In matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI TOF MS), analyte signals can be substantially suppressed by other compounds in the sample. In this technical note, we describe a modified thin‐layer sample preparation method that significantly reduces the analyte suppression effect (ASE). In our method, analytes are deposited on top of the surface of matrix preloaded on the MALDI plate. To prevent embedding of analyte into the matrix crystals, the sample solution were prepared without matrix and efforts were taken not to re‐dissolve the preloaded matrix. The results with model mixtures of peptides, synthetic polymers and lipids show that detection of analyte ions, which were completely suppressed using the conventional dried‐droplet method, could be effectively recovered by using our method. Our findings suggest that the incorporation of analytes in the matrix crystals has an important contributory effect on ASE. By reducing ASE, our method should be useful for the direct MALDI MS analysis of multicomponent mixtures. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

As an extention of previous reports, variables affecting the performance of columns modified by silicon deposition have been optimized. The aim of this work was to produce thermostable apolar glass capillary columns regardless of the glass type used and to maintain column performance under stressful conditions. The main parameters studied were the influence of glass type, leaching effects, and silicon layer thickness on column activity. Deactivation and stationary phase conditions were held constant. Both coated and uncoated columns were tested. The thickness of the silicon layer was found to be relatively unimportant. There was no difference whether soft glass or borosiiicate glass was used and leaching before silicon deposition did not influence column activity. Bare silicon surfaces showed considerable activity especially in respect to interactions with free acids and bases. There are indications that the surface consists of silicon oxide and other oxygenated forms of silicon rather than of the element. Treatment of the silicon layers with dilute hydrofluoric acid and the strict exclusion of traces of oxygen and water did not improve the situation. In spite of such specific interactions, silicon surfaces were easily deactivated by heat treatment with polysiloxanes. Silicon surfaces deactivated by baking with octamethyltetrasiloxane at 400°C are inert and temperature stable and show characteristics similar to persilylated surfaces.  相似文献

A new solvent extraction system was developed for extraction of PFCs from food. The extraction is carried out with 75:25 (v/v) tetrahydrofuran:water, a solvent mixture that provides an appropriate balance of hydrogen bonding, dispersion and dipole–dipole interactions to efficiently extract PFCs with chains containing 4–14 carbon atoms from foods. This mixture provided recoveries above 85% from foods including vegetables, fruits, fish, meat and bread; and above 75% from cheese. Clean-up with a weak anion exchange resin and Envi-carb SPE, which were coupled in line for simplicity, was found to minimize matrix effects (viz. enhancement or suppression of electrospray ionization). The target analytes (PFCs) were resolved on a perfluorooctyl phase column that proved effective in separating mass interferences for perfluorooctane sulfonate (PFOS) in fish and meat samples. The mass spectrometer was operated in the negative electrospray ionization mode and used to record two transitions per analyte and one per mass-labeled method internal standard. The target PFCs were quantified from solvent based calibration curves. The limits of detection (LODs) were as low as 1–5 pg analyte g−1 food; by exception, those for C4 and C5 PFCs were somewhat higher (25–30 pg g−1) owing to their less favourable mass response. To the best of our knowledge these are among the best LODs for PFCs in foods reported to date. The analysis of a variety of foods revealed contamination with PFCs at levels from 4.5 to 75 pg g−1 in 25% of samples (fish and packaged spinach). C10–C14 PFCs were found in fish, which testifies to the need to control long-chain PFCs in this type of food. The proposed method is a useful tool for the development of a large-scale database for the presence of PFCs in foods.  相似文献

A variety of new coordination compounds with transition-metal salts and the ligand trimethyl[1,2,4]triazolo[1,5-a]pyrimidine (abbreviated as tmtp) is described, together with several of their 3D crystal structures and spectroscopic and magnetic properties. The compounds were selected based on the coordination ability of the counterion, halide, nitrate, sulfate, thiocyanate and perchlorate. The formed coordination compounds and their coordination numbers were found to be strongly dependent on both the cation and the used counter-anion. The several compounds studied have the following structural formulae: [CuCl2(tmtp)2], [CuBr2(tmtp)2], [ZnBr2(tmtp)2], [Cu(NO3)2(tmtp)2], [CuSO4(tmtp)2]2(H2O)(MeOH), [Cu(H2O)(NCS)2(tmtp)2], [Zn(NCS)2(tmtp)2], [Cd(NCS)2(tmtp)2] and [M(H2O)2(tmtp)4](BF4)2, in which M = Co, Ni, Zn.The new coordination compounds have been further characterized by NMR, (far-)IR and LF spectra, as well as by C, H, N element analyses, and EPR spectra for the Cu(II) compounds. The coordination around the metal varies from 4 (Zn, Cu), via 5 (Cu) to 6 (for Co, Cu and Cd). The anions usually complete the coordination sphere; only the Co and Zn compounds with the tetrafluoridoborate anions have no coordinated anions, but water ligands complete the octahedral coordination sphere. In the 5-coordinated [Cu(H2O)(NCS)2(tmtp)2] water completes the square pyramid geometry.  相似文献

