Doxorubicin (DOX) belongs to the group of anthracycline antibiotics with very effective anticancer properties. On the other hand, the cardiotoxic effects limit its application over the maximum cumulative dose. To overcome this obstacle, encapsulation of this drug into the protective nanotransporter such as apoferritin is beneficial. In this study, fluorescent behavior of DOX in various solvents was determined by fluorescence spectrometry, demonstrating the fluorescence quenching effect of water, which is often used as a solvent. It was found that by increasing the amount of the organic phase in the DOX solvent the dynamic quenching is significantly suppressed. Ethanol, acetonitrile and dimethyl sulfoxide were tested and the best linearity of the calibration curve was obtained when above 50 % of the solvent was present in the binary mixture with water. Moreover, pH influence on the DOX fluorescence was also observed within the range of 4–10. Two times higher fluorescence intensity was observed at pH 4 compared to pH 10. Further, the DOX behavior in capillary electrophoresis (CE) was investigated. Electrophoretic mobilities (CE) in various pH of the background electrolyte were determined within the range from 16.3 to ?13.3 × 10 ?9 m?2 V?1 s?1. Finally, CE was also used to monitor the encapsulation of DOX into the cavity of apoferritin as well as the pH-triggered release.  相似文献

Matrix metalloproteinases (MMPs) play numerous roles in physiological and pathological processes including cancer. Interactions of MMPs with other zinc-binding proteins are of great interest mainly from the point of view of the participation of these proteins in carcinogenesis. The aim of this study was to utilize chip gel electrophoresis for investigation of matrix metalloproteinase 9 (gelatinase B, MMP-9) interactions with collagen. The interaction was observed and the effect of temperature was tested. We further focused on the study of activation of MMP-9 by the presence of zinc-binding proteins called metallothioneins (MT). We confirmed the basic presumption of the activation of MMP-9 by the presence of MT similar to the temperature effect. Moreover, we chose the method of differential pulse voltammetry Brdicka reaction to verify our results. It can be concluded that the activity of MMP-9 was higher in the presence of MT in comparison with the experimental collagen(type I)-MMP-9 mixture in vitro.  相似文献

In this study, two methods for metallothioneins (MT) determination in a biological sample were compared. Particularly, twenty five human and nine pig blood serum samples and liver and kidney samples from thirty five carps (Cyprinus carpio) were analyzed by enzyme‐linked immunosorbent assay and differential pulse voltammetry Brdicka's reaction. The results obtained by these two methods were in good agreement. For commercially available MT standard the correlation coefficient between the single concentrations signal height was 0.99. In biological samples the correlation coefficients were 0.90 for fish liver and kidney samples, 0.91 for pig blood serum and 0.93 for human blood serum.  相似文献

Metallothionein (MT) is a low‐molecular mass protein playing an essential role in homeostasis of heavy metal ions. Its relation with formation and progression of a tumour disease is discussed in this article. Here, we propose a new methodological approach for visualization of MT on PVDF membranes after dot‐ and electroblotting by using a commercial mouse monoclonal antibody E9 and polyclonal chicken antibodies. The optimized procedure was as follows. We dotted 1 μL sample volume on PVDF membrane and let it to dry. Then, we blocked the membrane surface with 2% BSA in PBS for 30 min. After that, the membrane was incubated in chicken primary antibody (diluted 1:500), washed, and incubated in rabbit‐anti‐chicken secondary antibody conjugated with horseradish peroxidase. To visualize the interaction, we used 3‐aminoethyl‐9‐carbazole. Under these conditions, we estimated detection limit as 3 pg of MT per 1 μL. The optimal approach was further utilized for detection of MT level in two human fibroblast cell lines and in blood serum obtained from children with medulloblastoma. The results were in good agreement with differential pulse voltammetry‐Brdicka reaction.  相似文献

In the present paper we employed adsorptive transfer stripping technique coupled with chronopotentiometric stripping analysis for determination of metallothionein (MT) in tumor cell lines and differential pulse voltammetry Brdicka reaction for determination of MT in blood serum of patients with head and neck cancer or retinoblastoma, and of rats treated with cisplatin with respect to discuss the role of MT in formation of resistance on treatment with heavy metal based cytostatics. The cisplatin or carboplatin sensitive and resistant neuroblastoma cell lines were derived from the maternal cell line isolated from the bone metastasis of patients with neuroblastoma. Based on the results obtained it can be concluded that level of MT increases with higher dose of platinum based cytostatics at cells. Further we focused on determination of MT in blood serum of rats treated with cisplatin (two doses 1.05 mg and/or 2.1 mg of cisplatin per kg). The highest level of MT at rats treated with 1.05 mg cisplatin was determined after four hours as 4.9 μmol/L. In the case of the second experimental group the maximum was reached even after two hours of the treatment as 4.8 μmol/L. In addition we were interested in the effect of cisplatin or carboplatin treatment of patients with a tumor disease. At patients with tumor in head and neck area treated with cisplatin we observed that the level of MT was going higher due to administration of the drug. This phenomenon was observed at all patients. However at patients with retinoblastoma treated with carboplatin we observed various phenomena including decreasing, increasing or no changes in MT level. Progression of MT levels was therefore individual and probably depended on tumor resistance to carboplatin.  相似文献

From the point of view of the information theory, the term relevancy is actual only in the case of multielement determinations, because the information of one component determination is basically relevant. In the multicomponent determination the surplus of information, the s.c. redundancy, is given on the one hand by the relation between the information parameters of the analytical order and the equivalent parameters derived from the experimental data. On the other hand, the information from the individual terms of determined information parameters also have individual degrees of redundancy. But the given redundancy, the weight of information, is not necessarily always useful. The main problem is the correct and true determination of the given redundancy and following degree of relevancy.  相似文献

A general measure for the amount of informationI(p,p 0) is defined, which allows to determine the relationships specific for different cases of probability distribution before carrying out the analysisp 0 (x) and after it has been carried outp(x).
