A comparative study of molecular structures of five L ‐proline (L ‐Pro) phosphonodipeptides: L ‐Pro‐NH‐C(Me,Me)‐PO3H2 (P1), L ‐Pro‐NH‐C(Me,iPr)‐PO3H2 (P2), L ‐Pro‐L ‐NH‐CH(iBu)‐PO3H2 (P3), L ‐Pro‐L ‐NH‐CH(PA)‐PO3H2 (P4) and L ‐Pro‐L ‐NH‐CH(BA)‐PO3H2 (P5) has been carried out using Raman and absorption infrared techniques of molecular spectroscopy. The interpretation of the obtained spectra has been supported by density functional theory calculations (DFT) at the B3LYP; 6–31 + + G** level using Gaussian 2003 software. The surface‐enhanced Raman scattering (SERS) on Ag‐sol in aqueous solutions of these phosphonopeptides has also been investigated. The surface geometry of these molecules on a silver colloidal surface has been determined by observing the position and relative intensity changes of the Pro ring, amide, phosphonate and so‐called spacer (−R) groups vibrations of the enhanced bands in their SERS spectra. Results show that P4 and P5 adsorb onto the silver as anionic molecules mainly via the amide bond (∼1630, ∼1533, ∼1248, ∼800 and ∼565 cm−1), Pro ring (∼956, ∼907 and ∼876 cm−1) and carboxylate group (∼1395 and ∼909 cm−1). Coadsorption of the imine nitrogen atom and PO group with the silver surface, possibly by formation of a weaker interaction with the metal, is also suggested by the enhancement of the bands at 1158 and 1248 cm−1. P1, P2 and P3 show two orientations of their main chain on the silver surface resulting from different interactions of the  C CH3,  NH and  CONH fragments with this surface. Bonding to the Ag surface occurs mainly through the imino atom (1166 cm−1) for P2, while for P1 and P3 it occurs via the methyl group(s) (1194–1208 cm−1). The amide group functionality (CONH) is practically not involved in the adsorption process for P1 and P2, whereas the Cs P bonds do assist in the adsorption. FT-IR and FT-RS spectra of three phosphonate tripeptides containing P-terminal L-Met-L-Ala [L-Gly-L-Met-L-Ala-PO3H2 (GMA), L-Leu-L-Met-L-Ala-PO3H2 ( LMA), and L-Phe-L-Met-L-Ala-PO3H2 (PMA)] were recorded and analyzed. Vibrational wavenumbers and intensities were calculated by density functional theory (DFT) at the B3LYP/6-311++G** level of theory and compared to these molecules in solid form. On the basis of this comparison, conclusions were drawn about the molecular structures. At the same time, the experimental data served as a test for the computational results. SERS spectra were recorded in a silver colloidal dispersion. Silver colloidal dispersions prepared by simple borohydride reduction of silver nitrate were used as substrates. A comparison is made between the SERS spectra and the spectra of the solid sample. Also, the capability of SERS for spectral fingerprinting of analytes with close structural properties using easily prepared substrates and relatively simple instrumentation is illustrated. By careful analysis, we obtained information on the orientation of these tripeptides and specific-competitive interactions of their functional groups with the silver surface. For example, all molecules are thought to adsorb on a silver surface via a P=O bond and a sulfur atom. In addition, the amide bond of GMA assists in the adsorption process, adopting a tilted orientation on the surface, with the N-H unit being closer to the surface than the C=O moiety. Conversely, the C=O unit of the LMA-CONH- bond lies closer to the silver surface than the N-H moiety. The -CH 3 group and P-O bond of LMA additionally interact with the silver surface, whereas for PMA the L-Phe lies almost flat on the colloidal silver surface.  相似文献

Here, we provide mechanistic insight to the photocleavage of a compound in the folate family, namely pteroic acid. A bis-decyl chain derivative of pteroic acid was synthesized, structurally characterized and photochemically investigated. We showed that, like folic acid, pteroic acid and the decylated derivative undergo a photocleavage reaction in the presence of H2O, while no reaction was observed in methanol solution. Furthermore, density functional theory calculations were carried out to predict relative stabilities of hypothetical mono-, bis- and tris-decylated pteroic acid derivatives to help rationalize the regioselectivity of the bis-decyl pteroic acid product. Additionally, the lipophilicity of the bis-decyl pteroic acid appears to confer a hydrophobic property enabling an interaction with biomembranes.  相似文献

Raman (RS) and surface-enhanced Raman scattering spectra (SERS) were measured for various length carboxyl terminal fragments (X-14 of amino acid sequence) of bombesin ( BN): BN13-14, BN12-14, BN11-14, BN10-14, BN9-14, and BN8-14 in silver colloidal solutions. Density functional theory (DFT) calculations of Raman wavenumbers and intensities with extended basis sets (B3LYP/6-31++G**) were performed with the aim of providing the definitive band allocations to the normal coordinates. The proposed band assignment is consistent with the assignment for similar compounds reported in the literature. The nonadsorbed and adsorbed molecular structures were deducted by detailed spectral analysis of the RS and SERS spectra, respectively. This analysis also allowed us to propose the particular surface geometry and orientation of these peptides on silver surface, and their specific interaction with the surface. For example, a SERS spectrum of BN8-14 indicates that the interaction of a thioether atom and Trp8 with the silver surface is favorable and may dictate the orientation and conformation of adsorbed peptide. One of the most prominent and common features in all of the fragments' SERS spectra is a approximately 692 cm (-1) band due to nu(C-S) accompanied by two or three bands of different C-S conformers for all, except BN8-14, which suggests that all of the above-mentioned compounds adsorb on the silver surface through the thioether atom and that the attachment of Trp8 produces limitation in a number of possible C-S conformers adopted on this surface. Our results also show clearly that His12 and CO do not interact with the colloid surface, which supports our earlier results.  相似文献

