Shotgun proteomics technology has matured in the research laboratories and is poised to enter clinical laboratories. However, the road to this transition is sprinkled with major technical unknowns such as long‐term stability of the platform, reproducibility of the technology and clinical utility over traditional antibody‐based platforms. Further, regulatory bodies that oversee the clinical laboratory operations are unfamiliar with this new technology. As a result, diagnostic laboratories have avoided using shotgun proteomics for routine diagnostics. In this perspectives article, we describe the clinical implementation of a shotgun proteomics assay for amyloid subtyping, with a special emphasis on standardizing the platform for better quality control and earning clinical acceptance. This assay is the first shotgun proteomics assay to receive regulatory approval for patient diagnosis. The blueprint of this assay can be utilized to develop novel proteomics assays for detecting numerous other disease pathologies. Copyright © 2013 John Wiley & Sons, Ltd. 相似文献

We consider the effect of a variable representing the competition between the frequency of the field and the frequency of the spin flipping (Ω) on the dynamics of the metamagnetic Ising model in a cubic lattice under the presence of a time varying (oscillating) external magnetic field. The system is modelled with a formalism of master equation at a mean-field level. The time averaged staggered magnetization (Ms) acts as the order parameter and divides temperature field plane into three regions: anti-ferromagnetic, paramagnetic and coexistence of anti-ferromagnetic and paramagnetic phases. It is observed that the topology of the dynamical phase diagram depends strongly on Ω as well as the ratio between interlayer and intralayer couplings. 相似文献

Conditions influencing the extent of P-C(aryl) vs P-C(alkyl) bond cleavage in the reaction of Ph(2)P(CH(2))(2)PPh(2) with lithium in THF have been investigated. The results complement and elucidate earlier work; they indicate that the mechanism of P-C bond cleavage in tertiary phosphines of this type involves a thermodynamic equilibrium between P-C(aryl) and P-C(alkyl) cleaved radicals and anions, followed by reaction and stabilization of these as lithium salts. The addition of water to the reaction mixture causes a reestablishment of the cleavage equilibrium prior to the formation of the secondary phosphines. A mechanism involving competitive release of leaving groups as the thermodynamically most stable anion or radical has been proposed. The preparation of (R, R)-(+/-)/(R, S)-PhP(H)(CH(2))(2)P(H)Ph by this route has been optimized. 相似文献

The aldol reactions of 2‐arylimino‐3‐aryl‐thiazolidine‐4‐ones with benzaldehyde carried out at ?78 °C were found to produce sec‐carbinols. Intramolecular hydrogen bonding within the aldol products forming a six‐membered ring enabled the assignment of stereochemistries of the major and minor diastereomers via analysis of the syn and anti 3JH,H 1H NMR coupling constants. Transformation of cyclohexa-1,3- and 1,4-dienes to carbasugars is described. Photooxygenation of dienes gave bicyclic endoperoxides, which were reduced with thiourea to the corresponding 1,4-diols with cis-configuration. Lactonization of the remaining double bond by oxidative addition of acetic acid to the double bond in the presence of Mn(OAc)3 followed by lactone ring-opening reaction gave the target branched carbasugars. 相似文献

An electrochemical dsDNA nanobiosensor was fabricated using amino‐functionalized multi walled carbon nanotubes modified glassy carbon electrode (NH2fMWCNTs/GCE) for the sensitive detection of DNA bases and electrochemical monitoring of drug‐DNA interaction. The influence of functional groups on MWCNT was studied by MWCNT functionalized with NH2 (NH2fMWCNTs) and COOH (COOHfMWCNT) groups based on the signal of DNA bases. The modified electrodes were characterized by scanning electron microscopy. One layer of calf thymus double stranded deoxyribonucleic acid (ct‐dsDNA) was immobilized onto the NH2fMWCNTs/GCE (dsDNA/NH2fMWCNTs/GCE). The dsDNA/NH2fMWCNTs/GCE were used to investigate the interaction between the dsDNA and the anticancer drug gemcitabine by differential pulse voltammetry in acetate buffer of pH 4.70. For the confirmation of interaction, the lowering in intensity of the current signals of guanine and adenine was considered as an indicator. Electrochemical impedance spectroscopy studies were performed for the comparison of the modified surfaces. In order to define and visualize the interaction mechanism between gemcitabine and dsDNA/NH2fMWCNTs/GCE at the molecular level, in silico methods including docking and molecular dynamics simulations were employed. 相似文献

Ascorbic acid is the most common electroactive biological compound found in some plant species (e.g., Citrus species, Rosa species). The electrochemical oxidation of ascorbic acid was investigated by cyclic, linear sweep, differential pulse (DPV),
and square wave (SWV) voltammetry. For analytical purposes, a very well-resolved diffusion-controlled voltammetric peak was
obtained in acetate buffer at pH 3.50 for DPV and SWV. The linear response was obtained in the range of 3.52–176.1 μg/mL with
a detection limit of 0.88 μg/mL for DPV and 0.52 μg/mL for SWV. Based on this study, simple, rapid, selective, and sensitive
voltammetric methods were developed for the determination of ascorbic acid in pharmaceutical dosage forms and Rosa species (R. dumalis ssp. boissieri var. boissieri, R. canina, R. pulverulenta, R. heckeliana ssp. vanheurckiana, and R. montana subsp. woronowii). The results obtained are compared with the HPLC data. The developed methods enable the extracts to be analyzed without
the necessity of any time-consuming separation.
