One new 3′,4′,5′,5,7-pentahydroxy-2′,2?-bis-dihydrobiflavonol, acuminatanol (1), was obtained from the aqueous extract of Trichoscypha acuminata. The structure elucidation process was performed primarily utilizing a capillary scale NMR probe. Acuminatanol (1) is the first example of a bis-dihydroflavonol linked exclusively via the B-rings at C-2′ and C-2? positions. To date, it is the first naturally-occurring compound reported from the genus Trichoscypha, and the first new natural product published from our compound libraries generated from the aqueous extracts of American and African plants. 相似文献
Poly(ethylene oxide) (PEO) hydrogels were synthesized in organic solvents, or for better results in water, via the free-radical homopolymerization of α,ω-methacryl-oyloxy PEO macromonomers. Their characteristics (amount of extractable material, equilibrium swelling degree, uniaxial compression modulus) could be controlled by the polymerization parameters (precursor molar mass, macromonomer concentration, polymerization time). In aqueous media, the hydrophobic end-standing polymerizable methacrylic units of the macromonomers self-organize, and their polymerization leads to networks with better mechanical properties than those prepared with the same macromonomers but in organic solvents. In vitro tests confirmed their good biocompatibility: almost no adhesion of cells was evident. It was confirmed that glucose diffuses through these hydrogels. Insulin diffusion was also studied but found to be more complex.

Time-course analysis of root protein profiles was studied by two-dimensional gel electrophoresis and silver staining in the model plant Medicago truncatula, inoculated either with the arbuscular mycorrhizal fungus Glomus mosseae or with the nitrogen fixing bacterium Sinorhizobium meliloti. Protein modifications in relation to the development of both symbioses included down- and upregulations, as well as newly induced polypeptides. Matrix assisted laser desorption/ionization-time of flight-mass spectrometry after trypsin digestion clearly identified one polypeptide induced in nodulated roots as a M. truncatula leghemoglobin. Internal sequencing with a quadrupole time-of-flight mass spectrometer and database searches confirmed the induction of proteins previously described in root symbioses, and revealed the implication of other proteins. In nodulated roots, one polypeptide was identified as an elongation factor Tu from S. meliloti, while another one could not be assigned a function. In mycorrhizal roots, analyzed proteins also included a protein of unknown function, as well as a glutathione-S-transferase, a fucosidase, a myosin-like protein, a serine hydroxymethyltransferase and a cytochrome-c-oxidase. These results emphasize the usefulness of proteome analysis in identifying molecular events occurring in plant root symbioses. 相似文献
A review about the application of response surface methodology (RSM) in the optimization of analytical methods is presented. The theoretical principles of RSM and steps for its application are described to introduce readers to this multivariate statistical technique. Symmetrical experimental designs (three-level factorial, Box-Behnken, central composite, and Doehlert designs) are compared in terms of characteristics and efficiency. Furthermore, recent references of their uses in analytical chemistry are presented. Multiple response optimization applying desirability functions in RSM and the use of artificial neural networks for modeling are also discussed. 相似文献
Herein, the extraction of bioactive compounds from umbu fruit peel was optimized using thermal-assisted solid–liquid extraction. In parallel, antioxidant, antimicrobial, and inhibitory effects against α-amylase of optimized extract were also evaluated. The combination of operational conditions including the temperature (32–74 °C), ethanol concentration (13–97%), and solid/liquid ratio (1:10–1:60; w/v) was employed using a rotational central composite design for optimization. The extracts were evaluated for total phenolic compounds (TPC), total flavonoid compounds (TFC) and antioxidant capacity by ABTS•+, DPPH• and FRAP assays. The bioactive profile of the optimized extract was obtained by ultra-performance liquid chromatography coupled to quadrupole/time-of-flight mass spectrometry in electrospray ionization in both negative and positive modes. The statistically evaluated results showed that the optimal operational conditions for the recovery of bioactive compounds from umbu fruit peel included 74 °C, 37% ethanol, and a solid–liquid ratio of 1:38. Under these conditions, the obtained values were 1985 mg GAE/100 g, 1364 mg RE/100 g, 122 µmol TE/g, 174 µmol/TE g and 468 µmol Fe2+/g for TPC, TFC, ABTS•+, DPPH•, and FRAP assays, respectively. In addition, the optimized extract was effective against Gram-positive and Gram-negative bacteria (MBC ranged from 0.060 to 0.24 mg GAE/mL), as well as it was effective to inhibit α-amylase (IC50 value of 0.076 mg GAE/mL). It will be shown in this paper that the input oriented DEA BCC model can generate negative efficiencies that are usually hidden in the model. The impact of these negative efficiencies becomes obvious when using input oriented Cross Evaluation models. With the help of an example with one input and one output, the conditions for the possible occurrence of negative efficiencies will be shown. Furthermore, we will show that a small intuitive change in the BCC multipliers model, previously presented in other papers, corrects this situation. We show why this change is used and compared it with an alternative formulation, which avoid negative efficiencies, namely the Non-Decreasing Returns to Scale (NDRS) model. We also show that the formulation studied in this paper is less restrictive than the NDRS model. The study of this variation in the DEA BCC model will be complemented with the formulation of the dual envelope model. This model changes the original frontier. Using the concept of non-observed DMUs, those variations can be graphically analyzed. We have also carried out some algebraic studies concerning benchmarks, multipliers and returns to scale. 相似文献
