The paper describes a sensitive method for simultaneous sensing of morphine (MOR) and diclofenac (DCF). The surface of a MgFe2O4/graphite paste electrode was modified with multi-walled carbon nanotubes, and the resulting sensor was characterized by cyclic voltammetry, differential pulse voltammetry, chronoamperometry, and electrochemical impedance spectroscopy. The electrode showed an efficient synergistic effect in term of oxidation of DCF and MOR, with sharp oxidation peaks occurring at +0.370 and 0.540 V (vs Ag/AgCl) at pH 7.0. The calibration plot for MOR is linear in the 50 nM to 920 μM concentration range, and the detection limit is 10 nM (at a signal-to-noise ratio of 3). The respective data for DCF are 100 nM to 580 μM, with a 60 nM LOD. The sensor was applied to the determination of MOR and DCF in spiked serum and urine samples, with recoveries ranging between 91.4 and 100.7 %.

Protein aggregation is commonly associated with a large number of neurodegenerative disorders such as Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, and other types of pathological conditions. Misfolding and aggregation of a number of peptides and proteins have been found to occur under these conditions. In the present review, some mechanistic features of the events related to the type of structure–function relationships which may define the outcome of the abnormal conditions are discussed. The immunological responses to the aggregates and possible therapeutic strategies for prevention or control of the diseases are also reviewed. Protein aggregation and its effect on human body have become an important issue over the last two decades. Many diseases in human are related to aggregation and misfolding of different kinds of proteins; therefore, diagnosis of causes of the aggregation and their mechanisms which provoke it are important. This review describes the relations between structures and functions of already aggregated proteins, as well as proteins, which only enter initial stages of aggregation. The consequences of aggregations, which provoke many kinds of neurodegenerative disorders, are explained in details and some factors that may influence their severity are described. In addition, the immunologic responses to these aggregates are discussed. Suggestions of plausible therapies of preventing or slowing down the protein condensation diseases are presented.  相似文献

Using stem cells to replace the lost beta cells is a hopeful strategy in the treatment of diabetic patients. Furthermore, during stem cell culture and therapy, it is a need to use a substrate to act as a supportive matrix to mimic 3D in vivo microenvironment. Therefore, in this study, human adipose‐derived stem cells were used to differentiate into insulin‐producing cells (IPCs) on a silk/polyethersulfone (PES) scaffold. After exposing to the differentiation media, 2D and 3D (silk/PES) cultured cells were gradually aggregated and formed spherical shaped clusters. The viability of cells was comparable in both 3D and 2D culture. As the results of gene expression assay in both RNA and protein level showed, the differentiation efficiency was higher in 3D culture. Furthermore, ELISA revealed that the release of C‐peptide and insulin was higher in 3D than 2D culture. It seems that silk/PES nanofibrous hybrid scaffold could provide an appropriate matrix to mimic in vivo microenvironment and therefore increases the IPC differentiation potency of stem cells.  相似文献

Breast cancer radioimmunoscintigraphy targeting HER2/neu expression is a growing field of work in nuclear medicine research. Trastuzumab is a monoclonal antibody that binds with high affinity to HER2/neu, which is over expressed on breast and other tumors. Developing new tracers for the detection of this cancer is of great interest. In this study, trastuzumab was successively labeled with [64Cu]CuCl2 after conjugation with DOTA-NHS-ester. The conjugate was purified by molecular filtration, the average number of DOTA conjugated per mAb was calculated and total concentration was determined by spectrophotometric method. DOTA–trastuzumab was labeled with 64Cu produced by 68Zn(p,αn)64Cu nuclear reaction (30 MeV protons at 180 μA). Radiochemical purity, integrity of protein after radiolabeling and immunoreactivity of radiolabeled mAb trastuzumab with HER2/neu antigen and SkBr3 cell line were performed by RIA. In vitro stability of radiolabeled mAb in human serum was determined by thin layer chromatography. In vitro internalization studies were performed with the SkBr3 cell line and the tissue biodistribution of the 64Cu–DOTA–trastuzumab was evaluated in wild-type rat (90 ± 5.5 μCi, 2, 6, 12, 24 h p.i.). The radioimmunoconjugate was prepared with a radiochemical purity of higher than 96 ± 0.5 % (ITLC) and specific activity as high as 5.3 μCi/μg. The average number of chelators per antibody for the conjugate used in this study was 5.8/1. The sample was showed to have similar patterns of migration in the gel electrophoresis. The 64Cu–DOTA–trastuzumab showed high immunoreactivity towards HER2/neu antigen and SkBr3 cell line. In vitro stability of the labeled product was found to be more than 94 % in PBS and 82 ± 0.5 % in human serum over 48 h. In vitro internalization studies of the 64Cu–DOTA–trastuzumab showed that up to 11.5 % of the radioimmunoconjugate internalized after 10 h. The accumulation of the radiolabeled mAb in liver, skin, intestine, lung, spleen, kidney and other tissues demonstrates a similar pattern to the other radiolabeled anti-HER2 immunoconjugates. 64Cu–DOTA–trastuzumab is a potential compound for molecular imaging of PET for diagnosis and treatment studies and follow-up of HER2 expression in oncology.  相似文献

