium to give pulp offering good physico-chemical and mechanical characteristics.  相似文献

A new methoxy functionalized 2‐(trichloromethyl)‐1,3‐diarylimidazolidin (6) was synthesized as the precursor for N‐heterocyclic carbene complexes of Pd(II) and Rh(I) by the condensation of N,N'‐bis(2,4‐dimethoxyphenyl)‐1,2‐diaminoethane with chloral. The structures of all compounds have been elucidated by a combination of multinuclear NMR spectroscopy, elemental analysis and in one instance, by single crystal X‐ray diffraction. Compound 8, C27H34N2O4ClRh, crystallizes in the triclinic space group P‐1 with cell dimensions a = 9.7642(12)Å, b = 11.1914(11)Å, c = 13.0102(14)Å, α = 104.034(9)°, β = 106.658(9)°, γ = 99.658(9)° with Z = 2. The molecular structure of 8 shows the geometry around the Rh metal to be a slightly distorted square planar. A fluorogenic probe based on a coumarin-derivative for Cu2+ sensing in CH3CN/H2O media (v/v, 95/5, 5.0 μM) was developed and applied in real samples. 3-(4-chlorophenyl)-6,7-dihydroxy-coumarin (MCPC) probe was obtained by synthetic methodologies and identified by spectral techniques. The probe MCPC showed remarkable changes with a “turn-off” fluorogenic sensing approach for the monitoring of Cu2+ at 456 nm under an excitation wavelength of 366 nm. The response time of the probe MCPC was founded as only 1 min. The detection limit of the probe MCPC was recorded to be 1.47 nM. The binding constant and possible stoichiometric ratio (1:1) values were determined by Benesi-Hildebrand and Job’s plot systems, respectively. The mechanism of the probe MCPC with Cu2+ was further confirmed by ESI-MS and FT-IR analyses, as well as supported by theoretical calculations. Furthermore, the probe MCPC was successfully employed for the practical applications to sense Cu2+ in different herbal and black tea samples. The proposed sensing method was also verified by ICP-OES method.  相似文献

A series of new N‐substituted isoquinolin‐1,3‐dione derivatives were prepared, starting from reaction of (Z)‐4‐((E)‐3‐phenylallylidene)isochromane‐1,3‐dione 4 with different nitrogen nucleophiles. The assigned structures of the prepared compounds were elucidated by spectral methods (IR, 1H NMR, 13C NMR, and mass spectroscopy). Some of the newly prepared compounds were tested in vitro against a panel of three human tumor cell lines, namely, hepatocellular carcinoma (liver) HepG2, colon cancer HCT‐116, and mammary gland breast MCF‐7. Also, they were tested as antioxidants. Some of the tested compounds showed very strong cytotoxic activity with respect to the standard.  相似文献

In this paper, we study the relation between the fuzzy separation axioms, which had been introduced by the authors in 2001, and the fuzzy proximity defined by Katsaras in 1980. We study also the relation between our fuzzy separation axioms and the G-compactness defined by Gähler in 1995. Moreover, we show here the relation between these fuzzy separation axioms and the fuzzy uniform structures introduced and studied by Gähler and the first author in 1998.  相似文献
