TNF-related apoptosis-inducing ligand (TRAIL) is an anticancer agent, which has greater apoptosis inducing capacity, but most of the cancer cells become resistant to TRAIL-induced apoptosis. The combined treatment of TRAIL with natural products could restore the cancer cell sensitivity to recombinant human TRAIL (rhTRAIL) protein and might enhance the TNF-related apoptosis-inducing ligand receptor (TRAIL-R) expression. This investigation was aimed to isolate flavonoids from leaves of Avicennia marina and evaluate their potential for sensitization of rhTRAIL in human cervical cancer cells (SiHa). The methanolic extract of A.marina leaves were purified and structure was elucidated as isoquercitrin by NMR and LC-MS analysis. Isolated isoquercitrin showed cytotoxicity against SiHa cell line at IC50 of 980 μM. Messenger RNA (mRNA) expression of TRAIL-Rs was quantified by qRT-PCR, combination of isoquercitrin, and/or rhTRAIL increased TRAIL-R1 and TRAIL-R2 gene expression by 7 folds and 4 folds, respectively. Also, FACS assay revealed that combined treatment has increased the early apoptosis up to 7.24%. In the present study, we found that isoquercitrin enhances the mRNA expression of TRAIL-Rs, but the percentage of apoptosis was meager, possibly due to the influence of other anti-apoptotic proteins.  相似文献

Summary Energy distributions are presented of the ions produced in a magnetron plasma source which hit the negatively biased substrate surface thus getting deeper insight into the coating process. By observing the mass analysed fluxes and the energy distributions of the impinging particles it is found that practically only atomic ions and atomic neutrals arrive at the substrate and contribute to the growth of the film.  相似文献

We demonstrate the accurate nanoscale mapping of near-surface loss and storage moduli on a polystyrene-polypropylene blend with contact resonance force microscopy (CR-FM). These viscoelastic properties are extracted from spatially resolved maps of the contact resonance frequency and quality factor of the AFM cantilever. We consider two methods of data acquisition: (i) discrete stepping between mapping points and (ii) continuous scanning. For point mapping and low-speed scanning, the values of the relative loss and storage modulus are in good agreement with the time-temperature superposition of low-frequency dynamic mechanical analysis measurements to the high frequencies probed by CR-FM.  相似文献

We present the first direct comparison of scanning ion conductance microscopy (SICM) with atomic force microscopy (AFM) for cell imaging. By imaging the same fibroblast or myoblast cell with both technologies in series, we highlight their advantages and disadvantages with respect to cell imaging. The finite imaging force applied to the sample in AFM imaging results in a coupling of mechanical sample properties into the measured sample topography. For soft samples such as cells this leads to artifacts in the measured topography and to elastic deformation, which we demonstrate by imaging whole fixed cells and cell extensions at high resolution. SICM imaging, on the other hand, has a noncontact character and can provide the true topography of soft samples at a comparable resolution.  相似文献

Results of scattering experiments with monochromatic electrons in the energy range from a few tenth of an eV to about 16 eV are reported. Below 1.8 eV collision energy no resonance structures have been found either in the total scattering (transmitted current) or in the elastic and inelastic differential cross sections. The resonances above 1.8 eV have been measured in the elastic and inelastic channels (up to vibrational quantum numberν=8 of the N2-molecule) in the angular range from 10° to 110°. In the inelastic channelsν≧2 especially the first resonance peak appears asymmetric, as predicted by calculations of the associated Franck-Condon-factors. In all inelastic channels the angular dependence (due to the pure resonance scattering) shows maxima at 0° and 90° and a minimum around 55°, probably indicating gerade-symmetry of the associated N 2 ? -state. Excitation functions of three states of N2 were measured between their thresholds (between 11 and 12 eV) and 15 eV, one of them showing several new resonance structures.  相似文献
