We prepared the PLGA‐loaded anti‐cancer drug and coated it with quantum dots to make it a dual‐function nanoparticles, and analyzed its potential use in cellular imaging and curing cancers. Two cancer cell lines, paclitaxel‐sensitive KB and paclitaxel‐resistant KB paclitaxel‐50 cervical carcinoma cells, were the relativistic models for analysis of the cytotoxicity of free paclitaxel and paclitaxel‐loaded PLGA conjugated with quantum‐dot nanoparticles. The paclitaxel‐loaded PLGA conjugated with quantum dots nanoparticles were significantly more cytotoxic than the free paclitaxel drug in paclitaxel‐resistant KB paclitaxel‐50 cells. This might have been because the cancer cells developed multi‐drug resistance (MDR), which hampered the action of free paclitaxel by pumping its molecules to extracellular areas. Addition of verapamil, a P‐glycoprotein inhibitor, reversed the MDR mechanism and significantly reduced KB paclitaxel‐50 cell viability. As a result, KB paclitaxel‐50 was highly associated with MDR on the cell membrane. The cytotoxicity results indicated that PLGA nanoparticles served as drug carriers and protected the drugs from MDR‐accelerated efflux. Combined quantum dots with PLGA nanoparticles allowed additional functionality for cellular imaging. 相似文献

Necroptosis is a type of programmed cell death executed through the plasma membrane disruption by mixed lineage kinase domain-like protein (MLKL). Previous studies have revealed that an N-terminal four-helix bundle domain (NBD) of MLKL is the executioner domain for the membrane permeabilization, which is auto-inhibited by the first brace helix (H6). After necroptosis initiation, this inhibitory brace helix detaches and the NBD can integrate into the membrane, and hence leads to necroptotic cell death. However, how the NBD is released and induces membrane rupture is poorly understood. Here, we reconstituted MLKL2–154 into membrane mimetic bicelles and observed the structure disruption and membrane release of the first brace helix that is regulated by negatively charged phospholipids in a dose-dependent manner. Using molecular dynamics simulation we found that the brace region in an isolated, auto-inhibited MLKL2–154 becomes intrinsically disordered in solution after 7 ns dynamic motion. Further investigations demonstrated that a cluster of arginines in the C-terminus of MLKL2–154 is important for the molecular conformational switch. Functional mutagenesis showed that mutating these arginines to glutamates hindered the membrane disruption of full-length MLKL and thus inhibited the necroptotic cell death. The hypergraph matching problem is to find a largest collection of disjoint hyperedges in a hypergraph. This is a well-studied problem in combinatorial optimization and graph theory with various applications. The best known approximation algorithms for this problem are all local search algorithms. In this paper we analyze different linear and semidefinite programming relaxations for the hypergraph matching problem, and study their connections to the local search method. Our main results are the following: We consider the standard linear programming relaxation of the problem. We provide an algorithmic proof of a result of Füredi, Kahn and Seymour, showing that the integrality gap is exactly ${k-1+\frac{1}{k}}$ for k-uniform hypergraphs, and is exactly k ? 1 for k-partite hypergraphs. This yields an improved approximation algorithm for the weighted 3-dimensional matching problem. Our algorithm combines the use of the iterative rounding method and the fractional local ratio method, showing a new way to round linear programming solutions for packing problems. We study the strengthening of the standard LP relaxation by local constraints. We show that, even after linear number of rounds of the Sherali-Adams lift-and-project procedure on the standard LP relaxation, there are k-uniform hypergraphs with integrality gap at least k ? 2. On the other hand, we prove that for every constant k, there is a strengthening of the standard LP relaxation by only a polynomial number of constraints, with integrality gap at most ${\frac{k+1}{2}}$ for k-uniform hypergraphs. The construction uses a result in extremal combinatorics. We consider the standard semidefinite programming relaxation of the problem. We prove that the Lovász ${\vartheta}$ -function provides an SDP relaxation with integrality gap at most ${\frac{k+1}{2}}$ . The proof gives an indirect way (not by a rounding algorithm) to bound the ratio between any local optimal solution and any optimal SDP solution. This paper is a study of the effects of anisotropic matter sources on the qualitative evolution of spatially homogenous cosmologies of Bianchi type VIII. The analysis is based on a dynamical system approach and makes use of an anisotropic matter family developed by Calogero and Heinzle which generalises perfect fluids and provides a measure of deviation from isotropy. Thereby the role of perfect fluid solutions is put into a broader context. The results of this paper concern the past and future asymptotic dynamics of locally rotationally symmetric solutions of type VIII with anisotropic matter. It is shown that solutions whose matter source is sufficiently close to being isotropic exhibit the same qualitative dynamics as perfect fluid solutions. However a high degree of anisotropy of the matter model can cause dynamics to differ significantly from the vacuum and perfect fluid case. 相似文献

Producing zeolite films with controlled preferred orientation on an industrial scale is a long‐standing challenge. Herein we report on a scalable approach to the direct wet deposition of zeolite thin films and membranes while maintaining a high degree of control over the preferred crystal orientation. As a proof of concept, thin films comprising aluminophosphate zeolite AEI were cast on silicon wafer or porous alumina substrates.
