A central question in biological water splitting concerns the oxidation states of the manganese ions that comprise the oxygen-evolving complex of photosystem II. Understanding the nature and order of oxidation events that occur during the catalytic cycle of five Si states (i = 0–4) is of fundamental importance both for the natural system and for artificial water oxidation catalysts. Despite the widespread adoption of the so-called “high-valent scheme”—where, for example, the Mn oxidation states in the S2 state are assigned as III, IV, IV, IV—the competing “low-valent scheme” that differs by a total of two metal unpaired electrons (i.e. III, III, III, IV in the S2 state) is favored by several recent studies for the biological catalyst. The question of the correct oxidation state assignment is addressed here by a detailed computational comparison of the two schemes using a common structural platform and theoretical approach. Models based on crystallographic constraints were constructed for all conceivable oxidation state assignments in the four (semi)stable S states of the oxygen evolving complex, sampling various protonation levels and patterns to ensure comprehensive coverage. The models are evaluated with respect to their geometric, energetic, electronic, and spectroscopic properties against available experimental EXAFS, XFEL-XRD, EPR, ENDOR and Mn K pre-edge XANES data. New 2.5 K 55Mn ENDOR data of the S2 state are also reported. Our results conclusively show that the entire S state phenomenology can only be accommodated within the high-valent scheme by adopting a single motif and protonation pattern that progresses smoothly from S0 (III, III, III, IV) to S3 (IV, IV, IV, IV), satisfying all experimental constraints and reproducing all observables. By contrast, it was impossible to construct a consistent cycle based on the low-valent scheme for all S states. Instead, the low-valent models developed here may provide new insight into the over-reduced S states and the states involved in the assembly of the catalytically active water oxidizing cluster. 相似文献

In this study, biobased furan dicarboxylate polyesters have been prepared using 2,5‐furandicarboxylic acid (FDCA) and diols with high number of methylene groups (long‐chain diols), namely, 8, 9, 10, and 12. Because of the high boiling points of these diols, a modified procedure of the well‐known melt polycondensation was applied in this work. According to this, the dimethyl ester of FDCA (DMFD) reacted in the first transesterification stage with the corresponding diols forming bis‐hydroxy‐alkylene furan dicarboxylates (BHFD). In the second stage, the BHFD reacted with DMFD again at temperatures of 150–170 °C (for 4–5 h), and in the final stage, the temperature was raised to 210–230 °C (vacuum was applied for 2–3 h). The molecular weight of the polyesters and the content of oligomers, as was verified by gel permeation chromatography analysis, depend on the polycondensation time and temperature. The chemical structure of the polyesters was verified from 1H NMR spectroscopy. All the polymers were found to be semicrystalline, with melting temperatures from 69 to 140 °C depending on the diol used. In addition, the mechanical properties also varied with the type of diol. The higher values were observed for poly(octylene 2,5‐furanoate), whereas the lowest values were observed for poly(dodecylene 2,5‐furanoate) with the higher number of methylene groups in its repeating unit. © 2015 Wiley Periodicals, Inc. The ability of a series of crown ether-functionalized dendrimers to function as alkali metal picrate extraction agents is assessed by liquid-liquid extraction and 1H NMR titration experiments. Crown ether-functionalized dendrimers that contain Fréchet-type poly(benzyl ether) dendrons of different generation as building blocks display different extraction characteristics toward alkali metal cations. Macroencapsulation of islets of Langerhans is a promising strategy for transplantation of insulin‐producing cells in the absence of immunosuppression to treat type 1 diabetes. Hollow fiber membranes are of interest there because they offer a large surface‐to‐volume ratio and can potentially be retrieved or refilled. However, current available fibers have limitations in exchange of nutrients, oxygen, and delivery of insulin potentially impacting graft survival. Here, multibore hollow fibers for islets encapsulation are designed and tested. They consist of seven bores and are prepared using nondegradable polymers with high mechanical stability and low cell adhesion properties. Human islets encapsulated there have a glucose induced insulin response (GIIS) similar to nonencapsulated islets. During 7 d of cell culture in vitro, the GIIS increases with graded doses of islets demonstrating the suitability of the microenvironment for islet survival. Moreover, first implantation studies in mice demonstrate device material biocompatibility with minimal tissue responses. Besides, formation of new blood vessels close to the implanted device is observed, an important requirement for maintaining islet viability and fast exchange of glucose and insulin. The results indicate that the developed fibers have high islet bearing capacity and can potentially be applied for a clinically applicable bioartificial pancreas. 相似文献

The present technique describes the development of a simple sensitive spot optical test and the construction of a portable biosensor for the rapid one-shot detection of naphthalene acetic acid (NAA) using stabilized lipid films supported on a methacrylate polymer on a glass fiber filter with incorporated auxin-binding protein 1 receptor. The lipid films without the receptor provided fluorescence under a UV lamp. The use of the receptor in these films quenched this fluorescence and the colour became similar to that of the filters without the lipid films. A drop of aqueous solution of naphthalene acetic acid provided a “switching on” of the fluorescence which allows the rapid detection of this stimulant at the levels of 10−9 M concentrations. It was also possible to have quantitative data based on a calibration graph. The effect of potent interferences included a wide range of compounds. The results showed no interferences from these compounds in concentration levels usually found in real samples. The method was applied for the determination of NAA in fruits and vegetables and the reproducibility of the method was checked in about 50 samples. A quantitative method for the detection of NAA in crops that can be complimentary to HPLC methods is provided in the present paper. These lipid films can be used as portable biosensors for the rapid one-shot detection of NAA in fruits and vegetables by non-skilled personnel in the field. 相似文献

A novel control volume finite element method with adaptive anisotropic unstructured meshes is presented for three-dimensional three-phase flows with interfacial tension. The numerical framework consists of a mixed control volume and finite element formulation with a new P1DG-P2 elements (linear discontinuous velocity between elements and quadratic continuous pressure between elements). A “volume of fluid” type method is used for the interface capturing, which is based on compressive control volume advection and second-order finite element methods. A force-balanced continuum surface force model is employed for the interfacial tension on unstructured meshes. The interfacial tension coefficient decomposition method is also used to deal with interfacial tension pairings between different phases. Numerical examples of benchmark tests and the dynamics of three-dimensional three-phase rising bubble, and droplet impact are presented. The results are compared with the analytical solutions and previously published experimental data, demonstrating the capability of the present method. 相似文献

The current work examines the role of working-memory capacity in problem solving in science education. It treats an information-processing model with tools of complexity theory. Nonlinear methods are used to correlate the subjects' achievement scores with working-memory capacity. Data have been taken from the achievement scores in simple organic-synthesis chemical problems. The subjects (N = 319) were in grade twelve (age 17–18). Problems of various Z-demands (that is the number of steps needed to solve the problem) from two to eight were used. Rank-order sequences of the subjects, according to their scores, were generated, and each score was then replaced by the value of subject's working memory capacity measured by the digit backward span test. Then the sequences were mapped onto a one-dimensional random walk model and when treated as dynamic flows were found to possess fractal geometry with characteristics depending on the Z-demand of the problem. The findings were interpreted using concepts from complexity theory, such as correlation exponents, fractal dimensions and entropy. The null hypothesis was tested with surrogate data. 相似文献
