Disruption of the parvulin family peptidyl prolyl isomerase (PPIase) Pin1 gene delays reentry into the cell cycle when quiescent primary mouse embryo fibroblasts are stimulated with serum. Since Pin1 regulates cell cycle progression, a Pin1 inhibitor would be expected to block cell proliferation. To identify such inhibitors, we screened a chemical compound library for molecules that inhibited human Pin1 PPIase activity in vitro. We found a set of compounds that inhibited Pin1 PPIase activity in vitro with low microM IC50s and inhibited the growth of several cancer lines. Among the inhibitors, PiB, diethyl-1,3,6,8-tetrahydro-1,3,6,8-tetraoxobenzo[lmn] phenanthroline-2,7-diacetate ethyl 1,3,6,8-tetrahydro-1,3,6,8-tetraoxo-benzo[lmn] phenanthroline-(2H,7H)-diacetate, had the least nonspecific toxicity. These results suggest that Pin1 inhibitors could be used as a novel type of anticancer drug that acts by blocking cell cycle progression.  相似文献

Two novel prostanoid-related marine oxylipins, tricycloclavulone (1) and clavubicyclone (2), were isolated from the Okinawan soft coral Clavularia viridis. The structures of 1, having a tricyclo[5.3.0.0(1,4)]decane ring system, and 2, having a bicyclo[3.2.1]octane ring system, were elucidated on the basis of spectroscopic analysis. Clavubicyclone showed a moderate growth inhibition activity against tumor cells in vitro.  相似文献

Theophylline has an anhydrous form and a monohydrated form, and the dissolution rate of the anhydrous form is higher than that of the monohydrated form. Terahertz (THz) spectra of theophylline tablet containing the theophylline anhydrous form, monohydrated form, microcrystalline cellulose and magnesium stearate exhibited a specific absorption peak at 0.96 THz, where the theophylline anhydrous form demonstrated an absorption peak. Additionally, the intensity of the peak at 0.96 THz gradually decreased as the proportion of the anhydrous form decreased. The multivariate data analysis was performed to correlate the THz spectra of theophylline tablets with the ratio of the theophylline anhydrous form. The calibration model used to predict the mixing ratio of the theophylline anhydrous form from the THz spectra achieved root-mean-squared errors of cross-validation (RMSECV) of 2.89%, a slope of 0.9934 and an R(2) of 0.9927. In addition, there were intentions to develop a prediction model for the dissolution rate of theophylline from the drug product. The dissolution rate of theophylline tablet was gradually delayed as the proportion of the anhydrous form was decreased. The multivariate data analysis was performed to correlate the THz spectra of theophylline tablets with the dissolution rate. The calibration model used to predict the percentage of theophylline dissolved in 45 min from the THz spectra achieved an RMSECV of 3.29%, a slope of 0.9260 and an R(2) of 0.9423. Furthermore, there were no significant differences between the predicted and measured percentages of theophylline dissolved in 45 min in the theophylline tablets that were stored at 84% relative humidity (RH) and 25 °C for 12 h or 3 d.  相似文献

Three-dimensional magnetic resonance (MR) images of four kinds of fruit (Japanese apricot, peach, Japanese pear, and apple) were acquired throughout their growing periods (from April to August) to visualize and quantify their vascular structures. The vascular structures were emphasized using T 1-weighted spin-echo or gradient-echo pulse sequences, visualized using a maximum-intensity projection technique, and quantified using an image-segmentation program. As a result, we demonstrated that the vascular volume changed almost in proportion to the whole volume and that larger fruits generally had larger proportionality constants. We therefore concluded that MR microimaging is a powerful tool for studies of the vascular structure of fruit.  相似文献

Water‐soluble luminescent material was developed by introducing europium (Eu(III)) ions into the core of a star polymer. Living radical polymerization was used to obtain the star polymer. The strategy to introduce Eu(III) ions into the star polymer was studied using poly(methyl methacrylate) as an arm. The best Eu(III) ion introduction was obtained by simultaneous introduction, resulting in about 30 µmol/g‐polymer, which needed only one step for synthesis. The utilization of a hydrophilic polymer such as poly(ethylene oxide) (PEO) as an arm produced a water‐soluble star polymer. The Eu(III)‐bearing PEO star polymer obtained in this study was water soluble and showed fluorescence. In addition, it was stable in water after 1 month. The Eu(III)‐bearing star polymer exhibited luminescent properties under UV light irradiation with relatively high quantum yields of 60% in organic solution and 19% in aqueous solution. © 2013 Wiley Periodicals, Inc. The morphology and formation process of interpenetrated spherulites of poly(butylene succinate)/poly(vinylidene choloride‐co‐vinyl chloride) (PBSU/PVDCVC) blends were investigated by confocal laser scanning microscopy (CLSM). CLSM images showed that the dense fibrils of PBSU spherulites penetrated into the sparse PVDCVC spherulites. For a blend with PBSU content 50% and crystallization temperature Tc = 368 K, the simultaneous growth of PBSU and PVDCVC spherulites was observed. After PBSU fibrils collided with PVDCVC spherulites, they kept growing through PVDCVC spherulites. For a blend with PBSU content 30% and Tc = 363 K, PBSU started to nucleate after PVDCVC spherulites filled the whole space.  相似文献