Recently, structure 8b has been assigned to the iridoid specionin. In the present letter the total synthesis of 8b is described. Comparison of spectral data reveals that the final product is not identical with natural specionin. Therefore the original structural assignment of specionin must be revised.  相似文献

The mass fragmentographic determination of medroxyprogesterone acetate (MPA) in serum, using as internal standard medroxyprogesterone propionate (MPP) synthesized from MPA, is described. After addition of MPP, the sera are extracted on Sep-Pak C18 cartridges and MPA and MPP are detected as their respective 3-enol trifluoroacyl esters. Serum samples from 84 patients with breast cancer, daily receiving MPA orally, were determined showing a large variation in MPA concentrations (4-349 ng/ml). Our proposed gas chromatographic-mass spectrometric (GC-MS) method, which can be considered as a reference, was compared with a radioimmunoassay (RIA) method showing a correlation coefficient of 0.73 (n = 69; p much less than 0.001). The assay was also used to determine sequential serum levels of patients receiving a single oral dose of MPA. With only minor adjustments, the GC-MS method allows the determination of serum concentrations of related steroids such as megestrol acetate and cyproterone acetate.  相似文献

The dynamics of water confined to mesoporous regions in minerals such as swelling clays and zeolites is fundamental to a wide range of resource management issues impacting many processes on a global scale, including radioactive waste containment, desalination, and enhanced oil recovery. Large-scale atomic models of freely diffusing multilayer smectite particles at low hydration confined in a silicalite cage are used to investigate water dynamics in the composite environment with the ReaxFF reactive force field over a temperature range of 300-647 K. The reactive capability of the force field enabled a range of relevant surface chemistry to emerge, including acid/base equilibria in the interlayer calcium hydrates and silanol formation on the edges of the clay and inner surface of the zeolite housing. After annealing, the resulting clay models exhibit both mono- and bilayer hydration structures. Clay surface hydration redistributed markedly and yielded to silicalite water loading. We find that the absolute rates and temperature dependence of water dynamics compare well to neutron scattering data and pulse field gradient measures from relevant samples of Ca-montmorillonite and silicalite, respectively. Within an atomistic, reactive context, our results distinguish water dynamics in the interlayer Ca(OH)(2)·nH(2)O environment from water flowing over the clay surface, and from water diffusing within silicalite. We find that the diffusion of water when complexed to Ca hydrates is considerably slower than freely diffusing water over the clay surface, and the reduced mobility is well described by a difference in the Arrhenius pre-exponential factor rather than a change in activation energy.  相似文献

In this work we present the preparation of highly luminescent anisotropic CdTe/CdSe colloidal heteronanocrystals. The reaction conditions used (low temperature, slow precursor addition, and surfactant composition) resulted in a tunable shape from prolate to branched CdTe/CdSe nanocrystals. Upon CdSe shell growth the heteronanocrystals show a gradual evolution from type-I to type-II optical behavior. These heteronanocrystals show a remarkably high photoluminescence quantum yield (up to 82%) and negligible thermally induced quenching up to temperatures as high as 373 K.  相似文献

Alloyed catalysts receive considerable attention, because of their unique catalytic properties; they often show higher selectivity, activity, and stability compared to the pure metal particles. To provide insights in the origins of these features, we report the structure and the interaction of hydrogen with each of the metals in an intimately mixed platinum-gold catalyst and compare these characteristics to those in the single metal particles. X-Ray absorption spectroscopy (XAS) and electron microscopy analysis showed that the structure of the mixed particle differed from the single metal particles. The interaction of platinum with hydrogen is stronger than the H-Au interaction and the adsorption sites were different. EXAFS analysis showed that the structure of the platinum clusters changes with increasing hydrogen coverage, observed as a relaxation of the contracted Pt-Pt distance and an increase in the Pt-Pt coordination number. No such changes were observed for gold clusters. Well-mixed PtAu-alloy clusters, with a bulk Au-to-Pt ratio of two, supported on SiO(2), adsorb hydrogen on both platinum and gold atoms, which indicates that gold cannot be regarded as an inert metal. The heat of adsorption on the platinum ensembles does not decrease upon alloying; the weakening of the overall hydrogen adsorption strength when alloying platinum with gold is an ensemble-size effect.  相似文献

The direct bioelectrocatalysis by an NAD(P)‐reducing hydrogenase is reported for the first time. In contrast to previous attempts to involve similar enzymes in bioelectrocatalysis [1–4], which were in fact unsuccessful, in our report an effective electrocatalysis by Pyrococcus furiosus hydrogenase is convincingly shown by (i) achievement of the hydrogen equilibrium potential and (ii) a high current of hydrogen oxidation (0.3 mA cm?2 at 100 mV overpotential and at 75 °C). The latter is just a few times lower compared to enzyme electrodes based on NAD(P)‐independent hydrogenases.  相似文献
