In this study, different cultivation systems such as roller bottles (RB), 5-L stirred-tank bioreactor (STR), and disposable bioreactors were used to cultivate hybridoma for lab-scale production of Salmonella Enteritidis O-antigen-specific monoclonal antibody (MAb). Hybridoma cell line was cultivated in either serum-containing or serum-free medium (SFM) culture conditions. In STR, MAb production scaled up to 4 L, and production capabilities of the cells were also evaluated in different featured production systems. Moreover, the growth parameters of the cells in all production systems such as glucose consumption, lactate and ammonia production, and also MAb productivities were determined. Collected supernatants from the reactors were concentrated by a cross-flow filtration system. In conclusion, cells were not adapted to SFM in RB and STR. Therefore, less MAb titer in both STR and RB systems with SFM was observed compared to the cultures containing fetal bovine serum-supplemented medium. A higher MAb titer was gained in the membrane-aerated system compared to those in STR and RB. Although the highest MAb titer was obtained in the static membrane bioreactor system, the highest productivity was obtained in STR operated in semicontinuous mode with overlay aeration.  相似文献

In this study, the new compounds; 3-(4-{[3-(trifluoromethyl)benzyl]oxy}phenyl)propan-1-ol 3 was prepared by the reaction of 4-(3-hydroxypropyl)phenol 1 with 1-(bromomethyl)-3-(trifluoromethyl)benzene 2 and 4-[3-(4-{[3 (trifluoromethyl)benzyl]oxy}phenyl)propoxy] phthalonitrile 5 was synthesized by the reaction of 4-nitrophthalonitrile 4 with 3-(4-{[3-(trifluoromethyl)benzyl]oxy}phenyl)propan-1-ol 3. Novel peripherally tetra substituted H2Pc 6, Co(II) 7, Cu(II) 8, Ni(II) 9 and Fe(II) 10 phthalocyanines, which have peripheral positions with 4-[3-(4-{[3 (trifluoromethyl)benzyl]oxy}phenyl)propoxy] groups, were synthesized and all of the new compounds characterized by IR, 1H NMR, 13C NMR, UV–Vis, mass spectroscopies and elemental analysis. The electrochemical and spectroelectrochemical investigation of the phthalocyanines carrying 4-[3-(4-{[3 (trifluoromethyl)benzyl]oxy}phenyl)propoxy] groups were studied using various electrochemical techniques in DMF on a glassy carbon electrode. Cyclic voltammetry and square wave voltammetry studies show that the complexes have either metal based or ligand-based diffusion controlled electron transfer properties. To shed more light on the electron-transfer steps of the complexes and assignments of the redox couples were carried out by spectroelectrochemical measurements. The color changes during spectral changes of redox species were recorded with in situ electrocolorimetric measurements. The electrochemical and in situ UV–Vis spectral change of complexes indicated their applicability in the fields of the electrochemical technologies.  相似文献

Cellulose has been used as a raw material for the manufacture of membranes and fibers for many years. This review gives the background of the most recent methods of treating or dissolving cellulose, and its derivatives to form polymer films or membranes for a variety of applications. Indeed, some potential applications of bacterial cellulose, nanofibrillar cellulose (NFC) for films showing enhanced barrier characteristics are reviewed as well as the utilization of cellulose nanonocrystals (CNC) for production of highly oriented super strong films or thin films is discussed. Because of the success of the Lyocell process as well as the amine/metal thiocyanate solvent blends of cellulose and other polysaccharides like starch, chitosan, and other natural polymers. Consequently, the use of cellulose (or its derivatives) and another polysaccharide dissolved as a blend is also elaborated. It is our hope that the reader will want to follow up and investigate these new systems and use them to develop end use materials for all sorts of applications, from medical to water filtration, or electrogels for use in batteries.  相似文献

