Pharmaceutical excipients should not interact with active substances, however, in practice, they sometimes do it, affecting the efficacy, stability and safety of drugs. Thus, interactions between active substances and excipients are not desirable. For this reason, two component mixtures of oral antidiabetic drug linagliptin (LINA) with four excipients of different reactivity, i.e., lactose (LAC), mannitol (MAN), magnesium stearate (MGS) and polyvinylpyrrolidone (PVP), were prepared in a solid state. A high temperature and a high humidity of 60 °C and 70% RH, respectively, were applied as stressors in order to accelerate the potential interactions between LINA and excipients. Differential scanning calorimetry (DSC) as well as Fourier transform infrared (FT-IR) and near infrared (NIR) spectroscopy were used to estimate the changes due to potential interactions. In addition, chemometric computation of the data with principal component analysis (PCA) and hierarchical cluster analysis (HCA) was applied to adequately interpret the findings. Of the excipients used in the present experiment, all of them were not inert in relation to LINA. Some of the interactions were shown without any stressing, whereas others were observed under high-temperature/high-humidity conditions. Thus, it could be concluded that selection of appropriate excipients for LINA is very important question to minimize its degradation, especially when new types of formulations with LINA are being developed and manufactured.  相似文献

In non-magnetic heavy fermion systems, valence fluctuations prevent formation of long-range magnetic correlations. The RKKY exchange fields fluctuate due to f-electron transfers to the conduction band. To simulate these processes, we use the Ising-type model with fluctuating fields. The linear temperature dependence of the specific heat and temperature independence of susceptibility suggest that the f-electron contribution to these quantities can be considered as complementary to the one originating from Kondo resonance.  相似文献

X-ray photoelectron spectroscopy (XPS) is used to estimate phase and element contents of the composites based on cubic boron nitride. A composite is a mixture of several compounds and the use of traditional X-ray diffraction for phase analysis in many cases can be difficult. By using XPS, we were able to evaluate the atomic concentration of elements and the content of different compounds of each element. SEM and XPS showed that samples are heterogeneous. The TiB2 phase, expecting from thermodynamic calculations, was formed in both investigated samples of cBN/Ti3SiC2/TiC but in different amount. The Ti3SiC2 additive was found more chemically active then TiC.  相似文献

Through temperature-programmed pyrolysis, the physicochemical properties of Pteria martensii (PM) before and after calcination are investigated. Our results show that the mass loss is 11.02% from room temperature to 600 °C with an average mass loss rate of 0.19% per min. The decomposition of organics coupling with the phase transition of calcium carbonate from aragonite to calcite occurs in the range of 367.4–423.0 °C, as confirmed by X-ray diffraction and Fourier transform infrared spectroscopy analyses. The decomposition pores and channels are changed forming complex porous structures. The surface of sample shows a much rougher fracture, with higher C, O and N element concentrations. At the stage of decomposition and phase transition of organics, the average activation energy value is 118.78 kJ mol?1. This study provides valuable information on the calcination process and calcined PM for use in medicines.  相似文献

A top‐down approach is applied for the production of polyurethane (PU)–polyhedral oligomeric silsesquioxane (POSS) nanocomposites, namely melt blending. As opposed to the typical chemical incorporation during synthesis, a POSS moiety with two hydroxyl groups is melt blended into a commercial thermoplastic polyurethane with mass fraction up to 2 wt %. POSS disperses in the matrix in submicrometer‐sized crystals, as well as in length scale of few tens of nanometers, in the bulk. Phase separation of the produced composites was studied by both standard dynamic and isothermal annealing experiments. In an approach rare in the literature, the dynamics of phase separation is discussed based on isothermal differential scanning calorimetry curves recorded during annealing. The blended‐in nanoparticles affect the micromorphology in a complicated manner, dependent on the intrinsically complex phase separation mechanism of PU. At higher temperatures, POSS slows down the phase separation, whereas at lower ones, it enhances and accelerates it. POSS decreases the mechanical modulus of the final material, presumably as a result of changes in the microphase separation. © 2019 Wiley Periodicals, Inc. J. Polym. Sci., Part B: Polym.
