UV‐induced toxicity is characterized by marked oxidative stress, accompanied by the depletion of key cellular antioxidants, particularly glutathione (GSH). Replenishing cellular GSH may represent a means of counteracting UV‐induced toxicity: however, treatment with free GSH is not therapeutically effective due to its unfavorable pharmacokinetic properties. In this study, we show that S‐acyl‐glutathione (acyl‐SG) derivatives, which consist of an acyl chain (of variable length and saturation) linked via a thioester bond to GSH, increase intracellular levels of reduced GSH in primary skin fibroblasts, adenocarcinoma HeLa and neuroblastoma SH‐SY5Y cells. Consistent with this, acyl‐SG derivatives protect against UV‐induced reactive oxygen species (ROS) production and UV‐B/C‐mediated lipid peroxidation and caspase‐3 activation in the analyzed cell lines, with unsaturated thioesters displaying a significantly greater protective effect. Taken together, our findings suggest that acyl‐SG thioesters may be therapeutically effective in the treatment of UV‐related skin disorders and oxidative stress‐mediated conditions in general. 相似文献

A series of cis-cis-triaminocyclohexane Zn(II) complex-anthraquinone intercalator conjugates, designed in such a way to allow their easy synthesis and modification, have been investigated as hydrolytic cleaving agents for plasmid DNA. The ligand structure comprises a triaminocyclohexane platform linked by means of alkyl spacers of different length (from C(4) to C(8)) to the anthraquinone group which may intercalate the DNA. At a concentration of 5 microM, the complex of the derivative with a C(8) alkyl spacer induces the hydrolytic stand scission of supercoiled DNA with a rate of 4.6 x 10(-6) s(-1) at pH 7 and 37 degrees C. The conjugation of the metal complex with the anthraquinone group leads to a 15-fold increase of the cleavage efficiency when compared with the anthraquinone lacking Zn-triaminocyclohexane complex. The straightforward synthetic procedure employed, allowing a systematic change of the spacer length, made possible to gain more insight on the role of the intercalating group in determining the reactivity of the systems. Comparison of the reactivity of the different complexes shows a remarkable increase of the DNA cleaving efficiency with the length of the spacer. In the case of too-short spacers, the advantages due to the increased DNA affinity are canceled due to the incorrect positioning of the reactive group, thus leading to cleavage inhibition. 相似文献

The novel coordination polymers [Cu(Hoxonic)(H2O)]n ( 1 ) and [Cu(Hoxonic)(bpy)0.5]n ? 1.5 n H2O ( 2?H2O ) (H3oxonic: 4,6‐dihydroxy‐1,3,5‐triazine‐2‐carboxylic acid; bpy: 4,4′‐bipyridine) have been isolated and structurally characterised by ab initio X‐ray powder diffraction. The dense phase 1 contains 1D zig‐zag chains in which Hoxonic dianions bridge square‐pyramidal copper(II) ions, apically coordinated by water molecules. On the contrary, 2?H2O , prepared by solution and solventless methods, is based on 2D layers of octahedral copper(II) ions bridged by Hoxonic ligands, further pillared by bpy spacers. The resulting pro‐porous 3D network possesses small hydrated cavities. The reactivity, thermal, magnetic and adsorptive properties of these materials have been investigated. Notably, the adsorption studies on 2 show that this material possesses unusual adsorption behaviour. Indeed, guest uptake is facilitated by increasing the thermal energy of both the guest and the framework. Thus, neither N2 at 77 K nor CO2 at 195 K are incorporated, and CH4 is only minimally adsorbed at 273 K and high pressures (0.5 mmol g?1 at 2500 kPa). By contrast, CO2 is readily incorporated at 273 K (up to 2.5 mmol g?1 at 2500 kPa). The selectivity of 2 towards CO2 over CH4 has been investigated by means of variable‐temperature zero coverage adsorption experiments and measurement of breakthrough curves of CO2/CH4 mixtures. The results show the highly selective incorporation of CO2 in 2 , which can be rationalised on the basis of the framework flexibility and polar nature of its voids. 相似文献

Field-flow fractionation (FFF) is a mature technique in bioanalysis, and the number of applications to proteins and protein complexes, viruses, derivatized nano- and micronsized beads, sub-cellular units, and whole cell separation is constantly increasing. This can be ascribed to the non-invasivity of FFF when directly applied to biosamples. FFF is carried out in an open-channel structure by a flow stream of a mobile phase of any composition, and it is solely based on the interaction of the analytes with a perpendicularly applied field. For these reasons, fractionation is developed without surface interaction of the analyte with packing or gel media and without using degrading mobile phases. The fractionation device can be also easily sterilized, and analytes can be maintained under a bio-friendly environment. This allows to maintain native conditions of the sample in solution.In this review, FFF principles are briefly described, and some pioneering developments and applications in the bioanalytical field are tabled before detailed report of most recent FFF applications obtained also with the hyphenation of FFF with highly specific, sensitive characterization methods. Special focus is finally given to the emerging use of FFF as a pre-analytical step for mass-based identification and characterization of proteins and protein complexes in proteomics. 相似文献

