Application of recombinant phage display antibody system in study of Codakia orbicularis gill proteins

We used the recombinant phage display antibody system (RPAS) to obtain chimeric single-chain fragment variable (ScFv) antibodies to gill proteins of the white clam Codakia orbicularis (Linné, 1758). After three rounds of selection on immunotubes loaded with total gill protein extract, recombinant phages exhibiting antibodies to gill proteins were isolated and tested by enzyme-linked immunosorbent assay (ELISA). Clones exhibiting a high affinity for the mollusk proteins were selected for production of soluble ScFv antibodies, which were purified for subsequent analysis. ScFv antibodies exhibited a reaction specific for a protein whose molecular mass was about 15,000 Daltons and that was detected by the antigen capture technique followed by sodium dodecyl sulfate polyacrylamide gel electrophoresis and Western blotting.     

Rapid separation of phenylthiohydantoin amino acids: ambient pressure ion-mobility mass spectrometry (IMMS)

An electrospray ionization (ESI) ambient pressure ion-mobility spectrometer (APIMS) interfaced to an orthogonal reflector time-of-flight mass spectrometer (TOFMS) was evaluated for the first time as a detector for the identification of phenylthiohydantoin (PTH)-derivatized amino acids, the final products in the Edman sequencing process of peptides and proteins. The drift and flight times of the twenty common PTH amino acids were characterized by a well-defined 2-D mobility/mass spectral pattern. The combination of mobility/mass modes of analysis gave rise to a unique trend-line formation for the series of PTH amino acids. In addition, each PTH amino acid had a unique reduced mobility constant K(o), thus enabling the differentiation of all the amino acid derivatives including the PTH-leucine and PTH-isoleucine isomers. More importantly it was shown that it was possible to resolve a complete reference mixture of PTH amino acids in a single experimental run in less than 1 min. Detection limits for the PTH amino acids were found to range from 1.04 to 3.52 ng; indicating that the limits of detection were less than 17.0 pmol for all of the PTH amino acids.     

Optimization of alkali-alkalkine earth cation separation on weak cation-exchangers

Summary The elution behavior of alkali and earth alkaline cations has been studied on a newly synthesized weak cationexchanger (WCX). Applying this stationary phase isocratic separation of these cations is possible. The dependence of retention on competing ion concentration is described via a simple model. The influence of crown ethers as well as organic eluent additives on selectivity and peak efficiency is demonstrated. With optimized separation such cations can be determined with detection limits of about 0.5 ppm using a conductivity detector without ion suppression.     

Temperature and magnetic field dependent correlations in the singlet ground state system CsFeBr3

The magnetic excitations in CsFeBr₃ have been measured with inelastic scattering of cold neutrons to high precision at 80 mK. The fact that the lowest frequency mode softens with decreasing temperature but stabilizes at 0.11 THz below 2.5 K is the indication that CsFeBr₃ remains a SGS system forT0.From dispersion curves measured earlier in a magnetic field along the chain axis experimental intensities were derived and in turn correlation lengths. Correlation lengths were also calculated using the new parameters for exchange and anisotropy. The experimental results and the calculations both show that the correlation lengths increase for increasing magnetic field, flatten off around the phase transformation at 4.1 Tesla with a maximum of about 35 Å and then decrease again for higher fields. No critical phenomena connected with this phase transformation could be detected.The Mirror Mode published earlier was found to be of spurious origin due to double scattering. It does not exist.     

Syntheses and Structures of Trilithium Cyclotriphosphazenates Equipped with 2‐Halo‐aryl Substituents

Hexakis (2‐halo‐anilino) cyclotriphosphazenes (2‐X‐C₆H₄NH)₆P₃N₃ {X = F ( 1d ), Cl ( 1e ), Br ( 1f )} were prepared by refluxing mixtures of hexachloro cyclotriphosphazene, 2‐haloaniline and triethylamine in toluene and characterized by single crystal X‐ray diffraction. 1d , 1e and 1f were reacted with ⁿBuLi in thf. Reactions were monitored with ³¹P NMR. Addition of three equivalents of ⁿBuLi yields lithium complexes of trianionic phosphazenates [{(thf)₂Li}₃{(2‐X‐C₆H₄N)₃(2‐X‐C₆H₄NH)₃P₃N₃}] {X= F ( 2d ), Cl ( 2e ) and Br ( 2f )}. 2d , 2e and 2f were structurally characterized by X‐ray diffraction, which reveals monomeric cis‐metalated phosphazenates featuring central P₃N₃ ring systems of chair conformation. Lithium ions reside in three N(eq)‐P‐N(endo) chelation sites at one face of the P₃N₃ ring system. Li…X distances are rather long (> 3Å) indicating no Li‐X interactions.     

Chiral Chromium Salen@rGO as Multipurpose and Recyclable Heterogeneous Catalyst

The first immobilization of a pyrene-tagged chromium salen complex through π-π noncovalent interactions on reduced graphene oxide (rGO) is described. A very robust supported catalytic system is obtained to promote asymmetric catalysis in repeated cycles, without loss of activity or enantioselectivity. This specific behavior was demonstrated in two different catalytic reactions (up to ten reuses) promoted by chromium salen complexes, the cyclohexene oxide ring-opening reaction and the hetero-Diels-Alder cycloaddition between various aldehydes and Danishefsky's diene. Furthermore, the chiral chromium salen@rGO has been found to be compatible with a multi-substrate type use, in which the structure of the substrate involved is modified each time the catalyst is reused.
