Fluorescence polarization (FP) has become widely employed for high throughput screening used in pharmaceutical drug discovery. Assays of important signal transduction targets are now adapted to FP. In this review we examine assays for cyclic adenosine monophosphate, phosphodiesterases, and protein kinases and phosphatases using FP competitive immunoassays and a direct enzymatic method called IMAP.  相似文献

An automatic procedure for the determination of ethanol in wines using a flow system based on multicommutation and enzymatic reaction is described. Alcohol oxidase was immobilized on aminopropyl glass beads and packed in an acrylic column. The peroxide due to enzymatic reaction with ethanol reacted with luminol and generated the chemiluminescence radiation that was monitored by using a laboratory-made detector based on photodiodes. The system manifold comprised a set of 3-way solenoid valves controlled by a microcomputer furnished with electronic interfaces, which ran on software written in Quick BASIC 4.5 to provide facilities to perform on-line sample dilution, reagent addition, and data acquisition. After system parameters optimization, ethanol samples were processed without prior pretreatment. The following suitable features were achieved: linear response ranging from 2.5 to 25% (v/v) ethanol, relative standard deviation of 1.8% (n = 10), detection limit of 0.3% (v/v) ethanol, sampling rate of 23 determinations per hour, and low reagent consumption of 0.23 mg luminol and 7 mg hexacyanoferrate (III) per determination. When the results were compared with those obtained using the AOAC Official Method, no significant difference at the 90% confidence level was observed.  相似文献

A synthesis of a novel electrophilic reagent--β-trifluoroacetylketene diphenyldithioacetal S,S-tetroxide 3 is described. The reaction of 3 with various electron-rich aromatics such as 1,3-dimethoxybenzene, 2-methylthiophene, N-methylpyrrole, and 2-methylindole was investigated. In the course of these reactions an unusual migration of a phenylsulfonyl group takes place. An easy and ready for scale-up procedure for the synthesis of previously unknown β-aryl, α-phenylsulphonyl substituted α,β-unsaturated trifluoromethyl ketones is reported.  相似文献

We consider the mean-field classical Heisenberg model and obtain detailed information about the total spin of the system by studying the model on a complete graph and sending the number of vertices to infinity. In particular, we obtain Cramér- and Sanov-type large deviations principles for the total spin and the empirical spin distribution and demonstrate a second-order phase transition in the Gibbs measures. We also study the asymptotics of the total spin throughout the phase transition using Stein’s method, proving central limit theorems in the sub- and supercritical phases and a nonnormal limit theorem at the critical temperature.  相似文献

Attempts are made to efficiently decouple (13)C nuclei without significant loss of coherence during the application of the decoupling package. Such attempts are based on the S(3)E spin-state selection method. A newly developed double S(3)E (DS(3)E) is particularly efficient for C(alpha) detection for proteins as large as 480 kDa.  相似文献

The C-C chemokine receptor 2 (CCR2) was proved as a multidrug target in many diseases like diabetes, inflammation and AIDS, but rational drug design on this target is still lagging behind as the information on the exact binding site and the crystal structure is not yet available. Therefore, for a successful structure-based drug design, an accurate receptor model in ligand-bound state is necessary. In this study, binding-site residues of CCR2 was determined using in silico alanine scanning mutagenesis and the interactions between TAK-779 and the developed homology model of CCR2. Molecular dynamic simulation and Molecular Mechanics-Generalized Born Solvent Area method was applied to calculate binding free energy difference between the template and mutated protein. Upon mutating 29 amino acids of template protein and comparison of binding free energy with wild type, six residues were identified as putative hot spots of CCR2.  相似文献

Electron–nuclear double resonance (ENDOR) spectroscopy provides useful information on hyperfine interactions between nuclear magnetic moments and the magnetic moment of an unpaired electron spin. Because the hyperfine coupling constant reacts quite sensitively to polarity changes in the direct vicinity of the nucleus under consideration, ENDOR spectroscopy can be favorably used for the detection of subtle protein–cofactor interactions. A number of pulsed ENDOR studies on flavoproteins have been published during the past few years; most of them were designed to characterize the flavin cofactor by means of its protonation state, or to detect individual protein–cofactor interactions. The aim of this study is to compare the pulsed ENDOR spectra from different flavoproteins in terms of variations of characteristic proton hyperfine values. The general concept is to observe limits of possible influences on the cofactor’s electronic state by surrounding amino acids. Furthermore, we compare ENDOR data obtained from in vivo experiments with in vitro data to emphasize the potential of the method for gaining molecular information in complex media.  相似文献