Neopentyl glycol (NPG) is a precursor for the manufacture of many valuable products of industrial importance such as polyester, polyurethane and alkyd resins, synthetic lubricants, hydraulic fluids, drugs, etc. The structure of NPG provides the resins with excellent hydrolytic stability, resistance to weather conditions, good flexibility-hardness balance, and outstanding functional properties. The paper presents a literature review on the development of methods for NPG preparation, focusing primarily on the synthesis of NPG by hydrogenation of hydroxypivaldehyde, which is obtained by the crossed aldol condensation of isobutyraldehyde and formaldehyde. Preparation of the substrates, catalysts, technical and apparatus solutions, and NPG purification were discussed.  相似文献

Salt concentrations in brine and temperature are the major environmental factors that affect activity of microorganisms and, thus may affect formation of biogenic amines (BAs) during the fermentation process. A model system to ferment cucumbers with low salt (0.5%, 1.5% or 5.0% NaCl) at two temperatures (11 or 23 °C) was used to study the ability of indigenous microbiota to produce biogenic amines and metabolize amino acid precursors. Colony counts for presumptive Enterococcus and Enterobacteriaceae increased by 4 and up to 2 log of CFU∙mL−1, respectively, and remained viable for more than 10 days. 16S rRNA sequencing showed that Lactobacillus and Enterobacter were dominant in fermented cucumbers with 0.5% and 1.5% salt concentrations after storage. The initial content of BAs in raw material of 25.44 ± 4.03 mg∙kg−1 fluctuated throughout experiment, but after 6 months there were no significant differences between tested variants. The most abundant BA was putrescine, that reached a maximum concentration of 158.02 ± 25.11 mg∙kg−1. The Biogenic Amines Index (BAI) calculated for all samples was significantly below that needed to induce undesirable effects upon consumption. The highest value was calculated for the 23 °C/5.0% NaCl brine variant after 192 h of fermentation (223.93 ± 54.40). Results presented in this work indicate that possibilities to control spontaneous fermentation by changing salt concentration and temperature to inhibit the formation of BAs are very limited.  相似文献

Radicals formed in γ-irradiated 1,3,5-trithiane (TT) and its three derivatives, α- and β-2,4,6-trimethyl-1,3,5-trithiane (α-TMT and β-TMT), and 2,4,6-trimethyl-2,4,6-triphenyl-1,3,5-trithiane (TMTPT), were studied by the electron paramagnetic resonance (EPR) method in the solid state. The sulfur radical cations (>S+•) were identified in all compounds at 77 K. In TT and its two derivatives, α-TMT and β-TMT, the >S+• decay via deprotonation-forming C-centered radicals. Further increase of temperature up to 293 K results in the appearance of thiyl-type radicals (RS). In TMTPT, the >S+• are stable up to 250 K. They formed the intermolecularly three-electron-bonded dimeric radical cations (S∴S)+ while RS radicals were not observed. Some of the radical assignments and their EPR parameters (g and a hyperfine splittings) obtained support from the DFT calculations.  相似文献

The aim of this study was to estimate the contamination of grain coffee, roasted coffee, instant coffee, and cocoa purchased in local markets with ochratoxin A (OTA) and its isomerization product 2′R-ochratoxin A (2′R-OTA), and to assess risk of dietary exposure to the mycotoxins. OTA and 2′R-OTA content was determined using the HPLC chromatography with immunoaffinity columns dedicated to OTA. OTA levels found in all the tested samples were below the maximum limits specified in the European Commission Regulation EC 1881/2006. Average OTA concentrations calculated for positive samples of grain coffee/roasted coffee/instant coffee/cocoa were 0.94/0.79/3.00/0.95 µg/kg, with the concentration ranges: 0.57–1.97/0.44–2.29/0.40–5.15/0.48–1.97 µg/kg, respectively. Average 2′R-OTA concentrations calculated for positive samples of roasted coffee/instant coffee were 0.90/1.48 µg/kg, with concentration ranges: 0.40–1.26/1.00–2.12 µg/kg, respectively. In turn, diastereomer was not found in any of the tested cocoa samples. Daily intake of both mycotoxins with coffee/cocoa would be below the TDI value even if the consumed coffee/cocoa were contaminated with OTA/2′R-OTA at the highest levels found in this study. Up to now only a few papers on both OTA and 2′R-OTA in roasted food products are available in the literature, and this is the first study in Poland.  相似文献

Petroleum, synthetic, and natural waxes have been used as hydrophobic bases for dispersions intended for use as barrier coatings for packaging paper. Oil-in-water dispersions with alkaline pH were prepared by a two-step homogenization procedure containing paraffin wax, with various characteristics, the Fischer–Tropsch synthesis product or beeswax. The size of the dispersed particles determined by dynamic light scattering depended on the type of hydrophobic base used and was in the range of 350–440 nm. The ability of dispersion particles in aggregation driven by electrostatic attraction, evaluated by Zeta potential analysis by electrophoretic light scattering, was from −26 to −50 mV. Static multiply light scattering was used for 30 days of stability assessment and helped to select the dispersion with a Sarawax SX70 wax base as the most stable. Dispersions were further used for coating the backing of kraft paper by the Meyer rod method. Coated paper with an applied coating of 6 g/m2 had very good hydrophobic properties (Cobb60 < 4 g/m2), sufficient strength properties, and air permeation, which enabled its application as a packaging material. The dispersions based on Sarawax SX70 wax were evaluated as the best coating for Mondi ProVantage Kraftliner 125 g/m2 backing paper. Good hydrophobic properties and strength properties indicate the possibility of using the SX70-based wax dispersion coating as a replacement for PFAS coatings in some applications.  相似文献
