The 13C-NMR spectra of the synthetic membrane modifying nonadecapeptide Boc-(Aib-l-Ala)5-Gly-Ala-Aib-Pro-Ala- Aib-Aib-Glu(OBz)-Gln-OMe (Aib = α-aminoisobutyric acid), and of synthetic intermediates were used for conformational analysis in solution. The assignments of the 13C-NMR signals of Aib are based on the magnetic nonequivalence (MNE) of the geminal Cβ-signals in asymmetric environment resulting in a shift difference of 0.2–0.5 ppm due to neighbouring chiral residues. More than 4 ppm MNE are observed due to α-helical conformation and about 2.5 ppm for Aib situated in the corners of a rigid β-turn. The Ala-Cα signal is also sensitive to different secondary structures. The Cα signal for C-terminal alanine is found at 49–50 ppm, and for alanine within unordered oligopeptides it absorbs at 50–51 ppm. α-Helical environment shifts the Ala-Cα signal to lower field down to 54 ppm. In methanolic solution the nonadecapeptide shows a α-helical N-terminal region. For the C-terminus beginning with proline-14 no periodically ordered conformation is observed, and we suggest a sequence of β-turns. Furthermore the typical E/Z isomerism of the prolyl-peptide bond can be observed on proline itself and on its neighbour alanine.  相似文献

A novel β-agarase AgaJ11 belonging to the glycoside hydrolase (GH) 16 family was identified from an agar-degrading bacterium Gayadomonas joobiniege G7. AgaJ11 was composed of 317 amino acids (35 kDa), including a 26-amino acid signal peptide, and had the highest similarity (44 % identity) to a putative β-agarase from an agarolytic marine bacterium Agarivorans albus MKT 106. The agarase activity of purified AgaJ11 was confirmed by zymogram analysis. The optimum pH and temperature for AgaJ11 activity were determined to be 4.5 and 40 °C, respectively. Notably, AgaJ11 is an acidic β-agarase that was active only at a narrow pH range from 4 to 5, and less than 30 % of its enzymatic activity was retained at other pH conditions. The K m and V max of AgaJ11 for agarose were 21.42 mg/ml and 25 U/mg, respectively. AgaJ11 did not require metal ions for its activity, but severe inhibition by several metal ions was observed. Thin layer chromatography and agarose-liquefying analyses revealed that AgaJ11 is an endo-type β-agarase that hydrolyzes agarose into neoagarohexaose, neoagarotetraose, and neoagarobiose. Therefore, this study shows that AgaJ11 from G. joobiniege G7 is a novel GH16 β-agarase with an acidic enzymatic feature that may be useful for industrial applications.  相似文献

11B (I=3/2) MAS NMR in the binary glass system xV2O5–B2O3 (x=0.053, 0.43) and the ternary glass system xV2O5–B2O3–PbO (0.1x1.5) has been investigated at room temperature. In the xV2O5–B2O3 glasses, one NMR line due to BO3 unit was observed. Meanwhile in the xV2O5–B2O3–PbO, two NMR lines which arise from BO3 and BO4 units were detected, where the appearance of BO4 units is produced by the presence of PbO. From the computer-simulation of the 11B NMR central transition line (m=−1/2↔1/2), the quadrupole parameters (e2qQ/h and η) for BO3 units in xV2O5–B2O3, and those for BO3 and BO4 units in xV2O5–B2O3–PbO were obtained as a function of x. As the V2O5 content increases in xV2O5–B2O3–PbO, the e2qQ/h and η values of the BO3 associated resonance are found to slightly decrease and increase, respectively. Meanwhile, the e2qQ/h and η values of BO4 associated resonance in xV2O5–B2O3–PbO are found to slightly increase and decrease, respectively. By comparing the intensities of the total transitions (m=−3/2↔−1/2,m=−1/2↔1/2, and 1/2↔3/2) for the 11B NMR line of BO3 and BO4 units contained in xV2O5–B2O3–PbO with those of respective standard samples of 0.053V2O5–B2O3 and NaBH4, the quantitative fractions of BO3 and BO4 in xV2O5–B2O3–PbO were obtained as a function of x.  相似文献