Gold nanoparticles (AuNPs) were electrodeposited on the surface of pencil graphite (PG) by fast scan cyclic voltammetry without using any additives in acidic medium. The effect of deposition time on the size of electrodeposited AuNPs was investigated in sulfuric acid as a supporting electrolyte. The deposition time was varied by varying the scan rate, number of cycles and applied potential range. Scanning electron microscopy (SEM), transmission electron microscopy (TEM) and X‐ray diffraction (XRD) were used for characterization of PG and electrodeposited AuNPs. The results confirmed that nanosized gold particles (20 ± 8 nm) were deposited on the PG substrate with almost spherical geometry.  相似文献

The use of thin-film solid-phase microextraction (SPME) as the sampling preparation step before direct analysis in real time (DART) was evaluated for the determination of two prohibited doping substances, cocaine and methadone, in urine samples. Results showed that thin-film SPME improves the detectability of these compounds: signal-to-blank ratios of 5 (cocaine) and 13 (methadone) were obtained in the analysis of 0.5 ng/ml in human urine. Thin-film SPME also provides efficient sample cleanup, avoiding contamination of the ion source by salt residues from the urine samples. Extraction time was established in 10 min, thus providing relatively short analysis time and high throughput when combined with a 96-well shaker and coupled with DART technique.

A new method is presented for simultaneous preconcentration of trace Fe(III) and Cr(III) by using polyacrylic acid-alumina as a sorbent. The separation/preconcentration conditions of analytes were investigated, including effect of pH, flow rate, elution conditions, sample volume, and interfering ions. At pH 4, the maximum sorption capacities of Fe3+ and Cr3+ were 8.0 and 13.0 mg/g, respectively, by the column method. The linearity was maintained in the concentration range of 0.175-6.0 x 10(3) ng/mL for iron and 0.175-8.0 x 10(3) ng/mL for chromium in the original solution. The RSD values under optimum conditions were +/- 1.73 and +/- 1.28% for 2.0 microg/mL Fe and Cr, respectively. The preconcentration factor was 400 for both of the elements, and detection limits were 0.025 and 0.023 ng/mL for Fe and Cr in the original solutions. The proposed method was successfully applied to the determination of trace amounts of Fe and Cr in plant samples.  相似文献

This paper deals with the phase behaviour of model abnormal emulsions of cyclohexane/water/polyethoxylated surfactant in the vicinity of the locus of transitional inversion (optimum formulation). Abnormal emulsions are formed under dynamic conditions if the phase containing the soluble surfactant becomes the dispersed phase. Phase maps have been suggested in the literature that define the boundaries of emulsion morphologies. On these maps, only one transformation can be observed for any formulation scan. Furthermore, the morphology of emulsions in the vicinity of the locus of transitional inversion is rather vague. One might assume that by changing the HLB of the surfactant used in an abnormal emulsion to favour the continuous phase, that abnormal emulsion will gradually transform to a normal emulsion of the same type. A new experimental procedure was adopted in which emulsification was started with abnormal emulsions. To transform the abnormal emulsions to the normal emulsion of the same type, they were exposed to variations in temperature, the surfactant HLB, or water–oil phase ratio. As the optimum formulation was approached, the abnormal emulsions became so unstable that could not exist anymore in the un-favoured morphology and inverted to the normal emulsion of the opposite type. Further variation in the formulation along the route led to a transitional inversion to the normal emulsion of the original type. The result indicates an important finding that the transformation of abnormal emulsions to normal ones of the same type occurred via two successive inversions of catastrophic and transitional nature. It appears that the boundaries of catastrophic inversion correspond to the emulsions with the finest drop size. A modification to the phase behaviour maps, to include the locus of catastrophic phase inversion in the vicinity of the transitional inversion, was thus suggested.  相似文献

In this work, a water-soluble polymer, polyethylenimine (PEI) was used for the simultaneous separation and preconcentration of trace Cu and Mn prior to their determination by flame atomic absorption spectrometry. For this purpose, the sample and the PEI solution were mixed and the metal-bound polymer was precipitated by adding acetone. The precipitate was separated and dissolved in a minimum amounts of water and aspirated into a flame AAS. By increasing the ratio of the volumes of sample to water used in dissolving the precipitate, the analyte elements were concentrated as needed. The sorption is quantitative in the pH ≥6. Detection limits were 5.2 μg/L for Cu and 5.4 μg/L for Mn. This method is simple, fast and precise.  相似文献
