Immunization with complex mixtures, like the human plasma resulted in the generation of cloned mAb libraries (PlasmaScan? and QuantiPlasma? libraries, with >1000 individual mAbs) reacting with a nonredundant set of antigenic epitopes. mAb proteomics refers to quasi‐hypothesis‐free profiling of plasma samples with nascent or cloned mAb libraries for the discovery of disease‐specific biomarkers. Once mAbs with biomarker potential have been identified, the next task is the determination of cognate antigens recognized by the respective mAbs. To determine the cognate protein antigen corresponding to each individual mAbs in the cloned mAb libraries, we have separated human plasma by consecutive steps of desalting and various chromatography procedures. The process resulted in 783 fractions, which we termed “Analyte Library” (AL). The AL represents the human plasma proteome in relatively low‐protein complexity fractions. Here, to determine the utility of the AL, we selected ten plasma proteins and checked for their presence in the fractions. Among the ten cases, the distribution of four selected plasma proteins matched expectations, as these proteins were present only in a few fractions corresponding to their physical, chemical, and biochemical properties. However, in six cases, we observed “smear” ‐like distribution or complete absence of the proteins, suggesting that protein–protein interactions or protein variants may alter the observed plasma distribution profiles. Nevertheless, we conclude that the AL is an efficient, high throughput tool to complement the mAb biomarker discovery process with cognate protein antigen identification for each mAbs. 相似文献

The folding of disulfide proteins is of considerable interest because knowledge of this may influence our present understanding of protein folding. However, sometimes even the disulfide pattern cannot be unequivocally determined by the available experimental techniques. For example, the structures of a few small antifungal proteins (PAF, AFP) have been disclosed recently using NMR spectroscopy but with some ambiguity in the actual disulfide pattern. For this reason, we carried out the chemical synthesis of PAF. Probing different approaches, the oxidative folding of the synthetic linear PAF yielded a folded protein that has identical structure and antifungal activity as the native PAF. In contrast, unfolded linear PAF was inactive, a result that may have implications concerning its redox state in the mode of action. 相似文献

A particle phi coupling to two photons couples also radiatively to charged particles, like protons. If the particle is a light scalar, this induced coupling leads to spin-independent non-Newtonian forces. We show that the experimental constraints on exotic, fifth-type forces lead to stringent constraints on the phigammagamma coupling. Spherical nanoparticles composed of MMX chains can be made by a polymerization strategy driven by electrochemical processes. In particular, the [Pt2(MeCS2)4I2] (MMI2) dimetal subunit is employed as a monomer for the formation of [Pt2(MeCS2)4I]n spherical nanostructures on surfaces. We have paid particular attention to elucidating the general mechanism of the deposition process on the basis of in situ electrochemical measurements. The reduction of MMI2 to give the electrodeposition of nanostructures agrees well with formation of the reduced [MMI2]? species followed by a disproportionation mechanism mediated by iodide anions. The chemical composition of the particles was determined by energy‐dispersive X‐ray spectroscopy (EDX) and X‐ray photoelectron spectroscopy (XPS) to reveal the MMI2 polymer. 相似文献

A short retrospection is given on the development of M?ssbauer spectroscopy and on the early period of its application to
heterogeneous catalysis in Hungary. Besides the historical view, we present results of recent studies of a latest application
on a nanosystem. Namely, catalytic centres in Fe-ZSM-5 are studied by in situ spectroscopy, the results are correlated with
parallel activity measurements in the decomposition of N2O. Changes in paramagnetic relaxation features are interpreted as the effects of Fe2+/Fe3+ interconversions. From the analysis, the dominance of the mononuclear species is suggested at lower temperature (620 K).

The utility and validity of toxicity tests for monitoring of wastewater treatment have been assessed. The evaluated acute toxicity tests have been Vibrio fischeri, Selenastrum capricornotum and Daphnia magna tests. The validation studies indicated that the acute toxicity tests can be considered as high sensitivity analytical tools to detect common environmental concentrations of the pollutants at concentration levels as low as ng l−1. The toxicity tests showed to have discriminatory ability to distinguish between different degrees of toxicity, and the toxic specificity of the compounds on target organisms. Synergistic, additive or antagonistic effects were evaluated indicating the capacity of the toxicity test to assess the combined effects of chemicals in wastewaters. The reproducibility of these tests, calculated as relative standard deviation, is acceptable in the range of 5-22.3%. The application of multivariate date analysis proved that toxicity and chemical measures are complementary analytical tools for monitoring of wastewaters quality. The toxicity tests are useful analytical tools for screening of chemical analysis and as an early warning system to monitor the treatment of WWTPs. The use of single toxicity test or battery of tests is the best approach to evaluate the risk because they are reliable indices of the toxic impact of effluents in the aquatic environment.