A new copper(II) complex, [Cu(Bphen)(Leu)(NO3)(H2O)] (Bphen = 4,7-diphenyl-1,10-phenanthroline, leu = L-leucine), has been synthesized and characterized by IR spectroscopy, CHN analysis, and single-crystal X-ray diffraction techniques. The CT-DNA binding properties of the complex have been investigated by both absorption and emission spectroscopy. The binding parameters for the fluorescence Scatchard plot were also determined. Further, the interaction of the complex with bovine serum albumin (BSA) has been investigated using absorption and emission spectroscopy. The thermodynamic parameters, free energy change (ΔG), enthalpy change (ΔH), and entropy change (ΔS), were calculated by the van’t Hoff equation and discussed. The distance between BSA and the complex has been obtained according to fluorescence resonance energy transfer. Conformational changes of BSA have been observed from synchronous fluorescence. Antioxidant and radical scavenging activities of the complex were determined by various in vitro assays such as 1,1-diphenyl-2-picryl-hydrazyl free radicals (DPPH˙), 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radicals (ABTS˙+), and reducing ability determination by H2O2 scavenging methods.  相似文献

A series of copolymers of N,N‐dialkyl‐N‐2‐(methoxycarbonyl)allyl allyl ammonium chloride, N,N‐dialkyl‐N‐2‐(ethoxycarbonyl)allyl allyl ammonium chloride, and N,N‐dialkyl‐N‐2‐(t‐butoxycarbonyl)allyl allyl ammonium bromide with diallyldimethylammonium chloride (DADMAC) were prepared in water at 60 °C with 2,2′‐azo‐bis(2‐amidinopropane)dihydrochloride. A strong effect of ester substituents on cyclopolymerization was observed. The methyl and ethyl ester monomers showed high cyclization efficiencies during homopolymerizations and copolymerizations. Unexpectedly, the t‐butyl ester derivatives showed high crosslinking tendencies. Water‐soluble copolymers were obtained only with a decrease in the molar fraction of t‐butyl ester monomer below 30%. Relative reactivities of the allyl‐acrylate monomers in photopolymerizations were compared with the relative reactivity of DADMAC. Allyl‐acrylate monomers were much more reactive than DADMAC; the photopolymerization rate decreased in the following order: N,N‐morpholine‐N‐2‐(t‐butoxycarbonyl)allyl allyl ammonium bromide > N,N‐piperidyl‐N‐2‐(t‐butoxycarbonyl)allyl allyl ammonium bromide > N,N‐dibutyl‐N‐2‐(ethoxycarbonyl)allyl allyl ammonium chloride > N,N‐piperidyl‐N‐2‐(ethoxycarbonyl)allyl allyl ammonium chloride ∼ N,N‐morpholine‐N‐2‐(ethoxycarbonyl)allyl allyl ammonium chloride ∼ N,N‐piperidyl‐N‐2‐(methoxycarbonyl)allyl allyl ammonium chloride > N‐methyl‐N‐butyl‐N‐2‐(ethoxycarbonyl)allyl allyl ammonium chloride. Intrinsic viscosities of the polymers measured in 0.09 M NaCl ranged from 1.06 to 3.20 dL/g. The highest viscosities were obtained for copolymers of the t‐butyl ester monomers with piperidine and morpholine substituents. The copolymer of the t‐butyl ester with piperidine substituent and DADMAC was hydrolyzed in acid to give a polymer with zwitterionic character. © 2001 John Wiley & Sons, Inc. Novel allyl‐acrylate quaternary ammonium salts were synthesized using two different methods. In the first (method 1), N,N‐dimethyl‐N‐2‐(ethoxycarbonyl)allyl allylammonium bromide and N,N‐dimethyl‐N‐2‐(tert‐butoxycarbonyl)allyl allylammonium bromide were formed by reacting tertiary amines with allyl bromide. The second (method 2) involved reacting N,N‐dialkyl‐N‐allylamine with either ethyl α‐chloromethyl acrylate (ECMA) or tert‐butyl α‐bromomethyl acrylate (TBBMA). The monomers obtained with the method 2 were N,N‐diethyl‐N‐2‐(ethoxycarbonyl)allyl allylammonium chloride, N,N‐diethyl‐N‐2‐(tert‐butoxycarbonyl)allyl allylammonium bromide, and N,N‐piperidyl‐N‐2‐(ethoxycarbonyl)allyl allylammonium chloride. Higher purity monomers were obtained with the method 2. Solution polymerizations with 2,2′‐azobis(2‐amidinopropane) dihydrochloride (V‐50) in water at 60–70°C gave soluble cyclopolymers which showed polyelectrolyte behavior in pure water. Intrinsic viscosities measured in 0.09M NaCl ranged from 0.45 to 2.45 dL/g. 1H‐ and 13C‐NMR spectra indicated high cyclization efficiencies. The ester groups of the tert‐butyl polymer were hydrolyzed completely in acid to give a polymer with zwitterionic character. © 1999 John Wiley & Sons, Inc. The competitive rate data and Taft relationships for the coupling of bromomagnesium n‐butyl (substituted phenyl) cuprates with alkyl bromides show that selective n‐butyl transfer can be explained by an oxidative addition mechanism. Taft reaction constants also show that the residual group FG‐C6H4 in the mixed cuprate n‐Bu(FG‐C6H4)CuMgBr changes the ability of the copper nucleophile to react with the electrophile RBr. These results provide support for the commonly accepted hypothesis regarding the dependence of the R1 group transfer ability on the strength of R2? Cu bond in reactions of R1R2CuMgBr reagents. A new LC method has been developed and validated for the direct determination of bupropion and its main metabolite, hydroxybupropion in human plasma. Plasma samples were analyzed after a simple, one step protein precipitation with trichloroacetic acid using a C8 column and mobile phase, consisting of methanol/acetonitrile/phosphate buffer (10 mM, pH 3.0) (40:10:50, v/v/v) and 20 mM 1-heptane sulfonic acid sodium salt with carbamazepine as the internal standard. UV detection was performed at 214 and 254 nm. The method was validated over the concentration range of 60–2,400 and 150–4,700 ng mL?1 for bupropion and hydroxybupropion, respectively. The intra- and inter-day assay variability was less than 15% for the two analytes. Limit of detection values were 24.8 and 63.4 ng mL?1 for bupropion and hydroxybupropion, respectively. The method developed was applied to quantification of bupropion and hydroxybupropion in human plasma.  相似文献

