In this work, differential mobility cytometry (DMC) was used to monitor cell separation based on aptamer recognition for target cells. In this device, open-tubular capillaries coated with Sgc8 aptamers were used as affinity chromatography columns for separation. After cells were injected into the columns, oscillating flow was generated to allow for long-term cell adhesion studies. This process was monitored by optical microscopy, and differential imaging was used to analyze the cells as they adhered to the affinity surface. We investigated the capture time, capture efficiency, purity of target and control cells, as well as the reusability of the affinity columns. Capture time for both CCRF-CEM cells and Jurkat T cells was 0.4 ± 0.2 s, which demonstrated the high separation affinity between aptamers and target cells. The capture efficiency for CCRF-CEM cells was 95% and purity was 99% in a cell mixture. With the advantage of both high cell capture efficiency and purity, DMC combined with aptamer-based separation emerges as a powerful tool for rare cell enrichment. In addition, aptamer-based DMC channels were found to be more robust than antibody based channels with respect to reuse of the separation device.  相似文献

Methodologies for the rapid screening of coating systems were developed and applied to photopolymer lacquers for photoembossing applications. Continuous and discrete gradient libraries were prepared with a gradient in grating period along the short axis and along the long axis, a gradient in exposure energy, development temperature, film thickness, photoinitiator concentration, or monomer to polymer mass ratio. Discrete gradient libraries consisted of arrays of rectangular films made by pipetting a certain amount of sample onto a chemically patterned substrate consisting of hydrophilic patches surrounded by hydrophobic, fluorinated barriers. The shape and height of the photoembossed gratings were measured using an automated AFM. Optimum grating height was obtained for a 20-microm period at intermediate exposure energies, photoinitiator concentrations, or both. Height improves with development temperature (max 110 degrees C), monomer-to-polymer ratio (max 55 wt % monomer), and film thickness. Surface topography can also be optimized, depending on any specific application.  相似文献

A new range of selenium‐based reversible addition‐fragmentation chain‐transfer (RAFT) agents is described and tested in the polymerization of styrene, acrylates, vinyl esters, and N‐vinylcaprolactam. The synthesized N,N‐dimethyldiselenocarbamates were poor control agents for styrene polymerization, whereas polyacrylates of controlled molar masses and bearing a diselenocarbamate terminal group could be synthesized. The polymerization of vinyl acetate and vinyl pivalate proceeded in a controlled manner as confirmed by size‐exclusion chromatography, matrix‐assisted laser desorption ionization‐time‐of‐flight mass spectrometry, and 77Se NMR analyses. The capability of these RAFT agents to control the polymerization of both more‐activated monomers and less‐activated monomers was exemplified through the synthesis of a poly(t‐butyl acrylate)‐b‐poly(vinyl acetate) diblock copolymer. Considering the very broad range of carbamate groups which can be envisioned, this finding opens numerous perspectives for diselenocarbamate‐mediated RAFT polymerization with its specificities yet to be explored. © 2013 Wiley Periodicals, Inc. The main goal of the present study was to develop a procedure for the simultaneous GC/MS analysis of a wide range of water-soluble organic compounds (WSOCs, mainly dicarboxylic acids and sugars) in atmospheric aerosol as chemical markers of atmospheric processes. The response surface methodology, including central composite design, was applied to systematically investigate and optimize the operating conditions of a solvent extraction method as the most widely used procedure for determining WSOCs in particulate matter (PM) samples. A solvent mixture of methanol/dichloromethane (90:10) and a volume of 10 ml were the optimum conditions that permit the simultaneous analysis of 30 target WSOCs, including dicarboxylic acids and sugars. Recoveries and limits of detection were determined for a standard mixture containing target analytes, and they varied from 79 to 103 % and 0.3 to 1 ng?m?3, respectively. The optimized procedure was used for the analysis of field PM filters and demonstrated its feasibility in detecting several important environmental WSOCs.  相似文献

To the best of our knowledge, this is the very first time that a thorough study of the synthetic procedures, molecular and thermal characterization, followed by structure/properties relationship for symmetric and non-symmetric second generation (2-G) dendritic terpolymers is reported. Actually, the synthesis of the non-symmetric materials is reported for the first time in the literature. Anionic polymerization enables the synthesis of well-defined polymers that, despite the architecture complexity, absolute control over the average molecular weight, as well as block composition, is achieved. The dendritic type macromolecular architecture affects the microphase separation, because different morphologies are obtained, which do not exhibit long range order, and various defects or dislocations are evident attributed to the increased number of junction points of the final material despite the satisfactory thermal annealing at temperatures above the highest glass transition temperature of all blocks. For comparison reasons, the initial dendrons (miktoarm star terpolymer precursors) which are connected to each other in order to synthesize the final dendritic terpolymers are characterized in solution and in bulk and their self-assembly is also studied. A major conclusion is that specific structures are adopted which depend on the type of the core connection between the ligand and the active sites of the dendrons.  相似文献