A new hydride vapor phase epitaxy (HVPE)-based approach for the fabrication of freestanding GaN (FS-GaN) substrates was investigated. For the direct formation of low-temperature GaN (LT-GaN) layers, the growth parameters were optimized: the polarity of ZnO, the growth temperature, and the V/III ratio. The FS-GaN layer was achieved by gas etching in an HVPE reactor. A fingerprint of Zn out-diffusion was detected in the photoluminescence measurements, especially for the thin (80 μm) FS-GaN film; however, a thicker film (400 μm) was effectively reduced by optimization of GaN growth.  相似文献

In this study, we postulated that the optical retardation magnitude could be used to evaluate the stability of the LC phase through the Mueller matrix polarimeter method in LC emulsion. In addition, we found that the increase of cetearyl alcohol concentration induced the more periodic dense lattice structure in crystalline phase of LC lamellae and the sorbitan olivate was relatively more responsible for higher ordering in LC phase compared to C12–20 alkyl glucoside in terms of molecular parallel packing on the basis of the optical retardation magnitude. Furthermore, it was confirmed that the LC emulsion had higher skin barrier function owing to lower TEWL and higher reflectance than ordinary emulsion on human skin and maintained a uniform phase without phase separation for 8 weeks.  相似文献

Os(II) hydridocarbonyl complexes of coumarinyl azoimidazoles, [Osh(CO)(PPh3)2(CZ‐4R‐R′)]0/+ ( 3 , 4 ) (CZ‐R‐H = 2‐(coumarinyl‐6‐azo)‐4‐substituted imidazole or 1‐alkyl‐2‐(coumarinyl‐6‐azo)‐4‐substituted imidazole), were characterized from spectroscopic data and the single‐crystal X‐ray data for one of the complexes, [Osh(CO)(PPh3)2(CZ‐4‐Ph)] ( 3c ) (CZ‐4‐Ph = 2‐(coumarinyl‐6‐azo)‐4‐phenylimidazolate), confirmed the structure. The complexes show higher emission (quantum yield ? = 0.0163–0.16) and longer lifetime (τ = 1.4–10.3 ns) than free ligands (? = 0.0012–0.0185 and τ = 0.685–1.306 ns). Cyclic voltammetry shows quasi‐reversible metal oxidation at 0.67–0.94 V for [Os(III)/Os(II)] and 1.21–1.36 V for [Os(IV)/Os(III)] and subsequent azo reductions (?0.68 to ?0.95 V for [? N?N? ]/[? N N? ]? and irreversible < ?1.2 V for [? N N? ]?/[? N? N? ]2?) of the chelated coumarinyl azoimidazole. The complexes are photostable and show better photovoltaic power conversion efficiency than free ligands. Also, the complexes were used as catalysts for the oxidation of primary/secondary alcohols to aldehydes/ketones using oxidizing agents like N‐methylmorpholine N‐oxide, t‐BuOOH and H2O2. Density functional theory computation was carried out from the optimized structures and the data obtained were used to interpret the electronic and photovoltaic properties. Agastache rugosa Kuntze (Korean mint) is used as a spice and in folk medicine in East Asia. The present study identified a total of 18 polyphenols from the flower, leaf, stem and roots of this plant using high‐performance liquid chromatography–tandem mass spectrometry. Fourteen of these compounds had not previously been identified in these plant tissues. Each polyphenol was validated in comparison with external calibration curves constructed using structurally related compounds, with determination coefficients >0.9993. The limits of detection and quantification were 0.092–0.650 and 0.307–2.167 mg/L, respectively. Recoveries of 61.92–116.44% were observed at two spiking levels, with 0.91–11% precision, expressed as relative standard deviation (except anthraquinone spiked at 10 mg/L). Hydroxycinnamic acid was the most abundant compound in the root, while the flowers showed the highest total flavonoid level. Antioxidant activities, determined in terms of reducing power, Fe2+ chelating activity and the radical scavenging activities using α,α‐diphenyl‐β‐picrylhydrazyl and 2‐2?‐azino‐bis‐3‐ethylbenzothiazoline‐6‐sulfonic acid, increased in a concentration‐dependent manner; the highest activity was identified in the stems, followed by leaves > flowers > roots.
