Background

The aim of this study is to examine the influence of the catechol-O-methyltranferase (COMT) gene (polymorphism Val158 Met) as a risk factor for Alzheimer's disease (AD) and mild cognitive impairment of amnesic type (MCI), and its synergistic effect with the apolipoprotein E gene (APOE). A total of 223 MCI patients, 345 AD and 253 healthy controls were analyzed. Clinical criteria and neuropsychological tests were used to establish diagnostic groups. The DNA Bank of the University of the Basque Country (UPV-EHU) (Spain) determined COMT Val158 Met and APOE genotypes using real time polymerase chain reaction (rtPCR) and polymerase chain reaction (PCR), and restriction fragment length polymorphism (RFLPs), respectively. Multinomial logistic regression models were used to determine the risk of AD and MCI.

Results

Neither COMT alleles nor genotypes were independent risk factors for AD or MCI. The high activity genotypes (GG and AG) showed a synergistic effect with APOE ε4 allele, increasing the risk of AD (OR = 5.96, 95%CI 2.74-12.94, p < 0.001 and OR = 6.71, 95%CI 3.36-13.41, p < 0.001 respectivily). In AD patients this effect was greater in women. In MCI patients such as synergistic effect was only found between AG and APOE ε4 allele (OR = 3.21 95%CI 1.56-6.63, p = 0.02) and was greater in men (OR = 5.88 95%CI 1.69-20.42, p < 0.01).

Conclusion

COMT (Val158 Met) polymorphism is not an independent risk factor for AD or MCI, but shows a synergistic effect with APOE ε4 allele that proves greater in women with AD.  相似文献

We report low temperature scanning tunneling microscopy and spectroscopy on graphene flakes supported on a graphite substrate. The experiments demonstrate that graphite is exceptionally well suited as a substrate for graphene because it offers support without disturbing the intrinsic properties of the charge carriers. The degree of coupling of a graphene flake to the substrate was recognized and characterized from the appearance of an anomalous Landau level sequence in the presence of a perpendicular magnetic field. By following the evolution of the Landau level spectra along the surface, we identified graphene flakes that are decoupled or very weakly coupled to the substrate. From the Landau level sequence in this flake, we extract the local Fermi velocity and energy of the Dirac point and find extremely weak spatial variation of these quantities confirming the high quality and non invasive nature of the graphite substrate.  相似文献

Electron tomography is a well‐known technique providing a 3D characterization of the morphology and chemical composition of nanoparticles. However, several reasons hamper the acquisition of tilt series with a large number of projection images, which deteriorate the quality of the 3D reconstruction. Here, an inpainting method that is based on sinogram interpolation is proposed, which enables one to reduce artifacts in the reconstruction related to a limited tilt series of projection images. The advantages of the approach will be demonstrated for the 3D characterization of nanoparticles using phantoms and several case studies.  相似文献

Magnetic nanoparticles have found broad applications in medicine, especially for cell targeting and transport, and as contrast agents in MRI. Our samples of ??-Fe2O3 nanoparticles were prepared by annealing in silica matrix, which was leached off and the bare particles were then coated with amorphous silica layers of various thicknesses. The distribution of particle sizes was determined from the TEM pictures giving the average size ~20 nm and the thickness of silica coating ~5; 8; 12; 19 nm. The particles were further characterized by the XRPD and DC magnetic measurements. The nanoparticles consisted mainly of ??-Fe2O3 with admixtures of ~1 % of the α phase and less than 1 % of the γ phase. The hysteresis loops displayed coercivities of ~2 T at room temperature. The parameters of hyperfine interactions were derived from transmission Mössbauer spectra. Observed differences of hyperfine fields for nanoparticles in the matrix and the bare ones are ascribed to strains produced during cooling of the composite. This interpretation is supported by slight changes of their lattice parameters and increase of the elementary cell volume deduced from XRD. The temperature dependence of the magnetization indicated a two-step magnetic transition of the ??-Fe2O3 nanoparticles spread between ~85 K and ~150 K, which is slightly modified by remanent tensile stresses in the case of nanoparticles in the matrix. The subsequent coating of the bare particles by silica produced no further change in hyperfine parameters, which indicates that this procedure does not modify magnetic properties of nanoparticles.  相似文献

Polypropylene (PP)/polyester (PES)–blend fibers were prepared by extruder melt spinning. The polymer blend consisted of PP and a “master batch” (MB) based on polytrimethylene terephthalate (PTT) or polyethylene terephthalate (PET), binary PTT/PET or PP/PTT blends, and also on a ternary PP/(PTT/PET) blend. The phase structure of PP/PES–blend fibers was examined. PES microfibers showed separation from the PP matrix in blend fibers. The impact of MB composition and rheological characteristics on phase structure parameters indicate a significant contribution of the PTT in the binary MB on the length of dispersed PES microfibers in the PP matrix. However, the blends of PP and ternary MB (PP/PTT/PET) have a lower diameter and length of the PES microfibers. The presence of PTT/PET (PES) enhances the structural and mechanical properties of the blend PP/PES fibers. In addition, PTT increases the tensile strength of the PP/PES–blend fibers if a binary MB is used, while the fiber nonuniformity is reduced in the presence of a ternary MB.  相似文献

The mobility, bioavailability and toxicity of mercury in the environment strongly depend on the chemical species in which it is present in soil, sediments, water or air. In mining districts, differences in mobility and bioavailability of mercury mainly arise from the different type of mineralization and ore processing. In this work, synchrotron‐based X‐ray absorption near‐edge spectroscopy (XANES) has been taken advantage of to study the speciation of mercury in geological samples from three of the largest European mercury mining districts: Almadén (Spain), Idria (Slovenia) and Asturias (Spain). XANES has been complemented with a single extraction protocol for the determination of Hg mobility. Ore, calcines, dump material, soil, sediment and suspended particles from the three sites have been considered in the study. In the three sites, rather insoluble sulfide compounds (cinnabar and metacinnabar) were found to predominate. Minor amounts of more soluble mercury compounds (chlorides and sulfates) were also identified in some samples. Single extraction procedures have put forward a strong dependence of the mobility with the concentration of chlorides and sulfates. Differences in efficiency of roasting furnaces from the three sites have been found.  相似文献