The feasibility of static secondary ion mass spectrometry (S-SIMS) for the detection of molecule specific information from complex materials, such as natural clay and soil samples, has been investigated. Ion trap (IT), as well as triple quadrupole (TQ) instruments, have been used for mass analysis. Secondary ion images have been acquired using time-of-flight (TOF) S-SIMS. The generation of molecular adduct ions from thin and thick layers on the mineral substrates has been investigated using KBr as a simple model system. Results show that molecular adducts of KBr can be indeed detected from the spiked materials. However, the concentrations of the spiking solutions have to be significantly larger than expected from the surface area measured by gas adsorption techniques. In addition imaging analysis has evidenced that the detection of adduct ions in the mass spectra directly relates to the presence of local micro-crystallites.  相似文献

Capturing pathogens on a sensor surface is one of the most important steps in the design of a biosensor. The efficiency of a biosensor at capturing pathogens has direct bearing on its sensitivity. In this work we investigated the capturing of Escherichia coli on substrates modified with antibodies targeting different types of fimbriae: K88ab (F4), K88ac (F4), K99 (F5), 987P (F6), F41, and CFA/I. The results suggest that all these fimbriae can be used for the efficient immobilization of living E. coli cells. The immobilization efficiency was affected by the purity and clone type of the antibody and the fimbriae expression level of the bacteria. For a specific fimbriae type, a higher immobilization efficiency was often observed with the monoclonal antibodies. Immunoimmobilization was utilized in an antibody microarray immersed in a mixed culture of pathogens to demonstrate the rapid and simultaneous label-free detection of multiple pathogens within less than 1 h using a single test. The capture rate of living pathogens exceeds a single bacterium per 100 × 100 μm(2) area per 0.5 h of incubation for a bulk concentration of 10(5) cfu/mL.  相似文献