This paper reports the development of calibration models for quality control in the production of ethylene/propylene/1-butene terpolymers by the use of multivariate tools and FT-IR spectroscopy.1-Butene concentration prediction is achieved in terpolymers by coupling FT-IR spectroscopy to multivariate regression tools. A dataset of 26 terpolymers (14 coming from a constrained experimental design for mixtures, plus 12 terpolymers used for external validation) was analysed by FT-IR spectroscopy. An internal method of “Polimeri Europa” plant, based on 13C NMR spectroscopy is used to determine the percentage of 1-butene in the samples. Then, different multivariate tools are used for 1-butene concentration prediction based on the FT-IR spectra recorded. Different multivariate calibration methods were explored: principal component regression (PCR), partial least squares (PLS), stepwise OLS regression (SWR) and artificial neural networks (ANNs). The model obtained by back-propagation neural networks turned out to be the best one. The performances of the BP-ANN model were further improved by variable selection procedures based on the calculation of the first derivative of the network.The proposed approach allows the monitoring in real time of the polymer synthesis and the estimation of the characteristics of the product attainable from the concentration of 1-butene. 相似文献

An investigation on filtration procedures and SPE sorbents used for the determination of traces of the most common veterinary fluoroquinolones (FQs), marbofloxacin (MAR) and enrofloxacin (ENR) used as antibacterial agents in cattle and swine farms in the province of Pavia (Italy), was performed in natural waters. The filter composition and the sorbent used in the SPE strongly influence the correct recovery, both in terms of total and dissolved FQs concentration. An accurate comparison among different filters and SPE sorbents showed that a full determination of analytes was possible on nylon filters followed by anionic (WAX) and hydrophilic–lipophilic balance (HLB) resins as SPE. Quantitative analysis was done by chromatography with fluorescence detection (HPLC‐FD). Fluoroquinolones recovery was between 90 and 116% with RSD not greater than 10% (sample volume 250 mL). The developed method allowed to determine both dissolved and NOM‐absorbed fractions of FQs, therefore a full determination of the analytes was possible. Limits of detection (LOD) and quantification (LOQ) were, respectively, 0.7 and 2.2 ng/L for ENR and 2 and 6 ng/L for MAR. The kinetics of degradation under solar light was explored. 相似文献

Anion–π interactions between a π‐acidic aromatic system and an anion are gaining increasing recognition in chemistry and biology. Herein, the binding features of an electron‐deficient aromatic system (1,3,5‐trinitrobenzene (TNB)) and selected anions (OH?, Br?, and I?) are examined in the gas phase by using the combined information derived from collision‐induced dissociation experiments at variable energy, infrared multiple‐photon dissociation spectroscopy, and quantum chemical calculations. We provide spectroscopic evidence for two different structural motifs of anion–arene complexes depending on the nature of the anion. The TNB–OR? complexes (R=H, or alkyl groups which were studied earlier) adopt an anionic σ‐complex structure whereby RO? attacks the aromatic ring with covalent bond formation, and develops a tetrahedral ring carbon bound to H and OR. The halide complexes rather conform to a structure in which the TNB moiety is hardly altered, and the halogen is placed on an unsubstituted carbon atom over the periphery of the ring at a C–X distance that is appreciably longer than a typical covalent bond length. The ensuing structural motif, previously characterized in the solid state and named weak σ interaction, is now confirmed by an IR spectroscopic assay in the gas phase, in which the sampled species are unperturbed by crystal packing or solvation effects. 相似文献

Ditopic salophen-UO(2) receptors 1-4 and 7 co-crystallize with tetramethylammonium (TMA) chloride and fluoride salts producing good quality crystals amenable for X-ray diffraction characterization. The arrangement of the receptor and salt units in the crystal lattice is such that tetrameric ball-shaped assemblies are formed, where an inner cluster of four TMA cations are surrounded by an outer shell of four UO(2)-bound anions. These elaborate architectures, which occur in all cases, regardless of a certain degree of structural modification on the receptors, lead to lattices that belong to non-centrosymmetric (NCS) space groups. Interestingly, the tetragonal symmetry of the tetrameric ball-shaped assemblies is either retained (I4?) or lost (R3c and I4?3d) at the lattice level, without compromising the NCS nature of the crystal lattices. The principal X-ray investigation on TMAX (X = Cl/F) co-crystals, that is, 1-(TMA)Cl, 2-(TMA)Cl, 3-(TMA)Cl, 4-(TMA)Cl, 7-(TMA)Cl, and 7-(TMA)F, is accompanied by NMR and electrospray ionization (ESI) mass spectrometry studies to gather additional insight on the modality of formation of the solid state structures observed. The important role of cation-π interactions in the receptor-salt recognition process is renewed and strengthened by comparison with NMR titration data with a novel reference compound, the salophen-UO(2) complex 8. Given the importance of NCS and polar crystalline solids in the development of functional materials, this study shows that this property can be introduced into elaborate host-guest systems, as those which assemble in the architectures described here, thus expanding its field of applicability. 相似文献
