Two new phenyl glycosides, mangliesides A and B (1, 2), a new ionol glycoside, manglieside C (3), two new lignan glycosides, mangliesides D and E (4, 5), were isolated from the leaves of Manglietia phuthoensis, along with two known lignans, 3-methoxymagnolol (6) and obovatol (7). Their structures were established by means of 1D and 2D NMR, electrospray ionization (ESI)-MS and HR-ESI-MS experiments. Among them, compounds 2 and 5 significantly (p<0.05) increased the growth and differentiation of osteoblastic MC3T3-E1 cells in vitro.  相似文献

The new tantalum(II) complex (eta (6)-C 7H 8)TaCl 2(PMe 3) 2 ( 1) was synthesized by the reduction of TaCl 5 with n-butyllithium in the presence of PMe 3 and cycloheptatriene. Compound 1 adopts a four-legged piano stool structure in which the tantalum center is bound to a eta (6)-cycloheptatriene ring in addition to two chlorides and two phosphine ligands in a transoid arrangement. Treatment of 1 with methyllithium results in a loss of the equivalents of HCl and formation of the eta (7)-cycloheptatrienyl complex (eta (7)-C 7H 7)TaCl(PMe 3) 2 ( 2), whereas treatment of 1 with sodium or sodium borohydride affords small amounts of the eta (5)-cycloheptadienyl complex (eta (5)-C 7H 9)TaCl 2(PMe 3) 2 ( 3). Compound 2 adopts a three-legged piano stool structure; the eta (7)-C 7H 7 ring is fully aromatic and planar. The molecular structure of 3 is similar to that of 1, except for the eta (5) binding mode of the seven-membered ring. Treatment of the previously described sandwich compound (C 5Me 5)Ta(C 7H 7) with allyl bromide affords the tantalum(V) product (C 5Me 5)Ta(C 7H 7)Br ( 4), which reacts with LiAlH 4 to give the tantalum(V) hydride (C 5Me 5)Ta(C 7H 7)H ( 5). Compound 4 also reacts with alkylating agents to generate the methyl, allyl, and cyclopropyl complexes (C 5Me 5)Ta(C 7H 7)Me ( 6), (C 5Me 5)Ta(C 7H 7)(eta (1)-CH 2CHCH 2) ( 7), and (C 5Me 5)Ta(C 7H 7)(c-C 3H 5) ( 8). Compounds 4- 8 all adopt bent sandwich structures in which the dihedral angle between the two carbocyclic rings is 34.9 degrees for the bromo compound 4, 26.6 degrees for the hydride 5, 33.1 degrees for the methyl compound 6, 34.2 degrees for the allyl compound 7, and 37.5 degrees for the cyclopropyl compound 8. Cytochrome P450 1A1 (CYP1A1) is involved in the 2-hydroxylation of estrogen, the hormone that plays a critical role in the etiology of breast carcinoma. We evaluated the associations between two CYP1A1 polymorphisms [MspI (rs4646903); Ile462Val (rs1048943)] and breast cancer in a multicenter case-control study of 513 breast cancer cases and 447 controls in Korea. Women carrying the T allele of the CYP1A1 MspI polymorphism were found to have a 1.72-fold (95% CI 1.11-2.68) greater risk of developing breast cancer. No association was found between any CYP1A1 Ile462Val polymorphism and breast cancer. Haplotype analysis of the two loci showed that the CA haplotype was associated with the lowest risk of breast cancer, and CA/CA diplotypes were associated with a lower risk of breast cancer [OR=0.28 (0.13-0.61)] than others/others diplotypes. Moreover, this reduced risk was more pronounced among women with a lower body mass index (BMI) [OR=0.18 (0.06-0.58)] or with a shorter lifetime exposure to estrogen [OR=0.23 (0.07-0.81)]. The results obtained suggest that the CYP1A1 MspI polymorphisms could affect susceptibility to breast cancer.  相似文献

TNF-alpha plays a variety of biological functions such as apoptosis, inflammation and immunity. PTEN also has various cellular function including cell growth, proliferation, migration and differentiation. Thus, possible relationships between the two molecules are suggested. TNF-alpha has been known to downregulate PTEN via NF-kappaB pathway in the human colon cell line, HT-29. However, here we show the opposite finding that TNF-alpha upregulates PTEN via activation of NF-kappaB in human leukemic cells. TNF-alpha increased PTEN expression at HL-60 cells in a time- and dose-dependent manner, but the response was abolished by disruption of NF-kappaB with p65 antisense phosphorothioate oligonucleotide or pyrrolidine dithiocarbamate. We found that TNF-alpha activated the NF-kappaB pathways, evidenced by the translocation of p65 to the nucleus in TNF-alpha-treated cells. We conclude that TNF-alpha induces upregulation of PTEN expression through NF-kappaB activation in human leukemic cells.  相似文献

Here, we report the extracellular biosynthesis of silver nanoparticles (AgNPs) and determination of their antibacterial and anticancer properties. We also explore the efficacy of bioAgNPs incorporated in cellulose nanocrystals (CNCs) and alginate (Alg) for the formation of an antibacterial hydrogel film. Streptomyces sp. PBD-311B was used for the biosynthesis of AgNPs. The synthesized bioAgNPs were characterized using UV-Vis spectroscopy, TEM, XRD, and FTIR analysis. Then, the bioAgNPs’ antibacterial and anticancer properties were determined using TEMA and cytotoxicity analysis. To form the antibacterial hydrogel film, bioAgNPs were mixed with a CNC and Alg solution and further characterized using FTIR analysis and a disc diffusion test. The average size of the synthesized bioAgNPs is around 69 ± 2 nm with a spherical shape. XRD analysis confirmed the formation of silver nanocrystals. FTIR analysis showed the presence of protein capping at the bioAgNP surface and could be attributed to the extracellular protein binding to bioAgNPs. The MIC value of bioAgNPs against P. aeruginosa USM-AR2 and MRSA was 6.25 mg/mL and 3.13 mg/mL, respectively. In addition, the bioAgNPs displayed cytotoxicity effects against cancer cells (DBTRG-0.5MG and MCF-7) and showed minimal effects against normal cells (SVG-p12 and MCF-10A), conferring selective toxicity. Interestingly, the bioAgNPs still exhibited inhibition activity when incorporated into CNC/Alg, which implies that the hydrogel film has antibacterial properties. It was also found that bioAgNP-CNC/Alg displayed a minimal or slow release of bioAgNPs owing to the intermolecular interaction and the hydrogel’s properties. Overall, bioAgNP-CNC/Alg is a promising antibacterial hydrogel film that showed inhibition against the pathogenic bacteria P. aeruginosa and MRSA and its application can be further evaluated for the inhibition of cancer cells. It showed benefits for surgical resection of a tumor to avoid post-operative wound infection and tumor recurrence at the surgical site.  相似文献
