A ten-residue fragment of an antibody (mini-antibody) directed against lysozyme as ligand in immunoaffinity chromatography

The interaction between an antibody molecule and a protein antigen is an example of "natural" protein modelling. Amino acids of the antigen-binding site consisting of three hypervariable segments (L1, L2, L3) of the light (L) and three (H1, H2, H3) of the heavy (H) chain of an antibody molecule interact with amino acids present in an epitope of a protein. A ten-residue peptide was synthesized with an amino acid sequence analogous to the hypervariable L3 segment of a monoclonal antibody directed against lysozyme. The peptide was immobilized on CH-Sepharose 4B and the affinity adsorbent was used to purify lysozyme added to a detergent extract of insect cells infected with a recombinant baculovirus. This methodology may also be applicable to other antigen-antibody combinations, in immunoaffinity chromatography for selective purification of a protein or in an immunosensor for detection of a protein.     

Azo coupling and aminomethylation of 2,5-diphenylpyrrole and its derivatives

The azo coupling of 2,5-diphenylpyrrole with arenediazonium chlorides has given previously unknown 3-arylazopyrroles and 4-phenylazo-3-phenylhydrazono-3H-pyrrole. The methylation and reductive acetylation of the anylazo derivatives have led to N-methylarylazo and acetylamino derivatives of 2,5-diphenylpyrrole. 3-Amino-2, 5-diphenylpyrrole has been obtained by the reduction of 3-p-chlorophenylazo-2,5-diphenylpyrrole. Dependence of the catalytic activity of organoaluminum compounds and some of their complexes on their composition

The influence of the acidity of organoaluminum compounds and their complexes on the catalytic activity in polymerization of trioxane has been studied. It is found that the catalytic activity of organoaluminum compounds of the type R_nALX_3?n depends on n as well as the nature of R or X. Catalytic activity of the complexes also depends on the type of cocatalyst used. The change of electroconductivity in the course of formation of some initiator complexes has been studied and an attempt has been made to determine the relation between conductivity and polymerization activity.     

Research on the aromas. On the aroma of coffee. I

The analysis of a coffee concentrate is described: 202 constituents have been identified, 154 of which for the first time. Some of these compounds are new chemicals; their synthesis is described. Most identifications are based upon mass spectrometry combined in many cases with IR. spectrometry and comparison with reference compounds. MS. and IR. data are given in detail. The analysis shows that a large number of minor constituents is present, many of which could not be identified.     

Über den Nachweis und die Bestimmung des Schädlingsbekämpfungsmittels E 605 in der gerichtlichen Chemie

Zusammenfassung Es wird gezeigt, daß sich Insektizide auf der Basis der Ester der Thiophosphorsäure (E 605) aus biologischem Material mittels Wasser-dampfdestillation auf verhältnismäßig einfache Weise abscheiden lassen. Im Destillat erfolgt die Bestimmung nach Ausäthern und Lösen des Rückstandes in Äthanol durch Messen der Lichtabsorption im UV. Hierbei zeigt sich, daß die rechnerische Eliminierung der Störstoffe bei geringeren Konzentrationen der Ester leichter ist als bei hohen.Durch Messung der Lichtabsorption im IR läßt sich, genügende Konzentration vorausgesetzt, entscheiden, welcher Ester vorliegt.

Summary It was shown that insecticides based on the ester of thiophosphoric acid (E 605) can be separated in relatively simple fashion from biological material by means of steam distillation. The distillate is shaken out with ether and the residue dissolved in ethanol and the determination is accomplished by measuring the absorption in ultraviolet light. It was found that the computational elimination of the interfering materials is easier at lower concentrations of the ester than when higher concentrations are present. By measuring the light absorption in infrared, it is possible to decide which ester is present, provided the concentration is high enough.

Résumé On montre que les insecticides se séparent d'une manière relativement simple de l'élément biologique par un entraînement à la vapeur d'eau sur la base de l'ester de l'acide thiophosphorique (E 605). Le dosage s'effectue sur le distillat d'après l'épuisement à l'éther et la solubilité du résidu dans l'éthanol et la mesure de l'absorption lumineuse dans l'ultraviolet. On en déduit que l'élimination calculée de la substance gênante est plus facile pour les faibles concentrations de l'ester que pour les grandes. En mesurant l'absorption de la lumière dans l'infrarouge, on peut savoir de quel ester il s'agit pour des concentrations supposées suffisantes.

     

Total synthesis of himandravine

[reaction: see text] The first total synthesis of (+)-himandravine (1) is described, starting from (2S,6S)-cis-2-formyl-6-methyl-N-Boc-piperidine (8) in 11 linear steps and 17% overall yield. The key step involves a highly diastereoselective intramolecular Diels-Alder reaction of the key intermediate 5 that contains the entire latent carbon framework and functional group substitution of himandravine.     

Transmembrane ion conductance by an acyclic bolaamphiphile

[reaction in text] An acyclic bolaamphiphile forms ion channels in vesicles and planar bilayer membranes that are closely similar to the channels formed by related bismacrocyclic compounds. Thus the function of these ion channels does not depend on synthetically difficult macrocyclic subunits.     

The Effects of Beating,Web Forming and Sizing on the Surface Energy of <Emphasis Type="Italic">Eucalyptus globulus</Emphasis> Kraft Fibres Evaluated by Inverse Gas Chromatography

In this work attention has been focused on the effects of papermaking beating, web forming and sizing operations on the physical/chemical surface properties of bleached Eucalyptus globulus kraft fibres. Inverse gas chromatography (IGC) was used to determine the dispersive component of surface tension (γ_s^d) as well as the acidic/basic character (according to the Lewis concept) of the solid surfaces (pulp fibres and pulp handsheets). The results have shown that the main effect of beating is to increase the fibre's Lewis acidic character. Web forming caused a strong decrease in γ_s^d and significant increments in the adhesion works of both acidic and basic probes, lowering the ratio between the two. Nevertheless, the surface of handsheets still exhibited a dominant acidic character. The sizing operation did not change the dispersive component of the surface tension significantly but decreased the difference between the adhesion works of the acidic and basic probes, rendering the handsheet surface less Lewis acidic and more Lewis basic. Thus, although internal sizing is expected to strongly influence liquid spreading at the paper surface and liquid penetration of the fibre's network, it is concluded that beating and web forming lead to important changes in the surface energetic properties of the Eucalyptus globulus kraft fibres.     

Physico-chemical aspects of polyethylene processing in an open mixer. Part 21: Improved product yields on true bimolecular hydroperoxide decomposition

The product yields from the reaction between two hydroperoxide groups have been re-calculated. This is a consequence of the fact that β-scission of secondary alkoxy radicals cannot be neglected in the high temperature range of the polyethylene processing experiments (170-200 °C). It must be taken into account in addition to disproportionation/combination and hydrogen abstraction by alkoxy radicals. The increased complexity caused by the additional reaction results mainly from the larger number of caged radical pairs involved in the reactions and also in the calculations. Among other products it becomes possible to calculate the yields of aldehyde and vinyl groups that would not result from hydroperoxide decomposition in the absence of β-scission. The yields of the main oxidation products such as alcohols, ketones and trans-vinylene groups are reduced to some extent in comparison with the values calculated if β-scission is neglected. The vinyl group yield corresponds to slightly more than 10% of the yield of trans-vinylene groups in the temperature range of the experiments. The aldehyde yield is significantly larger than the vinyl group yield and is important in the whole temperature range examined. Main-chain scissions are important at the temperatures of the experiments. They become more important than the sum of the different combination reactions from a temperature of 200 °C on.
