The effect of the CHCl3 and MeOH extracts from aerial parts from the Northwestern Argentina endemic species Ixorhea tschudiana Fenzl. on germination of the dicot tomato and the monoct onion and on soil microbial activity was studied by calorimetric and calorespirometric experiments. The CHCl3 extract inhibited seedling growth of tomato which increased with increasing concentrations. Seeds imbibed with 250 mg dm?3 extract germinated 24 h later than control and those imbibed with 100 and 500 mg dm?3 extract. This was reflected in the calculated low specific seedling growth rate (pSGΔHB) as determined from calorespirometric experiments. On the other hand, seedlings obtained in 500 mg dm?3 extract showed shorter and thicker hairy roots than control with significantly higher pSGΔHB. Probably, the extract induces inhibition of water uptake by seedling roots. Germination was reduced 50% in onion seeds imbibed with 500 mg dm?3 CHCl3 extract which apparently is due to higher rate of imbibition during the first stages as determined for calorimetry. On the other hand, 83 and 250 mg kg?1 of the MeOH and CHCl3 extract seem to selectively inhibit the growth of certain microorganisms and to enhance the activity of soil actinomycetes. Two actinobacteria were isolated from soil treated with these concentrations of both extracts: Kocuria sp. and Kocuria rosea; this latter species is known as a keratinolytic agent and seems to degrade complex carbon compounds of the soil and those incorporated by the MeOH extract. Both Kocuria specie seem to be beneficial for the soil converting substrate into biomass and thus I. tschudiana could be used as a soil phytoremediator.  相似文献

Bioluminescent solid-phase analysis was proposed to monitor the selection process and to determine binding characteristics of the aptamer–target complexes during design and development of the specific aptamers. The assay involves Ca2+-regulated photoprotein obelin as a simple, sensitive and fast reporter. Applicability and the prospects of the approach were exemplified by identification of DNA aptamers to cardiac troponin I, a highly specific early biomarker for acute myocardial infarction. Two structurally different aptamers specific to various epitopes of troponin I were obtained and then tested in a model bioluminescent assay.  相似文献

Background  

β-carotene 15,15'-monooxygenase (BCMO1) catalyzes the crucial first step in vitamin A biosynthesis in animals. We wished to explore the possibility that a carbocation intermediate is formed during the cleavage reaction of BCMO1, as is seen for many isoprenoid biosynthesis enzymes, and to determine which residues in the substrate binding cleft are necessary for catalytic and substrate binding activity. To test this hypothesis, we replaced substrate cleft aromatic and acidic residues by site-directed mutagenesis. Enzymatic activity was measured in vitro using His-tag purified proteins and in vivo in a β-carotene-accumulating E. coli system.  相似文献

Cyclophanes of different ring sizes featuring perylene-3,4:9,10-tetracarboxylic acid bisimide (PBI) linked by flexible malonates were designed, synthesized, and investigated with respect to their structural, chemical and photo-physical properties. It is predominantly the number of PBIs and their geometric arrangement, which influence dramatically their properties. For example, two-PBI containing cyclophanes reveal physico-chemical characteristics that are governed by strong co-facial π–π interactions. This is in stark contrast to cyclophanes with either three or four PBIs. Key to co-facial π–π stackings are the flexible malonate linkers, which, in turn, set up the ways and means for diastereoselectivity of the homochiral PBIs at low temperatures, on one hand. In terms of selectivity, diastereomeric (M,M)/(P,P) : (M,P)/(P,M) pairs with a ratio of approximately 10 : 1 are discernible in the 1H NMR spectra in C2D2Cl4 and a complete diastereomeric excess is found in CD2Cl2. On the other hand, symmetry-breaking charge transfer as well as charge separation at room temperature are corroborated in steady-state and time-resolved photo-physical investigations. Less favourable are co-facial π–π stackings in the three-PBI containing cyclophanes. For statistical reasons, the diastereoisomers (M,M,M)/(P,P,P) and (M,M,P)/(P,P,M) occur here in a ratio of 1 : 3. In this case, symmetry-breaking charge transfer as well as charge separation are both slowed down. The work was rounded-off by integrating next to the PBIs, for the first time, hydrophobic or hydrophilic fullerenes into the resulting cyclophanes. Our novel fullerene–PBI cyclophanes reveal unprecedented diastereoselective formation of homochiral (M,M)/(P,P) pairs exceeding the traditional host–guest approach. Hybridization with fullerenes allows us to modulate the resulting solubility, stacking, cavity and chirality, which is of tremendous interest in the field.

Perylene bisimide (PBI) cyclophanes linked by flexible malonates were functionalized with fullerenes. Modulation of the chemical environment enhances the chiral self-sorting, leading exclusively to the homochiral diastereomeric pair (M,M)/(P,P).  相似文献

Polyhedral oligomeric silsesquioxane (POSS) are hybrid nanostructures of about 1.5 nm in size. These silicon (Si)-based polyhedral nanostructures are attached to a polystyrene (PS) backbone to produce a polymer nanocomposite (POSS–styrene). We have solution blended POSS–styrene of with commercial polystyrene (PS), , and studied the rheological behavior and thermal properties of the neat polymeric components and their blends. The concentration of POSS–styrene was varied from 3 up to 20 wt.%. Thermal analysis studies suggest phase miscibility between POSS–styrene and the PS matrix. The blends displayed linear viscoelastic regime and the time–temperature superposition principle applied to all blends. The flow activation energy of the blends decreased gradually with respect to the matrix as the POSS–styrene concentration increased. Strikingly, it was found that POSS–styrene promoted a monotonic decrease of zero-shear rate viscosity, η 0, as the concentration increased. Rheological data analyses showed that the POSS–styrene increased the fractional free volume and decreased the entanglement molecular weight in the blends. In contrast, blending the commercial PS with a PS of did not show the same lubrication effect as POSS–styrene. Therefore, it is suggested that POSS particles are responsible for the monotonic reduction of zero-shear rate viscosity in the blends.  相似文献

This research investigates the influence of monomer composition on the thermal transitions, mi crostructure, and viscoelastic properties of thermotro pic liquid crystalline polymers (LCPs) based on 1, 4-hydroxy-benzoic acid (B) and 2,6-hydroxy-naphthoic acid (N). Compositions B–N of 1:1, 1:3, 3:1, and 2:1 mol% were studied. The solid-to-nematic transition T s→n was greatly influenced by the monomer composition, ranging from 230 to 280 °C. However, the decomposition temperature T dec was independent of composition, all the materials degrading at T dec = 509 °C. Polarized optical microscopy showed a threaded texture in the nematic phase. Fiber X-ray diffraction showed aperiodic meridional maxima for all LCPs, the number of maxima depended on composition. B–N 3:1 and 1:3 mol% favor the orthorhombic crystallographic phase whereas the 1:1 and 2:1 mol% crystallized in a pseudohexagonal phase. Dynamic time sweeps showed that the nematic phases are stable at the temperatures studied. Strain sweeps showed the existence of a linear viscoelastic (LVE) regime. The LVE properties ranged from predominantly viscous (G < G) for the 1:1 mol% composition to predominantly elastic (G > G) and with a well-defined rubber-like regime for the 1:3 mol% composition, resembling the rheology of flexible polymer melts. The persistence length q appears to be the parameter driving the rheological behavior.
